Expression von Hypoxia Inducible Factor 1 (HIF1) und HIF1-regulierten Genen unter Sauerstoffmangel und/oder erhöhter Temperatur im Herzen von Hühnerembryonen | Expression of Hypoxia Inducible Factor (HIF1) and HIF1 regulated genes under hypoxia and/or high temperature in hearts of chicken embryos

Die O2-Versorgung von Hühnerembryonen wird durch die Eischale begrenzt. Die physiologisch steigende Hypoxie ist ein wichtiger Stimulus für die embryonale Entwicklung. Es war anzunehmen, dass eine künstliche Reduzierung des O2-Gehaltes und/oder erhöhte Temperaturen zu morphologischen und metabolischen Anpassungen führen. Die Hühnerembryonen der Kontrollgruppe wurden bei 21% O2 und 37,5°C bebrütet. Die drei Versuchsgruppen wurden den Stressoren O2-Mangel (15%) und/oder erhöhten Temperaturen (39,5°C) ausgesetzt: die Hypoxiegruppe von Inkubationstag (D) 6-12, die Hyperthermiegruppe von D8-12 und die Hypoxie+Hyperthermiegruppe von D8-12. Die Massen der Embryonen und ihrer Herzen sowie der Massenverlust des Hühnereis, die Missbildungsrate und die Mortalitätsrate wurden an verschiedenen Inkubationstagen in allen Gruppen bestimmt. Die mRNA-Gehalte von Hypoxia Inducible Factor 1α (HIF1α) und der HIF1-regulierten Gene Vascular Endothelial Growth Factor A (VEGFA), Enolase 1 (ENO1), Hämoxygenase 1 (HO1), Glutathionperoxidase (GPx-3) und Hitzeschockprotein 90 (HSP90) wurden mittels quantitativer qPCR im Herzgewebe bestimmt. Die Expressionsraten der untersuchten Gene wurden anhand der Porphobilinogendesaminase (PBGD) normalisiert. Es kam in der Hypoxiegruppe zu einer Wachstumsverzögerung der Embryonen durch den Hypoxie-induzierten Hypometabolismus mit posthypoxischem Hypermetabolismus. Dem erhöhten Wachstum der Embryonen in der Hyperthermiegruppe folgte eine Wachstumsverzögerung nach Ende der Belastungsinkubation. Ein erhöhtes Wachstum ohne folgende Wachstumsverzögerung trat in der Hypoxie+Hyperthermiegruppe auf. Die relativen Herzmassen der Hypoxiegruppe waren an den Tagen D12, 18 und 20 erhöht, also an den Tagen an denen die Belastungsinkubation am längsten dauerte bzw. an denen die physiologisch steigende Hypoxie den Organismus am meisten belastete. Das Herz scheint die größten kompensatorischen Anpassungsmechanismen unter alleiniger Hypoxie zu haben. Die chronische Hyperthermie war am lebensbedrohlichsten von allen untersuchten Stressoren. Der durch die Hyperthermie aktivierte Stoffwechsel wurde durch gleichzeitige Hypoxie gebremst und hatte einen positiven Effekt auf das Überleben der Embryonen. Das ist auf die Kreuztoleranz zwischen verschiedenen Stressoren zurückzuführen. Die Hypoxiegruppe wies die geringsten und die Hyperthermiegruppe die größten Änderungen auf der mRNA-Ebene auf. Diese Arbeit zeigt, dass die Untersuchung des Gesamtherzens die organregionsspezifische, gewebs- und zelltypspezifische Regulation von HIF1 und der HIF1-regulierten Gene nicht berücksichtigt. Es ist davon auszugehen, dass auch im Herzen eine metabolische Zonierung existiert. Dass es trotzdem zu vereinzelten Genregulationen kam, lässt teilweise massive Veränderungen auf der mRNA-Ebene vermuten. Bei einer speziellen Untersuchung von Organregionen, Geweben- und Zelltypen und unter Berücksichtigung der metabolischen Zonierung sind deutliche Genregulationen anzunehmen. Die Ausbildung der Chorioallantoismembran (CAM) ist als kritischer Prozess beim Vogelembryo zu sehen. Der verfügbare O2 ist in Phase (D8-11) entscheidend für die akkurate Ausbildung der CAM und damit für das Überleben des Embryos. Aufgrund der Verteilung der Mortalität der Embryonen auf die Inkubationstage in der Hypoxiegruppe ist anzunehmen, dass das kritische Fenster für den O2-Verbrauch auf D8-11 eingeengt werden kann. Die Ergebnisse dieser Arbeit tragen zur erweiterten Kenntnis der Anpassungsmechanismen des Embryos an verschiedene Stressoren und der Kreuztoleranz bei. Die Auswirkungen verschiedener Stressoren während der embryonalen Entwicklung auf das Wachstum des Herzens und des gesamten Embryos sind bedeutend für das Verständnis später auftretender Krankheiten. Die ausgelösten Mechanismen aufgrund eines oder mehrerer Stressoren folgen dabei einem zeitlichen Verlauf. Kurzfristige Einflüsse erfolgen neuronal und humoral bzw. auf der Proteinebene. Mittelfristige Einflüsse lösen über Transkriptionsaktivatoren wie HIF Genregulationen aus. Längerfristig einwirkende Stressoren führen zu morphologischen Veränderungen, die entweder zum Überleben, Missbildungen oder zum Tod des Embryos führen.

