Entwicklung Ahp-Cyclodepsipeptid-basierter chemischer Sonden zur Funktionskontrolle von Serinproteasen | Development of Ahp-cyclodepsipeptide-based chemical probes to monitor the function of serine proteases

Alemán. Um die Hyperaktivität von Serinproteasen, insbesondere von HTRA1, und die daraus resultierenden Krankheiten wie z.B. der altersbedingten Makula-degeneration, zu bekämpfen, sind wirksame und selektive Inhibitoren und chemische Werkzeuge zur Regulation dieser Enzyme erforderlich. In diesem Zusammenhang stellt die Naturstoffklasse der Ahp-Cyclodepsipeptide einen vielversprechenden Ausgangspunkt dar.[10,99,116] Zur effizienteren Synthese neuer Ahp-CDP wurde im Rahmen der vorliegenden Arbeit eine bestehende Festphasenpeptidsynthese [10] auf einen neuen mikrowellenbasierten Syntheseroboter übertragen und angepasst. Dieser erlaubte eine deutlich schnellere Synthese der Zielverbindungen bei einem zeitgleich geringeren Materialeinsatz. Insbesondere beim Veresterungsschritt als auch bei der Kupplung auf die N-methylierte Aminosäure in P3´ konnte eine verbesserte Reaktionsführung erreicht werden. Zudem erlaubte die Mikrowellen-basierte Synthese einen besseren Durchsatz bei der Derivatesynthese. Ziel der etablierten Synthese war dann, diese zur zielgerichteten Synthese von hochaktiven und selektiven HTRA1-Inhibitoren anzuwenden. Zur Evaluierung der Derivate wurde dabei ein automatisierter biochemischer Enzymassay sowie ein zellbiologischer β-Amyloid-Abbauassay für ausgewählte Verbindungen verwendet. Die biochemische Evaluierung ergab, dass die Variation einiger Positionen in der Peptidsequenz deutlich größeren Einfluss sowohl auf die Aktivität (P3 und P2´) wie auch die Selektivität (P4) besitzen als andere. Die Variationen der Leitsequenz sind schematisch in Abb. 77 zusammengefasst. Abb. 77: Schematische Zusammenfassung der synthetisierten Ahp-CDP ausgehend von einer Leitsequenz. Die getesteten Variationen der Positionen P1, P2 und Px mit diversen Aminosäuren (vgl. Abb. 77) führte nur zu geringfügigen Veränderung der HTRA1-Hemmaktivität gegenüber der ursprünglichen Leitstruktur. Auch beim Austausch der Aminosäuren in Position P4 zeigte sich keine signifikante Aktivitätsänderung. Jedoch führte der Einsatz von 5-Nitropicolinsäure (CDP- 14) zu einer erhöhten Selektivität gegenüber HTRA1, dies könnte ein Anhaltspunkt für die Entwicklung eines selektiveren Inhibitors sein. In P3´ zeigten die aromatischen Aminosäuren NMe-Phe (CDP- 33) und NMe-Trp (CDP- 32) die höchsten Aktivitäten. Für P3 wurde eine Vielzahl von Derivaten (vgl. Abb. 77) hergestellt. Die stärksten Inhibitionen (IC50 ≈ 1 µM) wurden mit Lysin (CDP- 12) und 3-Chlorophenylalanin (CDP- 12) erreicht. Ähnliche Variationen wurden auch für P2´ durchgeführt (vgl. Abb. 77). Dabei zeigten meta- oder para-fluorierte Phenylalaninderivate IC50-Werte von unter 1 µM. Anzumerken ist, dass die Verwendung von Aminosäuren mit sauren Resten in P3, P3´und P2´ bei niedrigen Inhibitorkonzentrationen zu Messkurven führten, die auf eine Aktivierung der Protease hindeuten. Die molekulare Basis der Aktivierung ist bisher jedoch unklar. Die Kombination der bevorzugten Aminosäuren für jede Position zeigte, dass sich die Positionen P3 und P2´ gegenseitig zu beeinflussen scheinen. Setzte man in P2´ ein Phe(4Cl) ein, so ergab sich für ein Lysin (CDP- 12) in P3 ein IC50 von unter 1 µM, bei einem Alanin in P3 (CDP- 33) hingegen lag der Wert etwa zehnmal höher. Genau umgekehrt verhielt es sich für Phe(3F) in P2´ (CDP- 55, CDP- 70). Diese Erkenntnis sollte für zukünftige Optimierung der beiden Positionen berücksichtigt werden. Durch weitere Kombinationen bekannter Aminosäuren in diesen Positionen könnten diese Wechselwirkungen möglicherweise genauer identifiziert werden. Insgesamt zeigte das Derivat CDP- 55 die beste HTRA1-Hemmaktivität (vgl. Abb. 77, IC50: 0,56 µM für