1. A single-step isolation procedure is described for the hyperglycaemic hormones I and II (HGH I and II) from the corpora cardiaca (CC) of the American cockroach, Periplaneta americana, using reverse-phase high-performance liquid chromatography. 2. The efficacy of extraction and purification was found to be suitable to allow quantification of the two peptides in individual cockroach CC. 3. The CC always contained more HGH I than HGH II (overall molar ratio HGH I: HGH II was 4.3). 4. During late larval development the CC contained the same amount of hormones as adults. 5. in adults the difference between sexes was statistically significant. 6. Bisection of the CC revealed that the hyperglycaemic peptides are only present in the anterior part of the CC which contains the glandular and storage region.