The oxygen supply of chicken embryos is limited by the egg shell. The increasing physiological hypoxia is an important stimulus of embryonic development. The purpose of this study is to show that the artificial reduction of oxygen content and/or high ambient temperatures leads to morphological and metabolic adaptation by the organism. Chicken embryos of the control group were incubated under 21% O2 and 37.5°C. The experimental groups were incubated under hypoxia (15% O2) and/or high temperatures (39.5°C). The hypoxic group was incubated under modified conditions from incubation days (D) 6-12, the hyperthermic group from D8-12 and the combined hypoxic+hyperthermic group from D8-12. Body mass, heart mass, loss of mass of the egg, rate of malformation and mortality were all measured at different incubation days in all groups. The mRNA-levels of Hypoxia inducible factor 1α (HIF1α) and the HIF1α regulated genes: Vascular Endothelial Growth Factor A (VEGFA), Enolase 1 (ENO1), Heme Oxygenase (HO1), Glutathione Peroxidase (GPx-3) and Heat Shock Protein 90 (HSP90) in the heart of the embryos were determined via quantitative Polymerase Chain Reaction (qPCR). The expression rate of the genes of interest was normalized to Porphobilinogen Deaminase (PBGD). There was a retardation of growth due to the hypoxia-induced hypometabolism followed by a posthypoxic hypermetabolism. The increased body growth in the hyperthermic group was followed by growth retardation after the end of the stress incubation. In the hypoxic+hyperthermic group the body growth was increased without following growth restriction. The relative heart mass of the hypoxic group was elevated at D12, D18 and D20. On these days the embryos were incubated for the longest period of time. Under such conditions respectively the physiologically increasing hypoxia was stressing the heart, in which the hypoxic group seems to have the biggest compensatory mechanisms. The chronic hyperthermia was most life-threatening of all examined stress factors. The hyperthermiaactivated metabolism was limited by simultaneous hypoxia and had a positive impact on the survival of the embryos. This can be attributed to the cross tolerance between various stress factors. The mRNA levels of the genes of interest had the smallest changes in the hypoxic group and the biggest changes in the hyperthermic group. This study shows that the examination of the heart as a whole underestimates the specificity of HIF1α related gene expression regarding different organ regions, tissues and cell types. It is to be assumed that the metabolic zonation exists also in the heart. The occasional up-regulations imply considerable changes in gene expression, if taken into consideration the specific investigation of organ regions, tissues, cell types and the metabolic zonation. The formation of the chorioallantoic membrane (CAM) is to be considered a critical process of avian embryonic development. From D8-11 the available oxygen is crucial for the accurate formation of the CAM and therefore for survival. The distribution of the embryonic mortality on the incubation days in the hypoxic group suggests strongly that the critical window of oxygen consumption can be restricted on D8-11. These results contribute to the knowledge of the adaptation mechanisms of the embryonic stress response and the cross tolerance between stress factors. The influence of these various factors on the growth of the heart and the body as a whole during embryonic development are significant for the understanding of the incidence of later illnesses such as cardiovascular diseases. The mechanisms of a stress response follow a time-dependent course. A short-term response is based on neuronal and humoral reactions on the protein level. A medium-term response triggers off gene regulation e.g. via HIF1 (-induced) transcriptional activation. Long-lasting stressors induce morphological changes causing survival, malformations, or death of the embryos.