HTRA1 und 0,20 µM für HTRA3). Zudem zeigten die meisten Ahp-CDP für HTRA3 die deutlich besseren Hemmaktivitäten. Die Ergebnisse des β-Amyloidassays unterschieden sich zum Teil deutlich von denen des biochemischen Assays. Während CDP- 55 im biochemischen Assay die stärkste Inhibition aufwies, konnte mit diesem Derivat im Zellkulturassay keine Hemmung detektiert werden. Die Derivate mit Phe(3,4-Cl2) (CDP- 62), Phe(2,4-Cl2) (CDP- 59) und Phe(2I) (CDP- 61) in P2´ hingegen wiesen im biochemischen Assay IC50-Werte von rund 3 µM auf. Diese führten jedoch im β-Amyloid-Assay zu starken Erhöhungen der b-Amyloid-Level von 170 % und 190 % gegenüber der Kontrollprobe, womit die bisher besten zellulären Hemmeffekte erreicht wurden. Diese deutlichen Unterschiede zwischen biochemischen und Zellassay lassen den Schluss zu, dass zumindest einige Ahp-CDPs nur eine ungenügende proteolytische Stabilität im Zellmedium haben und dort z. B. durch weitere, sekretierte Proteasen teilweise abgebaut werden. Eventuell könnte die begrenzte Stabilität der Ahp-CDPs jedoch auch therapeutisch nutzbar sein, z. B. bei Medikamenten, die nur für eine begrenzte Zeit ihr Targetenzym hemmen sollen. Bezüglich des Ziels der Aktivitäts- und Selektivitätsverbesserung der Inhibitoren lässt sich zusammenfassen, dass eine einfache Kombination „guter" Aminosäuresubstitutionen in der Ahp-CDP-Sequenz nicht zwangsläufig zu einer Verbesserung der Hemmaktivität führte. Insbesondere Veränderungen in P3 und P2´ scheinen sich gegenseitig zu beeinflussen. Eine weitere Frage für die Entwicklung der HTRA1-Inhibitoren ist, inwieweit biochemische Hemmung auch in eine zellbiologische Inhibition translatiert. So konnten biochemisch teilweise sehr potente Verbindungen dargestellt werden, welche jedoch in einem Zellkulturmodell nahezu inaktiv waren. Dementsprechend sollte über weitere Optimierungen des biochemischen Assays nachgedacht werden, um die natürlichen Verhältnisse in der EZM besser zu simulieren. Dadurch sollten die Diskrepanzen zwischen biochemischen und biologischem Assay minimiert werden. Die biser hierzu ermittelte besten Leitsequenzen sind in Abb. 78 dargestellt. Durch das TML ist es möglich, eine Peptidbindung im Makrocyclus durch einen externen Trigger zu spalten. Zu diesem Zweck wurden vorab in Lösung zwei TML-Bausteine für die Festphasen-basierte Ahp-CDP-Synthese hergestellt, eines mit und eines ohne Methylgruppe am Amin. Dabei konnte die Syntheseroute des unmethylierten TML-Bausteins gegenüber der Literatursynthese um einen Schritt verkürzt und an einigen Stellen mit verbesserter Ausbeute bzw. geringerem Materialeinsatz durch Anpassung der Equivalente und Reaktionsbedingungen optimiert werden. Zudem gelang die erstmalige Synthese eines N-methylierten Bausteins. Jedoch scheiterten sämtliche Syntheseversuche des Einbaus eines NMe-TML in ein Ahp-CDP. Auch eine Kupplung eines normalen TML-Bausteines (d.h. ohne N-Methylgruppe) in P2´ auf eine N-methylierte Aminosäure in P3´ war nicht zu realisieren. Daher wurden im Rahmen der Arbeit Ahp-CDP-Derivate mit einem TML ohne N-Methylgruppe in Position P3´ bzw. in P2´ hergestellt. Dabei lieferte die Synthese zwei Produkte (CDP- 71a, CDP- 72), die auf ihre UV-induzierte Spaltbarkeit bzw. Hemmwirkung untersucht wurden. Dabei konnte für beide Derivate nach Bestrahlung mit UV-Licht die erwarteten Spaltprodukte mittels LC-MS nachgewiesen werden. Zudem konnte in einer Nachweisreaktion eine Fmoc-Schutzgruppe auf das neu entstandene freie, terminale Amin gekuppelt werden, was auf eine erfolgreiche Spaltung des Ringsystems hinwies. Jedoch bedurfte es nach der Bestrahlung einer Equilibrierdauer von mindestens 24 h, bevor sich aus den entschützten Edukten die linearen Produkte bildeten. Trotz vollständiger Abspaltung der Schutzgruppe zeigte keines der beiden Derivate einen vollständigen Umsatz zum gewünschten