Detecting oxidative stress in leaves of apple tree and white clover by analysis of ascorbic acid and pigments using high performance liquid chromatography | Ugotavljanje oksidativnega stresa v listih jablane in plazeče detelje z analizo askorbinske kisline in pigmentov s tekočinsko kromatografijo visoke ločljivosti

Ascorbic acid and photosynthetic pigments content in leaves of the apple tree (Malus domestica Borkh.) subjected to drought and white clover (Trifolium repens L.) exposed to high ozone concentrations were measured by high performance liquid chromatography in order to detect oxidative stress. Measurements showed a significant difference between stressed and control plants. Water deficit increased ascorbic acid and zeaxanthin, and decreased total chlorophyll (a+b) and b-carotene in apple tree leaves. High concentrations of ozone increased ascorbic acid, total carotenoids, neoxanthin, zeaxanthin and b-carotene, and decreased violaxanthin and antheraxanthin in white clover leaves. Observed changes in ascorbic acid and photosynthetic pigment content in leaves of tested apple tree and white clover plants indicate that oxidative stress may have occured under stress conditions used in our experiments.

Z analizami askorbinske kisline in fotosintetskih pigmentov v listih jablan (Malus domestica Borkh.) izpostavljenih sušnemu stresu in v listih bele detelje (Trifolium repens L.) izpostavljene ozonu s tekočinsko kromatografijo visoke ločljivosti, smo poskušali ugotoviti oksidativni stres. Meritve so pokazale, da obstajajo statistično značilne razlike med kontrolnimi rastlinami in rastlinami izpostavljenimi stresu. V listih jablan izpostavljenih sušnemu stresu smo izmerili večje koncentracije askorbinske kisline in zeaksantina ter manjše koncentracije skupnih klorofilov (a+b) in b-karotena kot v listih kontrolnih rastlin. V listih detelje izpostavljene ozonu smo izmerili večje koncentracije askorbinske kisline, skupnih karotenoidov, neoksantina, zeaksantina in b-karotena ter manjše koncentracije violaksantina in anteraksantina kot v listi kontrolnih rastlin. Iz rezultatov lahko sklepamo, da je merjenje askorbinske kisline in fotosintetskih pigmentov v listih testiranih jablan izpostavljenih suši in detelje izpostavljene ozonu, lahko koristno orodje za ugotavljanje oksidativnega stresa, vendar pa je potrebno upoštevati tudi trajanje in intenziteto stresa.
