Herança do teor de isoflavonas em sementes de soja (Glycine max (L.) Merrill) | Inheritance of the isoflavone content in soybean seeds (Glycine max (L.) Merrill)

Os efeitos benéficos à saúde associados às isoflavonas da soja incluem a atenuação dos sintomas da menopausa, redução da osteoporose, melhora dos níveis de colesterol do sangue, diminuição do risco de certos tipos de câncer e de doenças coronarianas. Os teores e a composição de isoflavonas variam de acordo com a parte morfológica da semente de soja (cotilédone, hipocótilo e tegumento); e, também, em função do genótipo e das condições ambientais. Os objetivos deste trabalho foram: avaliar os efeitos do genótipo do embrião, do genótipo nuclear e citoplasmático da planta-mãe, bem como os mecanismos de ação gênica e grau de dominância para a característica teor de isoflavonas em sementes de soja; estimar herdabilidades e correlações entre as formas de isoflavonas encontradas e entre cada forma e a característica teor de proteína. Para isso, foram estudadas populações derivadas dos cruzamentos recíprocos entre IAC-100 (alto teor de isoflavonas) e BARC-8 (baixo teor de isoflavonas), obtidas no verão de 2000/2001. Os teores das isoflavonas foram determinados por cromatografia líquida de alta eficiência e o teor protéico pelo método de Kjeldahl. Foram analisados três contrastes para verificar efeitos de dominância, citoplasmático e materno. O efeito citoplasmático foi testado juntamente com os efeitos epistáticos entre genes citoplasmáticos e nucleares pela análise de variância (ANOVA) do modelo genético Y ijk = m + n j + c i + (cn) ij + ε ijk. Em que: Y ijk = valor observado no citoplasma i no núcleo j na repetição k; m= média geral; c i = efeito do i-ésimo citoplasma; n j = efeito do j-ésimo núcleo e; (cn) ij = efeitos epistáticos entre os genes citoplasmáticos e nucleares; ε ijk = erro experimental. Os componentes de médias foram estimados utilizando dois modelos genéticos, um com e outro sem interação epistática citoplasma x núcleo, utilizando o método dos quadrados mínimos ponderados. A importância relativa de cada componente genético foi avaliada pelo método de eliminação de Gauss. Seis formas de isoflavonas foram detectadas nas sementes de soja: daidzina, genistina, glicitina, malonildaidzina, malonilgenistina e malonilglicitina. Glicitina e malonilglicitina não foram significativamente contrastantes entre os progenitores. Os resultados indicam a existência de efeito materno e citoplasmático atuando sobre os teores das isoflavonas estudadas. Tais informações são relevantes nos processos de seleção para estas características, pois na presença de pelo menos um desses efeitos, a seleção praticada em sementes F 2 é inferior àquela baseada na média das plantas F 2 , ou na média de progênies. Na análise dos componentes de médias os resultados foram similares para o teor de daidzina e malonildaidzina, pois todos os parâmetros foram significativos pelo teste t e o efeito citoplasmático foi o de maior importância relativa. Porém, para o teor de genistina, os efeitos aditivos, de desvio de dominância e de interação citoplasma x dominância não foram significativos e o efeito citoplasmático foi o efeito mais importante. Já para o teor de malonilgenistina os parâmetros genéticos significativos foram: efeitos aditivos e epistáticos citoplasma x aditivo e citoplasma x dominante, sendo o efeito aditivo o de maior importância relativa. Tais informações podem ser úteis na escolha do melhor progenitor materno, o qual possua o melhor citoplasma, para aumentar o ganho na seleção. Devido à influência dos efeitos materno e citoplasmático sobre os teores das isoflavonas estudadas foi necessário estimar as herdabilidades para essas características com base na progênies F 3 . As sementes F 3 foram obtidas, juntamente com sementes dos progenitores, na safra 2001/2002. A herdabilidade no sentido amplo para os teores de daidzina, genistina, malonildaidzina, malonilgenistina e teor de isoflavonas totais foram superiores a 90%, sugerindo sucesso na seleção feita em gerações precoces. A herdabilidade no sentido amplo para o teor de proteína foi 33%. Os coeficientes de correlações fenotípicas entre as isoflavonas foram positivos e de grande magnitude (>0,80), indicando que essas características podem ser melhoradas simultaneamente. Os coeficientes de correlações fenotípicas entre as isoflavonas estudadas e o teor de proteína foram todos negativos, e entre isoflavonas totais e teor de proteína foi 0,47, confirmando a tendência de correlação negativa entre essas características.

