Uso da técnica de swim-up para a remoção do vírus da artrite encefalite caprina e obtenção de espermatozoides viáveis.

Os procedimentos de lavagem seminal associados a testes moleculares de alta sensibilidade para o diagnóstico do vírus da Artrite Encefalite Caprina (CAEV), como a Nested PCR e a RT-nested PCR, podem ter um impacto significante na prevenção da transmissão de enfermidades e preservação do germoplasma de reprodutores portadores do CAEV, a fim de ser utilizado sem o risco de contaminação da fêmea por via reprodutiva. Nesse cenário, objetivou-se com o presente trabalho determinar a influência da técnica de swim-up sobre a viabilidade espermática e sua eficiência na remoção de partículas virais de amostras de sêmen de reprodutores caprinos portadores do CAEV. Nesse estudo, 67 ejaculados foram avaliados antes da técnica de swim-up, quanto ao volume, concentração, motilidade individual progressiva (MIP), vigor e a presença do DNA pró-viral do CAEV por Nested PCR. Após essa etapa, as amostras foram submetidas ao teste de swim-up seguido da retirada do sobrenadante, contendo os espermatozoides, para avaliação quanto à MIP e ao vigor, como também presença de formas pró-viral e RNA viral livre do CAEV pela Nested PCR e RT-nested PCR, respectivamente, para avaliar a influência do procedimento de lavagem seminal nos parâmetros seminais. Das 67 amostras testadas por Nested PCR, antes do swim-up, 47 (70,15%) foram positivas para o DNA pró-viral. Além disso, quatro amostras adicionais foram positivas ao RT-nested PCR após o swim-up, o que permite dizer que pelo menos 76,12% (51/67) delas estavam infectadas antes da lavagem. Todavia, em 23,88% (16/67) das amostras não foram detectados a presença do CAEV antes do swim-up. Após a aplicação da técnica de swim-up constatou-se, pela Nested PCR e RT-nested PCR, que houve uma redução significativa (?²= 9,078; p< 0,001) da presença do CAEV nas amostras seminais, onde 28 de 51 amostras positivas resultaram livres do vírus (54,90%) tanto para DNA pró-viral quanto para o vírus livre. Ao demonstrar os resultados dos dois testes moleculares e avaliá-los individualmente, observou-se que em relação aos resultados da Nested PCR, o procedimento de swim-up foi capaz de remover de forma significativa (?² = 0,1965, p <0,002), a maioria das células infectadas, uma vez que 30 amostras, de um total de 47 (63,83%) positivas para a presença de DNA pró-viral pré swim-up, obtiveram resultado negativo após lavagem espermática. No que diz respeito aos resultados da RT-nested PCR, foi possível constatar que nove amostras ainda continham o vírus em sua forma livre, sendo que quatro dessas eram negativas quanto à presença de DNA pró-viral antes do swim-up. Em relação à análise das respostas obtidas, antes e depois da técnica de swim-up, sobre a MIP e vigor espermático observou-se que houve uma diminuição, significativa na média das variáveis estudadas sendo o MIP 86,42% e 4,16 para vigor antes do swim-up, e após o procedimento as variáveis estudadas obtiveram média de 71,49% para a MIP e de 3,93 para o vigor. Além disso, todos os animais tiveram pelo menos um ejaculado negativo para a presença do vírus. Desta forma, conclui-se que a eliminação do CAEV no sêmen é de caráter intermitente. Finalmente, o monitoramento da presença do CAEV no sêmen não pode se basear apenas na presença de DNA pró-viral, pois pode conter o vírus em sua forma livre, RNA viral. Portanto, a associação da Nested PCR e RT-nested PCR é uma opção segura para a certificação sanitária individual das amostras seminais quanto à presença ou ausência do CAEV. A técnica de swim-up promove uma redução na infectividade de amostras de sêmen contaminadas, entretanto deve ser associada a outros protocolos de processamento seminal para alcançar melhores resultados. Mesmo, com a aplicação do swim-up é possível promover a recuperação de espermatozoides de alta motilidade individual progressiva e vigor espermático. Abstract: The washing seminal procedures associated with highly sensible molecular tests for the diagnosis of Caprine Arthritis-Encephalitis virus (CAEV), such as Nested PCR and RT-nested PCR, may have a significant impact in preventing the transmission of diseases as well as preserving germplasm in breeding CAEV carriers in order to be used with no risk of contamination of the female reproductive tract. In this scenario, the aim of the present work was to determine the influence of the swim-up technique on sperm viability and efficiency in the removal of viral particles in semen samples from male CAEV carries goats. In this study, 67 samples were evaluated before the swim-up technique, for volume, sperm concentration, individual progressive motility (MIP), vigor and the presence of CAEV proviral DNA by Nested PCR. After this step, samples were subjected to the swim-up test followed by removal of the supernatant, containing the spermatozoa in order to be evaluated for MIP and vigor and also the presence of CAEV?s proviral and free forms of the virus by nested PCR and RT-nested PCR, respectively, as well as to evaluate the influence of the washing procedure in seminal parameters. the 67 samples tested by Nested PCR, before swim-up, 47 (70.15%) were positive for viral DNA. Furthermore, four additional samples were positive for RT-nested PCR after swim-up, which allows us to say that at least 76.12% (51/67) were infected by CAEV. However, 23.88% (16/67) of the samples did not detect the presence of CAEV before the swim-up. After application of the swim-up technique was verified by Nested PCR and RT-nested PCR, there was a significant decrease (? ² = 9.078, p <0.001) in the presence of CAEV in semen samples, once 28 of 51 positive samples resulted free from the virus (54.90%) for both DNA proviral and the free form of the virus. Taking apart the results of the two molecular tests and evaluating them individually, it was observed that concerning results of the nested PCR, the swim-up procedure was able to significantly remove (?² = 0.1965, p <0.002) most infected cells, as 30 samples, from a total of 47 (63.83%), previously shown as positive for the presence of the proviral DNA pre swim-up, were negative after sperm washing. Concerning the RT-nested PCR results, it was determined that nine samples still contained the virus in its free form, and four of these were negative for the presence of proviral DNA. Regarding the analysis of the responses obtained before and after swim-up technique on MIP and spermatic vigor, it was observed that there was a decrease, in the average of significant variables, being the IPM 86.42% and 4.16 to vigor before the swim-up, and after the procedure the studied variables had an average of 71.49% for MIP and rate of 3.93 for the vigor. Besides, all animals had at least one ejaculate negative for the presence of virus. Thus, it is concluded that the removal of CAEV in semen has an intermittent character. Finally, the monitoring of CAEV presence in semen cannot be based only on nested PCR, once negative samples for the presence of DNA proviral may contain the virus in its free form, viral RNA. Therefore, the combination of PCR-nested and RT-nested PCR is a safe option for health certification of individual semen samples for the presence or absence of CAEV. The swim-up technique promotes a reduction in the infectivity of contaminated semen samples, however it should be associated with other sperm processing protocols to achieve better results. Even, with the implementation of the swim-up it is possible to promote the recovery of high individual progressive motility sperm and sperm vigor.
