Expression of the genes of non-specific immune response in the body of Gallus gallus domesticus under the influence of immunodepressive vaccine viruses | Изучение экспресии ключевых генов неспецифического иммунного ответа в организме Gallus gallus domesticus под влиянием инфицирования иммунодепрессивными вакцинными вирусами

Inglés. The immune response to vaccination is formed through the activation of gene expression processes in cells and tissues of various immunocompetent organs. Morphology of fabricii bursa, thymus and expression of immunity genes IRF7, PTGS2, IL8L2 were studied in commercial laying hens on the background of vaccination with live vaccines against infectious bursal disease and infectious anemia of chickens. In each experiment, an experimental (vaccinated) and a control (intact) group of chickens of 30 individuals each were formed. An immunocomplex vaccine from the strain "VNIVIP" was used against bursal disease, against infectious anemia of chickens - Nobilis CAVP4 vaccine produced by MSDAnimalHealth. In chickens vaccinated against bursal disease, changes were noted on the part of the fabricii bursa: its shape is oval, with folds protruding through the peritoneal cover, the cover itself is flabby, and the mucous membrane is hyperemic, spot hemorrhages are on the surface. The IL8L2 gene increased its expression 21.6 times, the PTGS2 gene by 26.6 times, and the IRF7 gene practically did not increase its expression. The thymus was examined in chickens vaccinated against infectious anemia. Spot hemorrhages were noted on its surface, the structure of the cortical layer was changed (replacement of thymus cells with adipocytes, fibroblasts, and other lymphoid cells). The IL8L2 gene increased expression 8.75 times, the gene PTGS2 - 22.94 times, and the IRF7 gene did not exceed expression compared to the control level. According to the results of the research, it has been concluded that the IL8L2 and PTGS2 genes responsible for the synthesis of inflammatory components are actively expressed when exposed to the cells of viruses of chicken infectious anemia and infectious bursal disease. The level of expression of the IRF7 gene in the immune organs of the experimental groups of chickens practically did not differ from those in the target organs of control birds due to the immunosuppressive effect of viruses.

Ruso. Иммунный ответ на вакцинацию формируется посредством активации процессов экспрессии генов в клетках и тканях различных иммунокомпетентных органов. Исследовали морфологию фабрициевой сумки, тимуса и экспрессию генов иммунитета IRF7, PTGS2, IL8L2 у промышленных кур-несушек на фоне вакцинации живыми вакцинами против инфекционной бурсальной болезни и инфекционной анемии цыплят. В каждом эксперименте формировали опытную (вакцинируемую) и контрольную (интактную) группу цыплят по 30 особей в каждой. Против бурсальной болезни применяли иммунокомплексную вакцину из штамма "ВНИВИП", против инфекционной анемии цыплят - вакцину Нобилис CAVP4 производства MSDAnimalHealth. У цыплят, вакцинированных от бурсальной болезни, отмечены изменения со стороны фабрициевой сумки: её форма овальная, с проступающими через перитонеальный покров складками, сам покров дряблый, слизистая оболочка гиперемирована, на поверхности точечные кровоизлияния. Ген IL8L2 увеличил свою экспрессию в 21,6 раз, ген PTGS2 - в 26,6 раз, ген IRF7 экспрессию практически не увеличил. У цыплят, вакцинированных от инфекционной анемии, исследовали тимус. На его поверхности отмечены точечные кровоизлияния, изменена структура коркового слоя (замена клеток тимуса на адипоциты, фибробласты и др. лимфоидные клетки). Ген IL8L2 увеличил экспрессию в 8,75 раз, ген PTGS2 - в 22,94 раза, а ген IRF7 не превысил экспрессию по сравнению с уровнем контроля. По результатам исследований были сделаны выводы, что гены IL8L2 и PTGS2, отвечающие за синтез воспалительных компонентов, активно экспрессируются при воздействии на клетки вирусов инфекционной анемии цыплят и инфекционной бурсальной болезни. Уровень экспрессии гена IRF7 в иммунных органах опытных групп цыплят практически не отличался от такового в органах-мишенях контрольных птиц по причине иммунодепрессивного действия вирусов.