The beneficial effects associated with isoflavone include the attenuation of the symptoms of menopause, reduction of osteoporosis, reduction on blood cholesterol levels, decrease on the risk of certain types of cancer and of coronary diseases. The objectives of this study were: to evaluate the effects of the genotype of the embryo, of the nuclear and cytoplasmatic genotypes, as well as of the mechanisms of genic action and of dominance on the isoflavone contents of soybean seeds, to estimate heritabilities for the isoflavone contents in soybean seeds and to estimate correlations between total isoflavone and between each isoflavone type, and protein content. Populations derived from the reciprocals cross between BARC-8 (low isoflavone content) and IAC-100 (high isoflavone content) were produced during the summer of 2000/2001. The isoflavones content were determined by high- performance liquid chromatography and the protein content were determined by the Kjekdahl method. Three contrasts were analyzed for to verify the effects of dominance, cytoplasmatic and maternal. The identified cytoplasmatic effect were tested with the epistatics effects among cytoplasmatic genes and nuclear genes by the analysis of variance (ANOVA) of the genetic model Y ijk = m + n j + c i + (cn) ij + e ijk . In which: Y ijk = value observed in the i cytoplasm in the j nucleus in the k repetition; m = general average; c i = effect of the i-th cytoplasm; n j = effect of the j-th nucleus and; (cn) ij = epistatic effects between cytoplasmatic and nuclear genes, e ijk = experimental error. The study of the components of means considered two genetic models, one with and the other without interaction cytoplasm x nucleus. The genetic components of the means were estimated by the weighted least square method squares and the relative importance of each genetic effect was evaluated by the Gauss elimination method. Six isoflavone kinds were detected: daidzin, genistin, glycitin, malonyldaidzin, malonylgenistin and malonylglycitin. Glycitin and malonylglycitin were not significantly contrasting among the progenitors. The results indicate the existence of maternal and cytoplasmatic effects on the determination of soflavone contents in soybean seeds. Such information is important for breeders during the processes of selection for high isoflavone contents, because in the presence of at least one of these effects, the success of the selection in F 2 seeds is lower than that based on the average of the F 2 plants, or on the average of the progenies. The resulted of the genetic components of the means were similar to daidzin and malonildaidzin contents,all the parameters were significant by the t test and the cytoplasmic effect was the most important one. However, for genistin content the addictive effects, of dominance deviation and of interaction cytoplasm x dominance were not significant and the cytoplasmic effect and of interaction cytoplasm x nucleus were the first and the second, respectively, most important effects. For malonylgenistin content the genetic parameters significant were: the addictive effects and of epistatic effects (cytoplasm x addictive and cytoplasm x dominant). The addictive effect was the one with the largest relative importance. Such information is especially important during the choice of the best maternal progenitor, with the best cytoplasm and interaction of the cytoplasm with the nucleus, increasing the probability of gain in the selection process. Due to the maternal and cytoplasmic effects on isoflavone contents it was necessary to estimate the heritability for that characteristic based on F 3 progenies. Heritability estimates for daidzin, genistin, malonyldaidzin, and malonylgenistin contents and for total isoflavone contents were superior to 90%, indicating a high probability of success in the selection in early generations. Heritability for protein content was 33%. The correlations coefficients for the kinds isoflavones were positives and of high magnitude (>0.80) among them, indicating that such characteristics can be breeding simultaneously. The correlations coefficients between isoflavone contents and protein contents were all negative. The phenotypic correlation between these characteristics was -0.47, agreeing with the literature.
