A rapid (1-hour), reproducible, quantitative method based on high-performance liquid chromatography (HPLC) analysis is described for the isolation and determination of the various carotenoids in cow's milk, and is applied to the determination of the effect of seasonal variation in carotenoid composition and content in milk from pasture-fed dairy cows. The method involves the saponification and extraction of milk, followed by HPLC analysis. An alternative colorimetric method is discussed for the determination of total carotenoids for use when an HPLC system is unavailable.(wz)
