Modulation of bioactivities of tocopherols by various environmental factors in cell culture systems

Tocopherols exhibit various physiological activities. To better understand the bioactivities and mechanisms of tocopherols in cell culture systems, the modulation of the cell growth inhibitory activity of tocopherols under different culture conditions was investigated. δ-Tocopherol (δT) showed the most potent inhibition against growth of PC-3 and HT- 29 human cancer cells. The inhibitory activity was significantly enhanced in the range of 1-2.5% fetal bovine serum (FBS) or 1-2 mg/mL bovine serum albumin; the inhibitory effect of δT was significantly less pronounced in media containing 95oC heated-FBS. The δT activity decreased with increasing volume of treatment media and was also affected by the initial seeding cell number. Different vehicles including dimethyl sulfoxide, deoxycholic acid, and polyvinyl alcohol (PVA), significantly enhanced the growth inhibitory activities of δT. PVA dramatically enhanced δT activity without any cytotoxicity. These results indicate that various factors affect the bioactivities of tocopherols in cell cultures.
