Laboratory diagnosis of Erwinia amylovora (Burrill) Winslow et al.

For detection of fireblight, caused by bacterium Erwinia amylovora (Burrill) Winslow et al., a testing procedure recommended by EPPO (European Plant Protection Organization) is used. It includes: isolation of bacteria by plating on semiselective and non-selective media, serological methods (agglutination and immunofluorescence), biochemical tests and pathogenicity test on immature pear fruits. As an addition to all these laboratory tests we have introduced a molecular biology based method (PCR), which both confirms the Erwinia amylovora (Ea) in a bacterial culture, and enables to distinguish Ea from other bacteria such as Pseudomonas syringae pv. syringae, a causal agent of a disease similar to fireblight regarding the symptoms, and from other bacteria frequently present on fruit trees. A testing on samples with and without symptoms of first finding of fireblight in Slovenia is presented.
