Effect of different phytohormones on the in vitro propagation and rooting of Citrus macrophylla

Factors affecting in vitro propagation from nodal explants of mature trees of Alemow (Citrus macrophylla Wester) were studied. When BA was the only phytohormone used, proliferation was not observed. Several combinations of BA (1, 2 and 3 mg/L) and kinetin (0, 1 and 2 mg/L) or GA (1 and 2 mg/L) were carried out for to optimize the proliferation phase. BA/kinetin combinations did not produce better results than BA alone; the number of shoot and the explant length were independent of the kinetin concentration, although the interaction of BA x kinetin was significant and the tallest shoots were observed in 2 mg/L BA and 1 or 2 mg/L kinetin. Productivity (number of shoots x the average shoot length) was higher with 1 or 2 mg/L BA and 1 mg/L kinetin. When BA/GA combinations were used, the number of shoots was independent of the BA concentration and best results were obtained with 2 mg/L of GA; the number of shoot per explant was superior to 2 in this concentration. Productivity was higher when 1 mg/L BA and 2 mg/L GA were added to the culture medium. Transferring in vitro shoots to rooting media, containing different combinations of IBA (1 and 3 mg/L) and IAA (0 and 1 mg/L), produced complete plantlets. The highest rooting percentage and number of roots were obtained on a medium containing IBA in combination with 1 mg/L IAA and the roots were longer when 3 mg/L IBA, with or without IAA, were used. The success during the acclimatisation was close to 100% and the plantlets exhibited normal growth in soil under greenhouse conditions.
