Effect of silver nitrate and carbohydrate composition of the nutrient medium on the induction capacity of pepper (Capsicum annuum L.) in anther culture in vitro | Влияние нитрата серебра и углеводного состава питательной среды на индукционную способность перца (Capsicum annuum L.) в культуре пыльников in vitro

Currently, there are no technologies for mass production of doubled haploids (DH plants) of pepper in vitro available in the Russian Federation, which are necessary for creating new genotypes. It is important to study the factors inducing androgenesis processes in anther culture. The research aimed to study the effect of silver nitrate and the carbohydrate composition of the nutrient medium on the induction of androgenesis in pepper (Capsicum annuum L.) breeding accessions. When cultivating pepper anthers on liquid two-layer Nitsch nutrient media with the inclusion of silver nitrate, morphogenesis can occur according to the type of callusogenesis with the formation of green morphogenic structures, direct embryoidogenesis and the simultaneous formation of callus, green morphogenic structures and embryoids. All pepper genotypes had the highest number of responsive anthers with morphologically altered, overgrown walls forming morphogenic structures with subsequent initiation of callus/embryogenesis processes (from 27.3 to 79.3%), observed on a medium containing 2% maltose as a carbohydrate component, 30 mg/L glutathione in the liquid component; 1% activated carbon, 0.3% herbal gel as a gelling agent in the solid part. The advantage of using a two-layer nutrient medium in the culture of pepper anthers is due to the adsorption of all toxic substances secreted by the anther tissues by activated carbon contained in the lower layer of the nutrient medium that inhibit embryogenesis, and the development of embryoids in the liquid part of the nutrient medium as a result of direct embryogenesis from washed-out microspores. When silver nitrate was included in the liquid part of the nutrient medium at concentrations of 10 and 15 mg/L, the intensity of callusogenesis increased by 2.8-7.1%, and the efficiency of embryogenesis increased by 7.9-32.9%, depending on the genotype. The highest efficiency of embryogenesis, amounting to 32.9%, was recorded in genotype No. 2 cultured on a nutrient medium with 15 mg/L of silver nitrate.

При отсутствии в РФ технологий массового производства удвоенных гаплоидов (DH-растений) перца in vitro, необходимых для создания новых генотипов, актуально изучение факторов, индуцирующих процессы андрогенеза в культуре пыльников. Цель исследований ‒ изучение влияния нитрата серебра и углеводного состава питательной среды на индукцию андрогенеза селекционных образцов перца (Capsicum annuum L.). При культивировании пыльников перца на жидких двухслойных питательных средах Nitsch с включением нитрата серебра морфогенез может проходить по типу каллусогенеза с образованием зеленых морфогенных структур, прямого эмбриоидогенеза и одновременного образования каллуса, зеленых морфогенных структур и эмбриоидов. У всех генотипов перца наибольшее количество отзывчивых пыльников, с морфологически измененными, разросшимися стенками, формирующих морфогенные структуры, с последующей инициацией процессов каллусо/эмбриогенеза (от 27,3 до 79,3%), отмечено на среде, содержащей в качестве углеводного компонента 2% мальтозы, 30 мг/л глутатиона в жидкой составной части; 1% активированного угля, 0,3% фитогеля в качестве желеобразующего агента в твердой части. Преимущество использования двухслойной питательной среды в культуре пыльников перца обусловлено адсорбированием активированным углем, содержащемся в нижнем слое питательной среды, всех тормозящих эмбриогенез токсичных веществ, выделяемых тканями пыльника, и развитием эмбриоидов в жидкой части питательной среды в результате прямого эмбриогенеза из вымываемых микроспор. При включении нитрата серебра в концентрациях 10, 15 мг/л в состав жидкой части питательной среды отмечено увеличение интенсивности каллусогенеза на 2,8…7,1%, эффективности эмбриогенеза – на 7,9…32,9% в зависимости от генотипа. Наиболее высокая эффективность эмбриогенеза, составлявшая 32,9%, зафиксирована у генотипа № 2 на питательной среде с 15 мг/л нитрата серебра.
