Expressionsanalyse, zelluläre Lokalisierung und proteinbiochemische Charakterisierung des porzinen CLCA-Homologen pCLCA4a | Unterschiede zu seinen Orthologen bei Mensch und Maus? | Expression pattern analysis, cellular localization and biochemical characterization of the porcine homolog CLCA4a | Are there differences from its human and murine orthologs?

Die CLCA (engl.: chloride channel regulators, calcium-activated) - Familie umfasst eine Gruppe heterologer Proteine, die funktionell möglicherweise eine Ca2+-abhängige Chloridleitfahigkeit an epithelialen Grenzflachen induzieren. Die CLCA-Proteine stellen dabei selbst keine echten Kanäle dar, sondern scheinen, neuen Erkenntnissen nach, eine indirekte Wirkung auf bislang unbekannte Proteine zu haben. Der humane Vertreter hCLCA4, der überwiegend im Intestinaltrakt und im Respirationstrakt exprimiert wird, scheint zudem eine bedeutende Rolle bei der Zystischen Fibrose (engl.: cystic fibrosis, CF) einzunehmen. Seine möglichen Funktionsmechanismen sind jedoch bislang noch unerforscht. Sein muriner Orthologe mCLCA6 ist mit dem mCFTR-Chloridkanal im Kolonepithel kolokalisiert, was ebenfalls für eine potenzielle Rolle als Modulator der „alternativen Chloridleitfahigkeit“ bei der CF sprechen konnte. Betrachtet man jedoch das Expressionsmuster dieser beiden nahe verwandten Vertreter, fallen gravierende speziesspezifische Unterschiede auf, da mCLCA6 im Gegensatz zu hCLCA4 nicht im CF-relevanten Respirationsepithel exprimiert ist. Daraus resultierende Funktionsunterschiede dieser beiden CLCA-Orthologen müssen daher zu diesem Zeitpunkt angenommen werden. Die somit eingeschränkte Übertragbarkeit von Daten der einen Spezies auf die andere schränkt den Gebrauch von Tiermodellen bei der translationalen Erforschung humaner Krankheiten wie der CF erheblich ein. Das bislang als Tiermodell verwendete CF-Mausmodell zeigt phänotypisch starke Abweichungen gegenüber dem humanen CF- Phänotyp. Gerade die Hauptmanifestation der humanen CF in der Lunge, deren Auswirkung bei den meisten Betroffenen der Grund für den tödlichen Krankheitsverlauf ist, fehlt im murinen CF-Modell fast gänzlich. Dadurch ist der Modellcharakter der CF-Maus zur Erforschung dieser dramatischen Erkrankung erheblich limitiert. Jüngst ist es gelungen, CF-Schweinemodelle zu generieren, die den CF-Phänotyp des Menschen wesentlich besser widerzuspiegeln scheinen. Insofern verfolgte diese Forschungsarbeit das Ziel, den porzinen Orthologen zu hCLCA4 und mCLCA6, pCLCA4a, proteinbiochemisch zu charakterisieren und sein Expressionsmuster zu ermitteln, um so einen Vergleich zwischen den Spezies zu ermöglichen und eventuell bestehende speziesspezifische Unterschiede zwischen den Orthologen aufzudecken. RT-PCR-Analysen zur Klärung dieser Frage ermittelten eine mRNA-Expression im oberen Respirationstrakt, im Intestinaltrakt und, deutlich schwächer, auch im Auge und im Uterus. Mittels spezifischer, hier erzeugter anti-pCLCA4a-Antikorper wurde das pCLCA4a-Protein immunhistochemisch im bronchialen Zilienepithel des oberen Respirationstrakts und in der apikalen Bürstensaummembran im oberen Drittel der Zotten des gesamten Dünndarms nachgewiesen. Computergestützte Analysen zur Proteinstruktur und die proteinbiochemischen Untersuchungen mittels Westernblot weisen darauf hin, dass pCLCA4a einen ähnlichen sekretorischen Weg durch die Zelle nimmt wie mCLCA6. Demnach unterliegt auch pCLCA4a den CLCA- typischen Proteinmodifikationen in Form einer posttranslationalen Spaltung des Vorläuferproteins im Endoplasmatischen Retikulum in ein 110 kDa- aminoterminales Spaltprodukt und ein 52 kDa- bzw 42 kDa-carboxy-terminales Spaltprodukt und Glykosylierungen im Endoplasmatischen Retikulum und im Golgi- Apparat. Schließlich wird pCLCA4a offensichtlich zur apikalen Zellmembran zielgeleitet, wo es mit einer carboxyterminalen Transmembrandomane membranverankert vorliegt. Lediglich das aminoterminale Spaltprodukt des pCLCA4a-Proteins kann in den extrazellularen Raum abgegeben werden. Trotz einiger hier beobachteter speziesspezifischer Abweichungen des pCLCA4a- Expressionsmusters von dem seiner humanen und murinen Orthologen besteht offensichtlich eine beträchtlich größere Homologie des Expressionsmusters in CF-relevanten Organen zwischen hCLCA4 und pCLCA4a als zwischen hCLCA4 und mCLCA6. Dies stützt die Auffassung, dass CF-Schweinemodelle geeigneter zu sein scheinen, die Rolle von hCLCA4 bei der CF zu studieren, als es durch die Verwendung der heute verfügbaren Mausmodelle möglich wäre. Die Ergebnisse dieser Forschungsarbeit konnten damit zum Verständnis der speziesspezifischen Unterschiede beim CF-Phänotypen verhelfen und ermöglichen nachfolgenden Arbeiten, die Funktionsmechanismen dieser Proteine im transepithelialen Ionentransport und ihre möglichen Modulatorfunktionen bei Krankheiten wie der CF aufzudecken.

