Sarıçam (Pinus sylvestris L.) Tohum Bahçesinde Mg ve Mn Element İçerikleri Bakımından Genetik Çeşitliliğin Belirlenmesi | Determination of Genetic Diversity in Scots Pine (Pinus sylvestris L.) Seed Orchard in Terms of Mg and Mn Element Contents

Orman ağaçlarında verimliliği artırmanın etkin yollarından biri de ıslah çalışmalarıdır. Islah çalışmaları ile ormanların büyüme hızını artırmak, biyotik ve abiyotik zararlılara karşı dirençli bireyler yetiştirmek mümkün olmaktadır. Bunun için ormanların genetik çeşitliliğinin belirlenmesi büyük önem arz etmektedir. Bu uygulamaların yapıldığı genetik ıslah çalışmaları içerisinde tohum bahçeleri ayrı bir öneme sahiptir. Bu nedenle yapılan araştırma Taşköprü Tekçam sarıçam klonal tohum bahçesinde gerçekleştirilmiştir. Sarıçam türü hem ekonomik değeri olan hem de ıslahının yapılması öngörülen bir tür olmasından dolayı tercih edilmiştir. Yapılan bu çalışmada tohum bahçesinde 30 klonun 8 rametinden 3 tekerrürlü olarak toplam 240 adet ağaç rastgele örneklenmiştir. Ağaçların son yıl ibrelerinden örnekler alınarak laboratuvarda yıkama işlemine, ardından iki farklı kurutma işlemine tabi tutulmuştur. Daha sonra laboratuvara gönderilerek öncelikle eritiş yöntemi ile numuneler hazırlanmış, sonrasında ICP-OES cihazı ile Mg (Magnezyum) ve Mn (Mangan) element konsantrasyonları belirlenmiştir. One of the most basic ways to increase productivity in forestry is tree breeding. With breeding studies, it is possible to increase the growth rate of forests and to grow individuals resistant to biotic and abiotic pests. For this purpose, determining forests' genetic diversity is of great importance. In addition, genetic diversity is very important for species to adapt to global warming and rapidly changing environmental conditions. Seed orchards are one of the important breeding facilities established within the scope of tree breeding studies. Therefore, the most important issue in seed orchards is the protection of genetic diversity. Because genetic diversity is the insurance of the species against possible future risks, many studies have been conducted to determine genetic diversity in seed orchards. However, most of the studies were conducted using morphological characters. In this study, genetic diversity in the seed orchard was evaluated by looking at needle element concentrations. In this study, a total of 240 trees from 8 rameters of 30 clones were randomly sampled in Taşköprü Tekçam Sarıçam seed orchard. Magnesium (Mg), a macronutrient, and manganese (Mn), a micronutrient, which play a role in carbon fixation in plants and are used in chlorophyll production, were studied. Samples were taken from last year’s needles of the trees and subjected to washing in the laboratory, followed by two different drying processes. Then, the samples were sent to the laboratory and firstly prepared by melting method and then element concentrations were determined by ICP-OES device.
