Charakterisierung des Na+-Transportes am isolierten Psalterepithel des Schafes | Characterisation of Na+ transport in the isolated omasal epithelium of sheep

Eine wichtige Funktion des Blättermagens der Wiederkäuer ist die Absorption von großen Mengen HCO3- im Austausch mit Cl-, um eine übermäßige Gasbildung in Form von CO2 im Labmagen zu verhindern und um eine ausreichende Ansäuerung der Ingesta im Labmagen gewährleisten zu können. Für den effektiven transepithelialen Transport (Absorption) von HCO3- müssen mindestens zwei Voraussetzungen gegeben sein: a) Wirksame Regulation des pHi und b) Energetisierung des apikalen Cl-/ HCO3- Austauschers. Die pH-Regulation im Blättermagenepithel erfolgt subapikal zur Kompensation der HSCFA Aufnahme wahrscheinlich zunächst primär über die Aktivität des Na+/H+-Austauschers (NHE). Die passive Aufnahme von HCO3- im Austausch mit Chlorid (Anionen- Austauscher) erfordert subapikal Chlorid und als Voraussetzung für den passiven Austausch einen entsprechenden Chloridgradienten. Dieser wird primär durch den NaCl-Cotransporter in der apikalen Membran sichergestellt, so dass die Aktivität des NaCl-Cotransporters als wichtige Voraussetzung für die Energetisierung des Bikarbonattransports anzusehen ist. Subapikales Chlorid ist zudem Voraussetzung für die Aktivität des NHE und somit für die pH- Regulation. Der HCO3- - Transport zeigt also eine eindeutige indirekte Abhängigkeit vom elektroneutralen Na+-Transport (NHE und NaCl-Cotransporter). Die vorliegende Studie bringt nähere Erkenntnisse über den elektroneutralen Na+-Transport am Blättermagenepithel des Schafes im Hinblick auf dessen Charakteristika, kinetische Eigenschaften und Regulationsmechanismen. 1\. Kinetik des elektroneutralen Na+-Transportes a. Der elektroneutrale Na+-Transport beschrieb sowohl bei einem pH-Wert von 7,4 als auch bei 6,4 auf mukosaler Seite eine Sättigungskinetik gemäß einer Michaelis-Menten-Kinetik. Eine Sättigung tritt ab ca. 40 mM Na+ ein. b. Die pH-abhängige Stimulierung des elektroneutralen Na+-Transportes wurde durch den Einsatz von Amilorid gehemmt werden, d.h. er wird über die Aktivitätssteigerung eines NHE realisiert. Die maximale Transportgeschwindigkeit (Vmax) war bei pH 6,4 signifikant größer als bei pH 7,4. c. Eine Aktivität des NHE3 war ab einer luminalen Na+-Konzentration von 12 mM nachzuweisen. Der NaCl-Cotransporter nimmt seine Aktivität erst ab etwa 25 mM Natrium auf. Bis diesen genannten Na+-Konzentrationen könnte es zu einer Einschränkung für den HCO3--Transport kommen, welcher auf einen Cl-Gradienten angewiesen ist. 2\. Differenzierung des elektroneutralen Na+-Transportes a. Der NHE-Blocker Amilorid und der NaCl- Cotransporter-Hemmstoff Hydrochlorothiazid (HCTZ) zeigten in der angewandten Dosierung von jeweils 1 mM keinen additiven Effekt auf den Na+-Transport. b. S3226, ein NHE3-Isoform-Blocker, war am Blättermagenepithel wirksam in einer Dosierung von 1 µM. Der Netto-Na+-Transport wurde durch S3226 auf die Größe des jeweiligen Kurzschlussstromes reduziert. Dementsprechend fand nach der Zugabe von 1 µM S3226 auf der mukosalen Seite kein elektroneutraler Na+-Transport mehr statt. Dies ist ein Hinweis auf die Existenz einer NHE3-Isoform an der apikalen Membran. c. 1 mM Amilorid und 1 µM S3226 zeigten einen additiven Effekt. Amilorid scheint eine geringere Sensitivität als S3226 zu haben. d. S3226 zeigte in der wirksamen Dosierung von 1 µM auf der serosalen Seite keinerlei Effekte. e. HOE 642, ein NHE1-Isoform-Blocker, zeigte keine Wirksamkeit auf der mukosalen Seite. 3\. Trafficking a. Die Zugabe von 1 nmol Wortmannin auf der mukosalen Seite hatte keinen Effekt auf den Na+-Transport. Die Stimulierung des Na+/H+-Austausches durch Absenkung des mukosalen pH-Wertes wurde nicht beeinflusst. Es gibt keinen Hinweis für Trafficking an der apikalen Membran des Blättermagenepithels als ein möglicher Mechanismus der Aktivitätssteigerung des NHE3. 4\. Korrelation von Jms und Jsm Na+ a. Jms und Jsm Na+ konnten ab einer Konzentration von über 50 mM Acetat bei einem pH von 6,4 auf der mukosalen Seite „entkoppelt“ werden. Dies bestätigt die mögliche Theorie der Existenz von zwei in Serie geschalteten Na+/Na+-Austauschern in der apikalen und basolateralen Membran, wobei der apikale Transporter in einen Na+/H+-Austauscher wechseln könnte. Dies könnte eine Art Servomechanismus für die pH-Regulation der Zelle darstellen. b. Die wachsende Differenz zwischen Jms und Jsm Na+ bei der „Entkopplung“ konnte durch 1 µM S3226 eliminiert werden, die Zunahme von Jms Na+ ist also auf die Aktivitätssteigerung einer NHE3-Isoform zurückzuführen. Art und Weise der Aktivierung bedarf weiterer Untersuchungen.