The CLCA (chloride channel regulators, calcium-activated) family contains a group of heterologous proteins which induce a Ca2+-dependent chloride current at epithelial interfaces. The CLCA do not represent real chlorid channels but, according to new insights, they have an indirect effect on so far unknown proteins. The human CLCA4 which is predominantly located in the intestinal tract and in the respiratory tract seems to play an important role in cystic fibrosis (CF). Possible functional mechanisms of hCLCA4 have not been investigated to date. Its murine ortholog mCLCA6 is colocalized with the murine cystic fibrosis transmembrane conductance regulator (CFTR) at the apical surface of colonic crypt cells, which may argue for a potential role as modulator of the “alternative chloride current“ in CF. However, considering the expression patterns of these two closely related CLCA members, species- specific differences are obvious because mCLCA6 is, unlike hCLCA4, not expressed in CF-relevant respiratory epithelial cells. Thus, functional differences of these two CLCA orthologs have to be assumed at the moment. The limited transferability of data from one species to the other restricts the use of animal models for translational research in human diseases such as CF. The CF mouse model which has been used as an animal model for the investigation of CF so far shows considerable variations compared to the human CF phenotype. The predominant CF lung phenotype of human patients is nearly totally missing in the murine CF model. The usefulness of the CF mouse model for the investigation of this severe disease is thus limited. Recently, CF pig models have been successfully generated which seem to reflect the human CF phenotype more closely. The intention of this work was to characterize biochemical properties of the porcine ortholog to hCLCA4 and mCLCA6, pCLCA4a, and to determine its expression patterns. With this knowledge, a comparison between the species should be possible, and species-specific differences of CLCA orthologs will become obvious. RT-PCR analyses revealed strong pCLCA4a mRNA expression in the upper respiratory tract, in the intestinal tract and considerably lower in the eye and in the uterus. Using new generated specific anti-pCLCA4a antibodies, the pCLCA4a protein was immunohistochemically detected in the bronchial ciliated epithelium of the upper respiratory tract and in the brush border of enterocytes in the apical third of small intestine villi. Computer-aided predictions of the protein structure as well as biochemical investigations by immunoblotting analyses suggest a way of intracellular movement for pCLCA4a that is similar to the secretory pathway of mCLCA6. According to this, pCLCA4a is posttranscriptionally regulated similar to other CLCA proteins, being cleaved into a 110 kDa-amino-terminal and two 52 kDa- and 42 kDa-carboxy-terminal products and glycosylated in the endoplasmic reticulum and Golgi apparatus. pCLCA4a is obviously transported to the apical cell membrane where it is found anchored by a carboxy-terminal transmembrane domain. Only the amino-terminal cleavage product of the pCLCA4a protein was detected in the extracellular medium. Despite some species-specific differences between the pCLCA4a expression pattern and that of its human and murine orthologs, the expression patterns of hCLCA4 and pCLCA4a in CF relevant tissues have more in common than hCLCA4 and mCLCA6 have. This supports the idea that porcine CF models are more suitable to study the role of hCLCA4 in CF than currently available murine models are. In this respect, the results of this work could help in investigating the species-specific differences of the CF phenotype. Future studies could use these results as a basis to investigate the functional mechanisms of these proteins in the transepithelial anion conductance and to discover their potential modulatory functions in diseases such as cystic fibrosis.
