Die Cysteinpeptidase HvPAP14 der Gerste und ihre Rolle beim Abbau plastidärer Proteine

Während der Seneszenz weist Gen der Papain-ähnliche Cysteinpeptidase (KDEL-Typ) HvPAP14 der Gerste eine hohe Expression auf. Zudem reichern sich die Proteinformen der HvPAP14 während der Seneszenz an, sind in geringeren Mengen aber auch in jungen Blättern zu finden. HvPAP14 konnte durch immunologische Analysen in Chloroplasten an der Thylakoidmembran lokalisiert werden. Der Abbau plastidärer Proteine ist essentiell für die Funktionserhaltung der Chloroplasten und spielt eine bedeutende Rolle bei Remobilisierungsprozessen während der Seneszenz, bei der Rubisco und Thylakoidmembranproteine die Hauptquelle des remobilisierten Stickstoffs bilden. Da Cysteinpeptidasen meist in vakuolären Kompartimenten auftreten und selten in Chloroplasten, wurde die subzelluläre Lokalisation von HvPAP14 mithilfe von RFP-Fusionsproteinen in lebenden Zellen untersucht. HvPAP14:RFP war im Endoplasmatischen Retikulum, in vesikulären Strukturen und an den Thylakoiden der Chloroplasten lokalisiert. Eine biochemische Charakterisierung rekombinanter HvPAP14 ergab, dass ihre Aktivierung bei pH 4-5 stattfindet und durch reduzierende Bedingungen begünstigt wird. Die Aktivität des reifen Enzyms wird von diesen Faktoren kaum beeinflusst. Zn2+-, Cu2+- und Fe2+-Ionen wirken inhibitorisch auf HvPAP14. Potentielle HvPAP14-Substrate wurden in in vitro-Untersuchungen auf RbcL, PSBO, LHCB1, LHCB2 bzw. LHCB3 sowie LHCB4, LHCB5 und LHCA3 eingegrenzt. In HvPAP14-überexprimierenden Protoplasten konnte der Abbau von RbcL, PSBO, LHCB1, LHCB5 und LHCA3 in vivo bestätigt werden. Der Abbau von LHC-Proteinen dient der Anpassung der Pflanzen an hohe Lichtintensitäten. In LHC-defizienten Gerstenpflanzen war die HvPAP14-Aktivierung verringert. Zudem gibt es Hinweise auf eine lichtabhängige Regulation der HvPAP14. Die Ergebnisse deuten darauf hin, dass HvPAP14 sowohl bei den Remobilisierungsprozessen während der Seneszenz als auch bei der Anpassung an die Lichtbedingungen in nicht-seneszenten Pflanzen eine Rolle spielt.

During senescence, the gene of the papain-like cysteine peptidase (KDEL-type) HvPAP14 of barley was found to be highly upregulated. Besides, the protein forms of HvPAP14 accumulate during senescence. In lower amounts the peptidase can also be found in young leaves. HvPAP14 could be localized in chloroplasts at the thylakoid membrane by immunological analyses. The degradation of plastid proteins is essential for maintaining the chloroplast functions and has a major importance for nutrient remobilization during senescence. The main sources of the mobilized nitrogen are Rubisco and the pigment-protein complexes of the thylakoid membrane. Because cysteine peptidases can usually be found in vacuolar compartments and the occurrence in chloroplasts is unexpected, the subcellular localization of HvPAP14 was analyzed in living cells with RFP-fusion proteins. HvPAP14:RFP was localized in the endoplasmic reticulum, in vesicular structures and also at the thylakoids of the chloroplasts. A biochemical characterization of recombinant HvPAP14 showed that the processing for activation of the protease occurs at a pH of 4-5 and is promoted by reducing conditions. The activity is hardly influenced by these factors. However, Zn2+, Cu2+ und Fe2+ have an inhibitory effect on HvPAP14. Potential substrates of HvPAP14 where first identified by in vitro-analyses. Beside RbcL, PSBO, LHCB1, LHCB2 or LHCB3 as well as LHCB4, LHCB5 and LHCA3 were identified as substrates. In HvPAP14-overexpressing protoplasts the degradation of RbcL, PSBO, LHCB1, LHCB5 and LHCA3 was confirmed in vivo. The degradation of LHC-proteins serves the plants´ adaption to high light intensities. In LHC-deficient barley plants the HvPAP14-activation was reduced. Besides, there is evidence of a light-dependent regulation of HvPAP14. The results suggest that HvPAP14 is involved in both nutrient remobilization processes during senescence and adaptation to light conditions in nonsenescent barley plants.
