Analysis of a Japanese field isolate of Mycoplasma gallisepticum using partial nucleotide sequence of Phase-variant Protein A gene

To clarify the epidemiological characteristics of a field Mycoplasma gallisepticum isolate from Saga prefecture, Japan, we determined a partial sequence of a phase-variant protein A (pvpA) gene of the field isolate and compared it with those from 69 M. gallisepticum isolates in a database. The field isolate from Saga (SagaCK150603: Accession Number (Acc. No.): LC200413) and a Chinese isolate (CG5: Acc. No.: HQ843992) shared the highest nucleotide similarity rate of partial pvpA gene (99.5%). The nucleotide similarity rate of strain SagaCK150603 compared with strain K5831B-19 (nucleotide sequence data from patent number CA2749213 A1) was 95.5% and strain ts-11 (Acc. No.: AY556382) was 95.5%. A phylogenetic analysis using neighbor joining method showed strain SagaCK150603 belonging to a lineage consist from Chinese isolates with 94% bootstrap values, and sharing node with strain CG5. This is the first report clarifying the molecular epidemiological characteristics of a Japanese M. gallisepticum field isolate.
