Primer System for Reindeer (Rangifer tarandus) Mitochondrial Genome Sequencing

In our study, we developed a primer system and optimized the PCR protocol for amplification of the reindeer (Rangifer tarandus) mitochondrial genome. We sequenced and assembled two mitochondrial genomes of wild and domestic reindeer from the Nenets Autonomous Okrug. These two genomes were compared with known sequenced mitochondria of domestic reindeers from Aoluguya (China) and Horonobe (Japan). Among 160 polymorphic sites found, some of them have private alleles for domestic and wild reindeer from the Nenets Autonomous Okrug. The developed system helps to provide new information for phylogeographic and genetic variation studies of reindeer and to add data from mitochondrial markers unexamined for reindeer such as ND1–ND6 and ATP6, ATP8.
