Efeitos do anestésico tricaína metanosulfonato sobre o estresse e aspectos reprodutivos em machos de jundiá (Rhamdia quelen)

A anestesia é uma prática comum em peixes utilizados para fins de pesquisa e piscicultura. Para ambas as aplicações, é importante entender os efeitos do anestésico no animal e nos tecidos de interesse, para garantir a validade dos dados e melhorar o bem-estar animal. O anestésico sintético tricaína metanosufonato (MS-222) é o anestésico para peixe mais difundido em todo mundo. Além disso, é um dos únicos anestésicos liberados para o uso em peixes destinados ao consumo humano nos Estados Unidos pelo FDA (Food and Drug Administration). A produção de peixes em cativeiro somente é possível com reprodução artificial, e sabe-se que a manipulação em peixes é um estímulo estressor. O método mais comum de determinar a resposta ao estresse em peixes é medir mudanças no nível circulante de cortisol. Assim, o estudo teve como objetivo avaliar os efeitos de diferentes concentrações (0, 100, 200 e 300 mg/ L) do anestésico MS-222 sobre os níveis de cortisol e sua influência sobre a manutenção da qualidade espermática de machos de jundiá (Rhamdia quelen). Após indução hormonal, 29 machos sexualmente maduros, com peso médio de 363,00 ±71,24g, foram distribuídos aleatoriamente entre os tratamentos, e em seguida foram realizadas as coletas de sêmen e sangue. Os seguintes parâmetros foram avaliados: tempo de indução à anestesia, taxa de motilidade, concentração espermática, morfologia espermática, concentração de cortisol plasmático, concentração plasmática de hormônios reprodutivos (testosterona, 17αHidroxiprogesterona e estradiol). Os dados foram analisados por meio de ANOVA seguido de teste Tukey (P<0,05) para os dados paramétricos, e por Kruskal-Wallis, seguido de Dunn (P<0,05) para os dados não paramétricos. Não houve diferença estatística nos níveis de testosterona e 17αHidroxiprogesterona, bem como na morfologia espermática. No tempo de indução a anestesia, o tratamento com 100 mg/L de MS-222 apresentou maior tempo de indução (440,14±51,32s, P=0,0145) comparado aos tratamentos de 200 mg/L (283,43±46,35s) e 300 mg/L (243,38±29,54s). A taxa de motilidade espermática foi significativamente maior no grupo controle (90,00±4,47%), quando comparada ao tratamento de 300 mg/L (66,25±5,65%), porém não diferiu dos tratamentos 100 e 200 mg/L. O nível de estradiol foi significativamente menor nos peixes anestesiados com 200 mg/L (574,10±75,49 pg/mL) em relação ao controle (1189,00±212,20 pg/mL). Nos níveis de cortisol plasmático, não houve diferença entre o controle (96,86±7,08 ng/mL) e os tratamentos 100 (143,43±24,97 ng/mL), 200 (138,29±23,20 ng/mL) e 300 mg/L (182,50±42,03 ng/mL). O MS-222 anestesia o peixe por meio do bloqueio dos canais de sódio impedindo o desenvolvimento de potenciais de ação nervosa. No entanto, no presente estudo o MS-222 não inibiu a secreção de cortisol. Sabe-se que em peixes anestesiados com MS-222, os níveis de cortisol plasmático aumentam ao longo do tempo. Além disso, a indução lenta da anestesia até estágio I durante a exposição ao MS-222 pode proporcionar tempo para os peixes detectarem as propriedades químicas do fármaco. Apesar ter ocorrido redução da concentração de estradiol (200 mg/L) e da motilidade espermática em todos tratamentos, outros parâmetros reprodutivos, como a taxa de fecundação, devem ser avaliados para uma melhor compreensão de como o MS-222 influencia a reprodução de peixes.

Anesthesia is a common practice in fish used for research and pisiculture purposes. For both applications, it is important to understand the effects of the anesthetic on the animal and the tissues of interest, to ensure the validity of the data and to improve animal welfare. The synthetic anesthetic tricain methanosufonate (MS-222) is the most widespread anesthetic for fish worldwide. Besides that, it is one of the only anesthetics released for use in fish intended for human consumption in the United States by the FDA (Food and Drug Administration). The production of captive fish is only possible with artificial reproduction, and it is known that manipulation in fish is a stressor stimulus. The most common method of determining the stress response in fish is to measure changes in the circulating level of cortisol. Therefore, the purpose of this study was to evaluate the effects of different concentrations (100, 200 and 300 mg / L) of the anesthetic MS-222 on cortisol levels and their influence on the maintenance of sperm quality in males of jundia (Rhamdia quelen). After hormonal induction, 29 sexually mature males, with an average weight of 363.00 ± 71.24 g, were randomly distributed among the treatments, and their semen and blood samples were collected. The following parameters were evaluated: anesthesia induction time, motility rate, sperm concentration, sperm morphology, plasma cortisol concentration, plasma concentration of reproductive hormones (testosterone, 17α-hydroxyprogesterone and estradiol). Data were analyzed using ANOVA followed by Tukey test (P <0.05) for the parametric data, and by Kruskal-Wallis, followed by Dunn (P <0.05) for non-parametric data. There was no statistical difference in the levels of testosterone and 17α-hydroxyprogesterone, as well as sperm morphology. At the time of induction of anesthesia, the treatment with tricaine 100 mg / L presented a longer induction time (440.14 ± 51.32s, P = 0.0145) compared to treatments of 200 mg / L (283.43 ± 46 , 35s) and 300 mg / L (243.38 ± 29.54s). The sperm motility rate was significantly higher in the control group (90.00 ± 4.47%) when compared to the 300 mg / L treatment (66.25 ± 5.65%), but did not differ from treatments 100 and 200 mg / L. The level of estradiol was significantly lower in fish anesthetized with 200 mg / L (574.10 ± 75.49 pg / mL) than the control (1189.00 ± 212.20 pg / mL). In the plasma cortisol levels, there was no difference between the control (96.86 ± 7.08 ng / mL) and the treatments 100 (143.43 ± 24.97 ng / mL), 200 (138.29 ± 23.20 ng / mL) and 300 mg / L (182.50 ± 42.03 ng / mL). The MS-222 anesthetizes the fish by blocking the sodium channels, preventing the development of nerve action potentials. However, in the present study MS-222 did not inhibit cortisol secretion. It is known that in fish anesthetized with MS-222, plasma cortisol levels increase over time. In addition, slow induction of anesthesia to stage I during exposure to MS-222 may provide time for fish to detect the chemical properties of the drug. Although a reduction in the concentration of estradiol (200 mg / L) and sperm motility in all treatments has occurred, other reproductive parameters, such as fertilization rate, should be evaluated for a better understanding of how MS-222 influences fish reproduction.
