Toxicity of endosulfan and citrinin alone and in combination in pregnant rats: Clinico-haematological and serum biochemical alterations

In the present investigation, the effects of endosulfan and citrinin on the clinico- haematological and serum biochemical parameters in pregnant rats on 20th day of gestation were studied. Timed pregnant female Wistar rats were randomly allotted to four groups (individual treatments, combination and control) of 10 rats each. Endosulfan was fed @ 1mg/kg bw in olive oil by oral gavage (Gr I), CIT @ 10 ppm infeed (Gr II) and both (endosulfan+citrinin) in combination (Gr III) during the gestation days 6–20. The fourth group was kept as a control and was fed toxin free feed. The treated rats were observed daily for clinical manifestations, feed and water intake and weekly body weight gains. Blood was collectedfrom heart for studying haematobiochemical parameters. Both Gr I and Gr II treated rats exhibited reduced feed intake along with intermittent anorexia, polydipsia, lacrimation and lowered body weight gain as compared to those of controls. These signs were more pronounced in the combination group. The treated rats became anaemic (normocytic normochromic anaemia) and also revealed lymphocytopenic leucocytopenia. Biochemical alterations included decrease in serum total protein, albumin and globulin values and an increase in the levels of AST, ALT, creatinine and LDH. Hormone estimation revealed increase in the serum oestradiol levels in Gr I as compared to those of Gr II and the controls. However, progesterone levels were comparable to that of controls in all the treatment groups. SDS-PAGE of urine revealed albuminuria. The present investigation led to the conclusion that low doses of endosulfan @ 1 mg/kg bw and citrinin @ 10 ppm infeed individually caused significant toxicity in pregnant rats and the effects were additive when both the toxins were given simultaneously.
