Model Hydrotime dan Hubungannya dengan Uji Vigor pada Beberapa Konsentrasi PEG 6000 dan Varietas Kedelai (Glycine max L.) | Hydrotime Model and Its Relationship with Vigor Tests at Different PEG 6000 Concentrations and Soybean Varieties (Glycine max L.)

Kacang kedelai (Glycine Max L.) merupakan tanaman palawija dari Famili Fabaceae yang penting di Indonesia. Pertumbuhan tanaman kedelai agar tumbuh dan berproduksi optimal diperlukan uji vigor. Pendekatan uji vigor dapat dikembangkan dengan model hydrotime. Penelitian ini bertujuan mendapatkan pola kadar air benih pada beberapa konsentrasi PEG 6000 dan hubungannya dengan uji vigor benih pada beberapa varietas kedelai. Terdapat dua percobaan, yakni pembuatan pola kadar air benih dan hubungan pola kadar air benih dengan mutu fisiologis. Percobaan pembuatan pola kadar air benih menggunakan Rancangan Kelompok Lengkap Teracak (RKLT) dengan faktor tunggal, yaitu konsentrasi PEG 6000. Perlakuan terdiri atas empat taraf konsentrasi PEG 6000, yaitu 0%, 5%, 7,5%, dan 10%. Perlakuan diacak lengkap dalam 3 kelompok waktu yang berfungsi sebagai ulangan, sehingga terdapat 12 satuan percobaan. Konsentrasi tersebut secara berurutan setara dengan potensial air 0 Mpa, -0,03 Mpa, -0,11 Mpa, dan -0,19 Mpa. Percobaan kedua adalah hubungan pola kadar air benih dengan mutu fisiologis yang menggunakan Rancangan Kelompok Lengkap Teracak (RKLT) faktorial dua faktor. Faktor pertama terdiri atas tiga taraf, yaitu Anjasmoro, Dega 1, dan Dering 1. Faktor kedua adalah konsentrasi PEG yang terdiri atas empat taraf, yaitu 0% (P0), 5% (P1), 7,5% (P2), dan 10% (P3). Perlakuan diulang sebanyak 3 ulangan, sehingga terdapat 12 perlakuan dengan 36 satuan percobaan. Pola kadar air menunjukan peningkatan kadar air pada perlakuan kontrol dan PEG 5% yang terdiri atas tiga fase imbibisi, yaitu fase I 0–8 jam, fase II 8–52 jam, fase ketiga =52 jam. Perlakuan pemberian PEG 7,5% dan 10% mebetuk pola pada fase I, yaitu 0-20 jam, fase II >20 jam, dan fase III belum terindentifikasi. Pemberian konsentrasi PEG 6000 diatas 5% memperpanjang fase II dan menunda kemunculan Radicle Emergence. Kemunculan radikula pada perlakuan kontrol dan konsentrasi PEG 5% berada pada jam ke-36 dan pada perlakuan konsentrasi PEG 7,5% dan 10% berada pada jam ke-48. Konsentrasi PEG 6000 perlakuan kontrol dan 5% membentuk model hydrotime yang potensial untuk pengujian vigor benih.

Soybean (Glycine Max L.) is an important crop with the Fabaceae family in Indonesia. The growth of soybean plants to grow and produce optimally requires vigour testing. The vigour test approach can be developed with the hydrotime model. This study aims to obtain the pattern of seed moisture content at several concentrations of PEG 6000 and its relationship with seed vigour test on several varieties of soybean. There were two experiments, namely the making of seed moisture content patterns and the relationship between seed moisture content patterns and physiological quality. The seed moisture content patterning experiment used a Randomized Complete Block Design (RCBD) with a single factor, namely PEG 6000 concentration. The treatment consisted of four levels of PEG 6000 concentration, namely 0%, 5%, 7.5%, and 10%. The treatments were completely randomised in 3 time groups that served as replicates, so there were 12 experimental units. The concentrations were equivalent to a water potential of 0 Mpa, -0.03 Mpa, -0.11 Mpa, and -0.19 Mpa, respectively. The second experiment was the relationship between seed moisture content pattern and physiological quality using a two-factor factorial Randomized Complete Block Design (RCBD). The first factor consisted of three levels, namely Anjasmoro, Dega 1, and Dering 1. The second factor was PEG concentration which consisted of four levels, namely 0% (P0), 5% (P1), 7.5% (P2), and 10% (P3). The treatments were repeated 3 times, so there were 12 treatments with 36 experimental units. The pattern of water content showed an increase in water content in the control treatment and 5% PEG which consisted of three phases of imbibition, namely phase I 0–8 hours, phase II 8–52 hours, the third phase =52 hours. The treatment of 7.5% and 10% PEG formed a pattern in phase I, namely 0–20 hours, phase II >20 hours, and phase III has not been identified. Giving PEG 6000 concentrations above 5% prolonged phase II and delayed radicle emergence. Radicle emergence in the control treatment and 5% PEG concentration was at the 36th hour and in the treatment of 7.5% and 10% PEG concentration was at the 48th hour. The control treatment and 5% PEG 6000 concentration formed a potential hydrotime model for seed vigour testing.
