Effect of carbon source on in vitro plant regeneration in anther culture of cyclamen (Cyclamen persicum Mill.)

To improve the plant-regeneration efficiency in anther culture of cyclamen (Cyclamen persicum Mill.), the effect of carbon source on plant regeneration in the anther culture was investigated. Anthers of F1 ('Kage Yellow'×'Anneke') were cultured on media containing different carbon source (sucrose, glucose, fructose, or maltose) to produce microspore-derived plants. The largest number of embryoids was obtained from the anthers cultured on media with maltose. The anther culture of five cyclamen genotypes, in addition to F1 ('Kage Yellow'×'Anneke'), was also examined on media containing sucrose or maltose. While no plant was obtained on media with sucrose in all of the five genotypes, embryoid formation on media with maltose was observed in four genotypes. The optimum concentration of maltose for the anther culture of F1 ('Kage Yellow'×'Anneke') was 394.4 mM. The present study demonstrates that maltose is one of the most effective carbon sources for in vitro plant regeneration in anther culture of cyclamen.