linearen Produkt. Die TML-basierten Ahp-CDP (CDP- 71a, CDP- 72) wurden sowohl als HTRA1- als auch als Elastaseinhibitoren getestet. Dabei ergaben sich die stärksten Hemmeigenschaften für CDP- 71a und Elastase (IC50: 81 nM) sowie für CDP- 72 und HTRA1 (IC50: 1,2 µM) nach einer Vorinkubation von 5 min. Bei einer Verlängerung der Vorinkubation von CDP- 71a mit Elastase nahm die Aktivität der Protease im Laufe der Zeit zu, was auf einen Abbau des Inhibitors durch das Enzym schließen lässt. Dies könnte auf die fehlende N-Methylgruppe am Amin des TML zurückzuführen sein, was wiederum die Stabilität des Ahp-CDPs negativ beeinflussen kann. Anders verhielt es sich bei CDP- 71a nach UV-Bestrahlung und 48-stündiger Equilibration. Hier blieb die Hemmung bei Inkubationszeiten von 5 min - 2 h konstant und stieg erst danach deutlich an, jedoch sollte dieser Versuch zur Validierung der Ergebnisse wiederholt werden. Im Weiteren wurde die Vorinkubation auf 5 min beschränkt. Die beiden TML-basierten Ahp-CDP wurden mit UV-Licht bestrahlt und in mehreren Zeitabständen nach der Bestrahlung auf ihre Aktivität getestet. Für beide Systeme zeigte sich eine leichte Aktivitätszunahme der jeweiligen Protease direkt nach der Abspaltung der Schutzgruppe, welche einige Stunden nahezu konstant blieb, 24 h - 48 h nach der UV-Behandlung weiter zunahm und sich danach nicht weiter veränderte. Dieser Versuch zeigte somit einen leicht positiven Einfluss der Schutzgruppe auf den inhibitorischen Effekt. Zum anderen bestätigte er die Ergebnisse, dass sich die linearen Spaltprodukte erst ca. 24 h nach der Bestrahlung bilden und sich deren Anteil auch nach ca. 48 h nicht weiter erhöht. Abschließend lässt sich zusammenfassen, dass TML-basierten Ahp-CDP nur bedingt als Modell-Inhibitoren für Serinprotesen geeignet sind, da sie von der Protease abgebaut werden können. Zudem gelingt die Spaltung eines Ahp-CDP mittels TML zum linearen Peptid nur in geringen Mengen und ist zudem stark sequenzabhängig, da intramolekulare Wasserstoffbrücken im Macrocyclus sowie sterische Hinderungen die vollständige Öffnung, und damit zum Teil den Abbau des UV-entschützten Inhibitors durch die Protease, verhindern. To combat the hyperactivity of serine proteases, especially HTRA1, and the resulting diseases such as age-related macular degeneration, effective and selective inhibitors and chemical tools to regulate these enzymes are needed. In this context, the natural product class of Ahp cyclodepsipeptides represents a promising starting point.[10,99,116] For more efficient synthesis of new Ahp-CDPs, an existing solid-phase peptide synthesis [10] was transferred and adapted to a new microwave-based synthesis robot in the present work. This allowed a significantly faster synthesis of the target compounds with a simultaneously lower material input. In particular, improved reaction performance was achieved in the esterification step as well as in the coupling to the N-methylated amino acid in P3'. In addition, the microwave-based synthesis allowed a better throughput in the derivative synthesis. The goal of the established synthesis was then to apply it to the targeted synthesis of highly active and selective HTRA1 inhibitors. In this regard, an automated biochemical enzyme assay and a cell biological β-amyloid degradation assay for selected compounds were used to evaluate the derivatives. The biochemical evaluation revealed that the variation of some positions in the peptide sequence possess significantly greater influence on both activity (P3 and P2') and selectivity (P4) than others. The variations in the guide sequence are summarized schematically in Fig. 1. Fig. 1: Schematic summary of the synthesized Ahp-CDP starting from a guide sequence. The tested variations of positions P1, P2 and Px with various amino acids (see Fig. 1) resulted in only minor changes