Embryogenic cell lines and germination of shoot from somatic embryos of Punica granatum 'Ganesh'

Somatic embryo formation occurred from nodal segments of pomegranate (Punica granatum 'Ganesh'). An embryogenic callus was induced from segments of node from mature plants cultured on ¾ MS liquid medium with filter paper boat supplemented with individual Indole-3-acetic acid (IAA) and thidiazuron (TDZ) at three different concentrations (0.5, 1 and 2 mg L-1). Nodulated embryogenic yellowish green mass of callus was induced on IAA containing media and greenish nodulated callus with somatic embryos was noticed on media supplemented with TDZ. Globular to bipolar stage embryos were more prominent after 90 days of culture on filter paper boat. Such calli has embryogenic activity for over three years through secondary embryogenesis. Germination of somatic embryos occurred on ¾ MS semi-solid media supplemented with IAA individually or in combination with TDZ and coconut water. The present study is more efficient for the maintenance of embryogenic cultures for longer time without any subculture.
