Rapid screening assay of downy mildew resistance in grapevines by application of the leaf disc method

A highly accurate and simple screening assay was developed with the goal of improving efficiency in the development of Downy Mildew Resistant cultivars. (1)The leaf disc method, which requires little labor for verification and makes it possible to control environmental variables when performing verification, was applied. (2)The inoculation condition of Plasmopara viticola was examined. As a result, the leaf position of 3 to 5 and an inoculum sporangia concentration of 10E4 /ml were determined to be suitable. (3)The inoculated leaf discs were cultured for 7 days at a temperature of 19degC. Afterwards, the disease severity was calculated on the basis of the area in which sporangia developed, and the degree of resistance was evaluated. (4)The assay result in this method corresponded with the results of researching a natural pathogeny in the field. As a result, it became possible to judge the field resistance of downy mildew accurately and simply.
