Enfoque molecular e análise computacional relacionados à qualidade e amadurecimento dos frutos de café | Molecular approach and computational analysis related to the quality and ripening of coffee fruit​

Coffee is one of the most commercialized beans in the world, grown under different conditions that differ from one region to another, such as climate, altitude, soil and light, conditions that cause changes in the desirable characteristics of the bean. Another determining factor is the species, of which Coffea arabica and Coffea canephora are the ones of commercial interest. All of this causes changes from sequential flowering, with consequent uneven fruit development and ripening, to the final content of storage compounds in the grains. Therefore, three chapters have been developed in this work to address the problems mentioned above. The first provides a general review of the subject, including its importance, phenology, ripening, harvesting and the technological tools used to monitor it. In the second chapter, a study was carried out with the aim of evaluating the effect of applying ethylene to accelerate coffee fruit ripening and homogenize the harvest, as well as to monitor and compare this effect using visual vs. images counting. The analysis of quantification and yield showed that the application of ETEFOM had no effect on ripening, which could be due to other factors. Despite this, it was possible to determine that, by means of images quantification and according to the grouped classification of the fruit, better precision was obtained with a margin of error of 4% in relation to the harvest, while in visual quantification it was 7%. The third chapter presents a review of the literature on the molecular quality of coffee, focusing on the main storage compounds, as well as the processes related to fruit ripening. This review shows that there is little information available on the gene expression that regulates these processes. Furthermore, correlations of these genes are needed in future studies to determine key points that help understand the networks of interactions that trigger the synthesis of these compounds.

O café é um dos grãos mais comercializados no mundo, cultivado sob diferentes condições que diferem de uma região para outra, como clima, altitude, solo e luz, condições essas que causam mudanças nas características desejáveis do grão. Outro fator determinante é a espécie, das quais Coffea arabica e Coffea canephora são as de interesse comercial. Tudo isso causa alterações desde a floração sequencial, com consequente desenvolvimento e amadurecimento desigual dos frutos, até o teor final de compostos de armazenamento nos grãos. Portanto, três capítulos foram desenvolvidos neste trabalho para abordar os problemas mencionados acima. No primeiro, uma revisão geral do assunto, como importância, fenologia, maturação, colheita e ferramentas tecnológicas utilizadas para o monitoramento. No segundo capítulo, foi realizado um estudo com o objetivo de avaliar o efeito da aplicação de etileno para acelerar o amadurecimento dos frutos de café e homogeneizar a colheita, bem como monitorar e comparar este efeito utilizando a contagem visual vs.imagens. A análise da quantificação e do rendimento mostrou que a aplicação de ETEFOM não teve efeito sobre a maturação, o que pode ter sido devido a outros fatores. Apesar disso, foi possível determinar que, por meio da quantificação imagens e de acordo com a classificação agrupada dos frutos, foi obtida uma melhor precisão com uma margem de erro de 4% em relação à colheita, enquanto na quantificação visual foi de 7%. O terceiro capítulo apresenta uma revisão da literatura sobre a qualidade molecular do café, com foco nos principais compostos de armazenamento, bem como nos processos relacionados à maturação do fruto. Essa revisão mostra que há pouca informação disponível sobre a expressão gênica que regula esses processos. Além disso, são necessárias correlações desses genes em estudos futuros para determinar pontos-chave que ajudem a entender as redes de interações que desencadeiam a síntese desses compostos.