in HTRA1 inhibitory activity compared to the original lead structure. Similarly, replacement of the amino acids in position P4 showed no significant change in activity. However, the use of 5-nitropicolinic acid (CDP- 14) resulted in increased selectivity towards HTRA1, this could be a clue for the development of a more selective inhibitor. In P3', the aromatic amino acids NMe-Phe (CDP- 33) and NMe-Trp (CDP- 32) showed the highest activities. A variety of derivatives were prepared for P3 (see Fig. 1). The strongest inhibitions (IC50 ≈ 1 µM) were obtained with lysine (CDP- 12) and 3-chlorophenylalanine (CDP- 12). Similar variations were also performed for P2' (compare Fig. 1). Here, meta- or para-fluorinated phenylalanine derivatives showed IC50 values below 1 µM. It should be noted that the use of amino acids with acidic residues in P3, P3'and P2' at low inhibitor concentrations resulted in measurement curves indicating activation of the protease. However, the molecular basis of activation remains unclear to date. Combining the preferred amino acids for each position showed that the P3 and P2' positions appear to influence each other. When a Phe(4Cl) was placed in P2', the IC50 for a lysine (CDP- 12) in P3 was less than 1 µM, whereas for an alanine in P3 (CDP- 33) the value was about ten times higher. Exactly the opposite was true for Phe(3F) in P2' (CDP- 55, CDP- 70). This finding should be considered for future optimization of the two positions. Further combinations of known amino acids in these positions could potentially identify these interactions more precisely. Overall, the derivative CDP- 55 showed the best HTRA1 inhibitory activity (compare Fig. 1, IC50: 0.56 µM for HTRA1 and 0.20 µM for HTRA3). In addition, most Ahp-CDPs showed the significantly better inhibitory activities for HTRA3. Some of the results of the β-amyloid assay were significantly different from those of the biochemical assay. While CDP- 55 showed the strongest inhibition in the biochemical assay, no inhibition could be detected with this derivative in the cell culture assay. In contrast, the derivatives containing Phe(3,4-Cl2) (CDP- 62), Phe(2,4-Cl2) (CDP- 59), and Phe(2I) (CDP- 61) in P2' showed IC50 values of around 3 µM in the biochemical assay. However, these resulted in strong increases in -amyloid levels of 170% and 190% in the β-amyloid assay compared to the control sample, achieving the best cellular inhibitory effects to date. These marked differences between biochemical and cellular assays suggest that at least some Ahp-CDPs have insufficient proteolytic stability in the cell medium, where they are partially degraded by, for example, additional secreted proteases. However, possibly the limited stability of Ahp-CDPs could also be therapeutically useful, e.g. for drugs that are intended to inhibit their target enzyme only for a limited time. Regarding the goal of improving the activity and selectivity of the inhibitors, it can be summarized that a simple combination of "good" amino acid substitutions in the Ahp-CDP sequence did not necessarily improve the inhibitory activity. In particular, changes in P3 and P2' seem to influence each other. Another question for the development of HTRA1 inhibitors is to what extent biochemical inhibition also translates into cell biological inhibition. Thus, biochemically very potent compounds could be presented in some cases, which, however, were almost inactive in a cell culture model. Accordingly, further optimization of the biochemical assay should be considered to better simulate natural conditions in ECM. This should minimize the discrepancies between biochemical and biological assay. The best guide sequences determined to date are shown in Fig. 2. Furthermore, in order to obtain novel serine protease inhibitors with controllable inhibition duration via UV light