Estudio de los mecanismos asociados a la neuropatogénesis del alfaherpesvirus bovino 5 (BoHV5) en su hospedador natural | Study of the mechanisms associated with bovine alphaherpesvirus 5 (BoHV5) neuropathogenesis in its natural hospedator

El alfaherpesvirus bovino 5 (BoHV5) es el agente etiológico de la meningoencefalitis no supurativa en terneros, causal de brotes fatales frecuentes en Argentina y Brasil y cuya patogenia aún no ha sido completamente esclarecida. Este virus está estrechamente relacionado con el alfaherpesvirus bovino 1 (BoHV1), el cual causa enfermedades respiratorias, abortos, trastornos genitales y, ocasionalmente, encefalitis en el ganado. Aunque ambos virus son neurotrópicos y comparten propiedades biológicas similares, se desconoce el motivo por el cual estos BoHV difieren en su capacidad para causar enfermedades neurológicas. Con el objeto de estudiar diversos aspectos de la neuropatogénesis del BoHV5 y comparar los hallazgos con BoHV1, se obtuvieron muestras de tejido neural de terneros inoculados intranasalmente con BoHV1 y BoHV5 durante la infección aguda, la latencia y la reactivación. Se determinó la distribución del genoma de ambos virus en sistema nervioso observándose que en la infección aguda, la distribución de BoHV1 es más amplia que la de BoHV5, mientras que durante la latencia y reactivación sólo el genoma de BoHV5 está presente en este tejido. También se analizó la expresión de genes virales representativos de la cinética inmediata temprana (IE), temprana (E) y tardía (L) en los distintos estadios de infección. No se detectó expresión génica viral en tejido nervioso durante la infección aguda de terneros infectados con BoHV1 o BoHV5. En terneros latentemente infectados con BoHV5 se detectó la expresión del gen TK en todos los sectores del sistema nervioso central evaluados y se detectaron genes L en la corteza cerebral y en ganglio trigémino (GT) durante la latencia de BoHV5 y BoHV1, respectivamente, lo cual permite asumir que en este estadio existe cierto nivel de expresión de genes IE que iniciarían la cascada de expresión génica. Durante la etapa de reactivación no se detectó la expresión de genes IE o E en tejido nervioso de bovinos infectados. En esta etapa se detectó la expresión de genes L, gD de BoHV1 y gB de BoHV5. Esto sugiere que la expresión de genes L es un evento clave para que se logre la infección productiva en esta etapa. Por otro lado se evaluaron las variaciones en los niveles de expresión de los receptores tipo toll (TLR) en diferentes regiones del SNC bovino. En general, los resultados de este estudio demostraron que la infección del sistema nervioso por los BoHV estimuló la expresión de los TLR3, TLR7 y TLR8 y no modificó significativamente la expresión de TLR9, y fue posible determinar que la magnitud en la variación de la expresión de los TLR se correlaciona con las manifestaciones clínicas que se pueden observar en cada etapa del ciclo infeccioso de los BoHV. Por otro lado, se analizaron los niveles de expresión de ARNm de ciclinas mediante qRT-PCR, observandose que existen diferencias notables en la modulación de la expresión de ciclinas por BoHV1 y BoHV5, particularmente en GT, el sitio principal de latencia de los alfaherpesvirus. En este trabajo se evaluó la presencia de linfocitos CD8+ citotóxicos mediante inmunohistoquímica y se determinó la expresión de perforina y granzimas en el tejido nervioso. No se detectó una presencia abundante de linfocitos T CD8+. Estos fueron más evidentes durante la infección aguda por BoHV1 y menos evidentes en el GT de los bovinos infectados con BoHV5. Del mismo modo, la ausencia de expresión de perforina y granzimas durante la infección aguda facilitaría la diseminación viral en el tejido nervioso. Durante la latencia de BoHV5, fue posible determinar la expresión de granzima B, lo cual sugiere que antígenos virales mantendrían activos a los linfocitos citotóxicos durante esta etapa. Por último, se analizó la capacidad de BoHV5 para inducir apoptosis en cultivos celulares y en tejido nervioso. Pudiendose observar que BoHV1 tendría más capacidad para inducir apoptosis in vitro e in vivo. Los cambios apoptóticos se corresponden con la capacidad de replicación de las cepas de BoHV1 y BoHV5. En este estudio se demostró que la reactivación molecular espontánea puede ser un evento frecuente en las infecciones por ambos alfaherpesvirus. Se detectó una regulación positiva significativa de todos los TLR después de la infección primaria de los tejidos neurales por ambos BoHV, a excepción de TLR9 que no demostró variaciones significativas. Las diferencias notables en los niveles de ARNm de TLR durante la infección aguda y la reactivación proporcionan evidencia de que la respuesta inmune innata puede estar involucrada en los resultados clínicos observados en cada etapa. Se observaron también, diferencias marcadas entre la inducción in vitro de apoptosis por las cepas BoHV1 Cooper y BoHV5 97/613. Se demostró una asociación entre la magnitud de la replicación de los BoHV y la inducción de apoptosis en el GT. Por otro lado se demostró por primera vez la modulación de BoHV5 sobre la expresión de ciclinas en los tejidos neurales de su hospedador natural. Estos hallazgos sugieren que la inducción de apoptosis y la respuesta inmune innata orquestan el resultado final de la infección por alfaherpesvirus del sistema nervioso bovino.

