Fractionation of secondary metabolites extracted from Streptomyces sp. strain SSJ 6-10 (Streptomycetales:Streptomycetaceae) through preparative thin layer chromatography

A range of secondary metabolites found in the crude extract of Actinomycete bacteria have the potential to be used in pest control management. Purifying these compounds into simpler mixtures, however, may be more efficient than using the crude extract itself. Preparative Thin Layer Chromatography (Prep-TLC) is one of the purification methods that is cost-effective and is highly recommended for small-scale studies. In this study, Prep-TLC was used to fractionate secondary metabolites isolated from ethyl acetate-crude extract of Streptomyces sp. strain SSJ 6-10 and the resulting fractions were tested against 3-5 days old Spodoptera frugiperda larvae. Prep-TLC analysis revealed that the crude extract contained at least six compounds with retention factor (rf) value ranging from 0.09 - 0.91. Consequently, among the six fractions at 6000 ppm, only Fraction 4 with rf value of 0.30 demonstrated potential biological activity (29%). Preliminary spectrometry analysis using Fourier Transform Infrared Spectrophotometry (FT-IR) also suggested that fraction 1 contains alkane, ester, and amine functional groups, whereas fraction 2 mostly contains alkane and carboxylic groups. Fractions 3 and 4 have alkane and imine functional groups, while fractions 5 and 6 mostly have alkane and ester functional groups. Although no significant increase in activity was observed in this study, it still showed the potential biological activity of the different fractions of Streptomyces metabolite extracts against FAW. Therefore, further investigation into its synergistic effect with other insecticides, as well as its efficacy in different insect species, can be conducted.
