Papel del microARN-181c en el desarrollo del Proepicardio y Epicardio Embrionario

[ES] El desarrollo cardíaco es un proceso complejo y regulado, en el que el epicardio y sus derivados participan en la formación de estructuras vasculares, musculares y fibroblásticas. Durante la transición del proepicardio a epicardio embrionario se activan programas de migración, diferenciación y regulación génica. En este trabajo se ha analizado el papel del microRNA-181 (miR-181), evaluando su impacto en la expresión de genes implicados en migración y control transcripcional como Pum1, Podxl, Nfib, Sox6, Pbx3, Tbx18 o Malat1. Mediante sobreexpresión de miR-181 en células MEC1 y posterior análisis por qPCR, se observó una regulación negativa significativa en varios genes, lo que sugiere un posible papel modulador post-transcripcional. Para evaluar su efecto funcional, se realizaron ensayos de migración tipo scratch. Los resultados, cuantificados por microscopía y analizados con la prueba t de Student, no mostraron diferencias significativas entre los grupos control y tratados a las 6 y 12 horas. Estos datos indican que, aunque miR-181 regula genes relacionados con migración y transcripción, no altera directamente la motilidad celular. Dado que miR-181 está diferencialmente expresado durante esta transición, podría desempeñar un papel relevante en la regulación génica durante el desarrollo del epicardio. [EN] Heart development is a complex and tightly regulated process in which the epicardium and its derivatives play a crucial role in the formation of vascular, muscular, and fibroblastic structures. During the transition from the proepicardium to embryonic epicardium, many critical programs of cell migration, differentiation, and gene regulation are activated. This study investigates the role of microRNA-181 (miR-181) in this context, evaluating its impact on the expression of several target genes involved in both cell migration and transcriptional control, such as Pum1, Podxl, Pbx3, Tbx18, and Malat1. Overexpression of miR-181 in MEC1 epicardial cells followed by qPCR analysisrevealed significant negative regulation of several genes, suggesting a possible post-transcriptional modulation. To evaluate its functional impact, scratch migration assays were performed. Results, quantified by optical microscopy and analyzed using Student’s t-test, showed no significant differences between control and miR-181 groups at 6 and 12 hours. These findings suggest that although miR-181 regulates genes related to migration and transcription, it does not directly affect cell motility. Since miR-181 is differentially expressed during the proepicardium-to-epicardium transition, it may play a relevant role as a gene expression regulator during epicardial development.