Bovine alphaherpesvirus 5 (BoHV5) is the etiological agent of non-suppurative meningoencephalitis in calves, the cause of frequent fatal outbreaks in Argentina and Brazil. BoHV5 pathogenesis of which has not been fully elucidated yet. This virus is closely related to bovine alphaherpesvirus 1 (BoHV1), which causes respiratory diseases, abortions, genital disorders, and occasionally encephalitis in cattle. Although both viruses are neurotropic and share similar biological properties, the reason why these alphaherpesviruses differ in their ability to cause neurological disease is unknown. In order to study various aspects of BoHV5 neuropathogenesis and to compare the findings with BoHV1 infection, neural tissue samples were obtained from calves intranasally inoculated with BoHV1 and BoHV5 during acute infection, latency, and reactivation. The distribution of the genome of both viruses in the nervous system was determined, observing that during acute infection, the distribution of BoHV1 is broader than that of BoHV5, while during latency and reactivation only the BoHV5 genome is present in this tissue. The expression of viral genes representative of the immediate-early (IE), early (E) and late (L) kinetics in the different stages of infection was also analyzed. No viral gene expression was detected in nervous tissue during acute primary infection of calves infected with BoHV1 or BoHV5. In calves latently infected with BoHV5, the expression of the TK gene was detected in all the sectors of the central nervous system evaluated and L genes were detected in the cerebral cortex and in the trigeminal ganglion (TG) during the latency of BoHV5 and BoHV1, respectively, which allows us to assume that at this stage there is a certain level of expression of IE genes that would initiate the gene expression cascade. During the reactivation stage, the expression of IE or E genes was not detected in nervous tissue of infected cattle. At this stage, the expression of L genes, BoHV1 gD and BoHV5 gB was detected. This suggests that L gene expression is a key event for productive infection to be achieved at this stage. On the other hand, variations in the levels of expression of toll-like receptors (TLR) in different regions of the bovine CNS were evaluated. In general, the results of this study demonstrated that infection of the nervous system by BoHV stimulated the expression of TLR3, TLR7, and TLR8 and did not significantly modify the expression of TLR9. It was possible to determine that the magnitude in the variation of TLR expression is correlated with the clinical manifestations that can be observed at each stage of the infectious cycle of BoHV. The expression levels of cyclins mRNA were analyzed using qRT-PCR, observing that there are notable differences in the modulation of cyclin expression by BoHV1 and BoHV5, particularly in the TG, the main latency site of alphaherpesviruses. In this work, the presence of cytotoxic CD8+ lymphocytes was evaluated by immunohistochemistry and the expression of perforin and granzymes in nervous tissue was determined. An abundant presence of CD8+ T lymphocytes was not detected. These cells were more evident during acute BoHV1 infection and less evident in the TG of BoHV5- infected cattle. Similarly, the absence of perforin and granzyme expression during acute infection would facilitate viral spread in nervous tissue. During BoHV5 latency, it was possible to determine the expression of granzyme B, which suggests that viral antigens would keep cytotoxic lymphocytes active during this stage. Finally, the ability of BoHV5 to induce apoptosis in cell cultures and in nervous tissue was analyzed. It was evident that BoHV1 would have more capacity to induce apoptosis in vitro and in vivo. The apoptotic changes are in agreement with the level of replication of BoHV1 and BoHV5 strains.This study demonstrated that spontaneous molecular reactivation can be a frequent event in infections by both BoHV. A significant positive regulation of all TLR was also detected after primary infection of neural tissues by both viruses, except for TLR 9 who did not show significant variations. The notable differences in the TLR mRNA levels during acute infection and reactivation provide evidence that the innate immune response may be involved in the clinical outcomes observed at each stage. Marked differences were also observed between in vitro induction of apoptosis by the BoHV1 Cooper and BoHV5 97/613 strains. An association between the magnitude of BoHV replication and the induction of apoptosis in the TG was demonstrated. On the other hand, the modulation of BoHV5 on the expression of cyclins in the neural tissues of its natural host was demonstrated for the first time. These findings suggest that the induction of apoptosis and the innate immune response orchestrate the final outcome of alphaherpesviruses infection of the bovine nervous system.