One crucial function of the ruminant‘s omasum is the absorption of huge amounts of HCO3- in exchange with chloride in order to prevent an excessive formation of gas in the abomasum. Moreover, it guarantees a sufficient drop of the pH in the abomasum. Concerning the effective transepithelial transport of HCO3- two requirements must be met: a) effective regulation of the internal pH and b) provision of the apical anion exchanger. The regulation of subapical pHi depends very likely primarily on the activity of Na+/H+ exchange, which is required for the compensation of H+ uptake via HSCFA. The absorption of HCO3- in exchange with chloride requires subapical chloride and an according chloride-gradient for the passive anion exchange. This gradient is primarily achieved through NaCl-Cotransport. Thus the activity of NaCl-Cotransport is an important condition for the provision of HCO3--Transport. Moreover subapical chloride is needed for the activity of the NHE and therefore also for pH- regulation. HCO3--Transport thus shows an indirect dependence to the electroneutral Na+ transport (NHE and NaCl-Cotransport). The underlying study provides detailed conclusions to the electroneutral Na+ transport investigating its characteristics, kinetics and regulatory mechanisms. 1\. Kinetics of the electroneutral Na+ transport a. Electroneutral Na+ transport showed with a pH of 7.4 as well as with a pH of 6.4 a saturation kinetic on the mukosal site, according to a Michaelis-Menten type kinetic. Saturation is noticed at a luminal Na+ concentration of about 40mM. b. pH-dependent stimulation of electroneutral Na+ transport could be inhibited by amiloride. This means the stimulation is realized through a rise in activity of the NHE. Maximum transport (Vmax) was higher at pH-value of 6.4 than 7.4. c. The NHE seems to start its activity at a luminal concentration of 12 mM Na+. NaCl- Cotransporter starts its activity at 25 mM Na+. Below these Na+ concentrations, a limitation of the HCO3--Transport – which depends on a gradient of chloride - could occur. 2\. Differentiation of electroneutral Na+ transport a. 1 mM of NHE-inhibitor amiloride and 1 mM of the NaCl-Cotransport- inhibitor hydrochlorothiazide (HCTZ) showed no additive effect on Na+ transport. b. S3226, an inhibitor of the NHE3-Isoform, showed effects at the omasal epithelia with a concentration of 1 µM. There was no significant difference between Jnet Na+ and the short circuit current in the presence of this inhibitor. Therefore there was no remainig electroneutral Na+ transport after the addition of 1 µM S3226 on the mucosal side. This is evidence to the existence of the NHE3-Isoform on the apical membrane of the omasum. c. 1 mM amiloride and 1 µM S3226 showed an additive effect. Amiloride seemed to have a lower sensitivity than S3226. d. In its effective dosage of 1 µM, S3226 showed no effect on the serosal side. e. The NHE1-Isoform-Inhibitor, HOE 642, showed no effect on the mukosal side. 3\. Trafficking a. The addition of 1 nmol wortmannin on the mucosal side showed no effect on the Na+ transport. There was no influence on the stimulation of Na+/H+-exchange through lowering the pH on the mucosal side. There is no evidence for trafficking on the luminal side of the omasum as a possible mechanism for rising of NHE-activity. 4\. Correlation of Jms und Jsm Na+ a. Jms and Jsm Na+ could be ‘decoupled’ using a concentration of more than 50 mM Acetat at a pH of 6.4. This supports the assumption of the possible existence of two parallel working Na+/Na+ exchangers on the apical and serosal membrane. According to this the apical Na+/Na+ exchanger could substitute in a Na+/H+ exchanger. This could be a possible kind of ‘servomechanism’ for the pH-regulation of the cell. b. The increasing difference between Jms und Jsm Na+ while the ‘decoupling’ could be eliminated through 1 µM S3226. Thus the increase of Jms Na+ is due to a rise of activity of a NHE3-Isoform. The specific way of activation needs further investigation.