irradiation, trimethyllock-based Ahp-CDPs were synthesized. UV-triggered activation of TML allows cleavage of a peptide bond in the macrocycle. Accordingly, two TML building blocks for solid-phase-based Ahp-CDP synthesis were prepared in solution, one with and one without a methyl group on the amine. The published synthesis route of the unmethylated TML building block was optimized regarding reaction yields and reagent consumption and shortened by one reaction step. In addition, the first synthesis of an N-methylated building block was achieved. However, all synthesis attempts of incorporating a NMe-TML moiety into an Ahp-CDP failed. Also, a coupling of a normal TML building block (i.e. without N-methyl group) in P2' to an N-methylated amino acid in P3' could not be realized. Therefore, in this work, Ahp-CDP derivatives with a TML residue in position P3' or in P2' lacking an N-methylamino acid were prepared instead. Two TML-incorporated Ahp-CDP products (CDP- 71a, CDP- 72) were obtained which were investigated separately for their cleavage and inhibition properties. Irradiation with UV light and analysis via LC-MS confirmed successful cleavage and ring-opening. Moreover, a Fmoc protecting group could be successfully coupled to the newly formed free terminal amine, also indicating cleavage of the ring system. However, the cleavage reaction is a slow process and an equilibration period of at least 24 h was required after irradiation for the formation of linear products. In addition, no complete transformation of the nitrobenzyl-deprotected derivative to the linear peptide could be achieved with any of the two derivatives. The TML-based Ahp-CDPs (CDP- 71a, CDP- 72) were tested for their inhibitory potential both vs. HTRA1 and elastase. CDP- 71a potently inhibited elastase (IC50: 81 nM) and CDP- 72 was a good HTRA1 inhibitor (IC50: 1.2 µM) if a preincubation period of 5 min was used. When the elastase preincubation time of CDP- 71a was prolonged, a stronger residual protease activity was observed, suggesting partial degradation of the inhibitor by the enzyme, probably due to the lack of N-methyl group on the amine of the TML. The situation was different for UV-treated CDP- 71a (48 h after irradiation) with elastase. Here, the activity did not change with incubation times of 5 min- 2 h and only increased significantly thereafter, but this experiment needs further repetition to validate the results. Due to their proteolytic instability, further biochemical experiments were therefore performed after only 5 min pre-incubation. The two TML-based Ahp-CDPs were next irradiated with UV light and tested for their inhibitory potential after different time periods. For both compounds, a slight increase in proteolytic activity immediately after deprotection was observed, that however remained almost constant for several hours, before it continued to increase again after 24 h – 48 h after UV treatment. After 48 h no further increase in inhibition was observed. These findings indicate that 1) the protecting group slightly contributed to inhibition, 2) confirmed previous findings that generation of the linear peptide requires a longer time period such as 24 h after irradiation and that formation of this product stops after 48 h. In conclusion, the generated TML-based Ahp-CDPs are unfortunately currently only of limited use as model inhibitors for serine proteases because even the non-activated derivatives are not completely stable to proteolytic degradation. In addition, cleavage of a TML-based Ahp-CDP to a linear peptide is a very slow and low yielding process, thereby also hampering the proteolytic degradation and thus triggered release of inhibition mechanism of the compounds. Clearly, further optimizations to obtain proper “inhibiton-releasable” Ahp-CDPs are still required befor they can be used as chemical tools.
