Retenção de clorofila e seus efeitos sobre a qualidade fisiológica de semente de soja | Chlorophyll retention and its effects on physiological quality of soybean seed

This study was composed by three experiments related to the effects of chlorophyll retention in soybean seeds. The first one determined the effects of water deficit and high temperature stresses during maturation on the expression level of green seeds. The second evaluated the susceptibility of different genotypes to the expression of this problem. The third determined the maximum level of green seed that can be tolerated in a seed lot after storage. With the objective of determining the stage of development of soybean plants in which environmental stresses can be imposed, resulting in the production of high levels of green seed, plants of ´CD 206´ were cultivated in green house. At the end of R6 e R7.2 plants were transferred to growth chambers (phytotrons) for imposition of water deficit and high temperature stresses. Seeds harvested at R9 had their quality evaluated by germination, tetrazolium and electrical conductivity tests. Percentage of green seed and weight of 100 seed at the upper, medium and lower thirds of each plant were also evaluated. In the second experiment, plants of BRS 133, CD 206, MG/BR 46 (Conquista) and BRS 251 (Robusta) cultivars were transferred to phytotrons at R6 for inducing water and temperature stresses. The effects of the treatments were evaluated by the percentage of green seed production and by the weight of 100 seeds in the three thirds of the plants. Contents of a, b and total chlorophylls and chlorophyllase activity were determined. The third experiment determined the maximum tolerated level of green seed in lots of CD 206 and FMT Tucunare cultivars after storage, by mixing green and yellow seed in the proportions of 0%, 3%, 6%, 9%, 12%, 15%, 20%, 30%, 40%, 50%, 75% and 100%. Seed quality was evaluated by germination, accelerated aging, tetrazolium and electrical conductivity tests. Additionally, seed contents of a, b and total chlorophylls were determined. Imposing high temperature stress (28oC to 32oC) associated with no water supply at R6 resulted in high level of expression of green seed. Intensive water stress after R6 associated with normal temperatures (19oC to 26oC) does not result in the occurrence of significant levels of green seed. The expression of green seed production under these unfavorable environmental conditions varies among cultivars: Conquista and Robusta were considered more susceptible to the production of green seeds under these stressful conditions as compared to ´BRS 133´ and ´CD 206´. These cultivars produced lower levels of green seeds, lower retention of chlorophylls and higher activity of chlorophyllase when compared with Robusta and Conquista. Soybean plants submitted to water and temperature stresses produce high levels of green seed which are small and light and have high contents of chlorophylls and low chlorophyllase activity. The contents of a, b and total chlorophylls in green seeds was inversely proportional to the activity of the enzyme chlorophyllase. The study related to the level of tolerance of green seed indicated that seed quality was negatively affected after storage. Content of chlorophyll increased with the increment of the percentage of green seed. Levels above 6% of green seed significantly reduce the quality of the seed. Seed lots with 9% or more green seed have their physiological quality significantly reduced to the point their commercialization is not advised.

Foram conduzidos três experimentos relativos à ocorrência de sementes verdes, em função de estresse hídrico e térmico, durante o período de maturação, ao comportamento de genótipos considerados tolerantes e suscetíveis ao problema e ao nível máximo de tolerância de sementes esverdeadas em lotes após armazenamento. Para verificar em qual estádio de desenvolvimento da planta a imposição de estresses propicia maiores ocorrências de sementes esverdeadas, foram cultivadas plantas da cultivar CD 206, em casa de vegetação. No final do processo de maturação das sementes, nos estádios R6 e R7.2, as plantas foram transferidas para câmaras de crescimento (fitotrons), para a indução de estresse hídrico e térmico. As sementes colhidas em R9 foram avaliadas pelo teste de germinação, de tetrazólio e de condutividade elétrica. Foram também determinados a porcentagem de semente verde e o peso de 100 sementes nos terços superior, médio e inferior de cada planta. No segundo experimento, plantas das cultivares BRS 133, CD 206, MG/BR 46 (Conquista) e BRS 251 (Robusta), produzidas em casa de vegetação, foram transferidas para fitotrons no estádio R6, para indução de estresses hídrico e térmico. Os efeitos dos tratamentos foram avaliados pelo porcentual de semente verde e pelo peso de 100 sementes nos três terços da planta. Foram também determinados os teores de clorofila a, b e total e a atividade da enzima clorofilase. O terceiro experimento envolveu a determinação do nível máximo de tolerância de sementes esverdeadas, das cultivares CD 206 e FMT Tucunaré, após armazenamento, mescladas com sementes amarelas, nas proporções de 0%, 3%, 6%, 9%, 12%, 15%, 20%, 30%, 40%, 50%, 75% e 100%. A qualidade das sementes foi avaliada pelos testes de germinação, envelhecimento acelerado, tetrazólio e condutividade elétrica. Determinaram-se, ainda, os teores de clorofila a, b, e total nas sementes. A imposição de estresses de elevada temperatura (28oC a 32oC) e o corte total de irrigação, no estádio de desenvolvimento R6, propiciam acentuada ocorrência de sementes verdes. Estresse hídrico intenso (20% de Ug e corte total de irrigação) a partir de R6, quando associado com temperaturas amenas (19oC a 26oC), não resulta na ocorrência de índices significativos de semente verde. As cultivares Conquista e Robusta são suscetíveis à retenção de clorofilas nas sementes e a ´BRS 133´ é menos suscetível às condições de estresse que causam o esverdeamento. As cultivares BRS 133 e CD 206 apresentam menor índice de semente verde, menor retenção de clorofilas a, b e total e maior atividade da enzima clorofilase, em relação às cultivares Conquista e Robusta, sob condição de estresse. Plantas de soja submetidas às condições de estresse hídrico e térmico produzem altos índices de sementes verdes, menores e mais leves, com elevado teor de clorofilas e baixa atividade de clorofilase. O conteúdo de clorofilas a, b e total em sementes verdes de soja é inversamente proporcional à atividade da enzima clorofilase. Com relação aos níveis de tolerância de sementes esverdeadas, foi observado que a qualidade das sementes foi afetada negativamente após o armazenamento e os teores de clorofila se elevaram com o aumento dos porcentuais de sementes esverdeadas. A partir de 6% de sementes esverdeadas, a sua qualidade apresenta reduções significativas. Níveis superiores a 9% comprometem a qualidade de lotes de sementes de soja para fins comerciais, por seus efeitos significativos na redução do potencial fisiológico.
