Assessment of genetic variation in an artichoke European collection by means of molecular markers

A core collection, constituted of accessions from former germplasm collections present in Italy, France and Spain, has been built up to be representative of the four main artichoke typologies, i.e. 'Petit Violet de Provence', 'Catanese', 'Spinoso Sardo', 'Romanesco'. The core collection was analyzed at the molecular level, with the aim of: assessing the diversity present; measuring the genetic distance among accessions; and clustering the accessions according to genetic distances. Genomic DNA was extracted from 556 genotypes belonging to the artichoke germplasm collection and analysed using different types of molecular markers (i.e. ISSR, SSR, AFLP). Here we present the results obtained using four ISSR primers and four AFLP primer combinations. The different markers were able to detect different levels of polymorphism, with AFLP markers detecting higher levels of variation. The data obtained from all markers were scored and used to construct genetic distance matrices. Accessions were then clustered based on their genetic distance. Finally, discriminant analysis was carried out to detect the correct assignment of each genotype with respect to the country of origin and artichoke typologies.
