Simultaneous LC Analysis of Five Bioactive Alkaloids in an Anti-Angiogenesis Herbal Formula, Qing-Luo-Yin

A rapid and reliable gradient reversed-phase LC method has been developed for simultaneous analysis of five bioactive alkaloids (matrine, oxymatrine, oxysophocarpine, sinomenine, and berberine) in the decoction of Qing-Luo-Yin (QLY), an anti-angiogenesis herbal formula. The compounds were separated on a C₁₈ column and UV detection was at 214 nm. The separation conditions including pH, mobile-phase composition, and gradient were optimized. The effect of pH on the retention behavior of the five alkaloids on the C₁₈ column was investigated. The method was validated for specificity, linearity, precision, recovery, and reproducibility and successfully used for analysis of five alkaloids in the QLY decoction and in decoctions prepared from its individual herbs. Inter-herb effects were deduced by comparing the compounds present in the decoction of QLY with those in the decoction of the individual herbs.
