Animal botulism in Poland – laboratory and epidemiological investigations

The aim of the study was to present cases of botulism in animals found in Poland in 2019–2021. The analytical laboratory diagnosis and difficulties that occurred in the interpretation of the results are described. From 2019 to 2021, samples of serum, intestinal content, liver, spleen, kidney, faeces, wet feed, dry feed, ensilage, water and mixed samples of internal organs associated with 10 suspected animal botulism cases were sent to the National Veterinary Research Institute. Samples were analysed using a mouse bioassay and culture methods in combination with ntnh and bont gene detection. Among the ten putative botulism cases, only four (40%) were confirmed in the laboratory on the basis of the detection of botulinum toxin (BoNT) or the ntnh or bont genes. The remaining six (60%) were determined as probable despite observable characteristic clinical signs. The diagnosis of botulism in animals is a very difficult task, made so by the heterogeneity of Clostridium botulinum strains and possible loss of toxinogenicity during laboratory processing or the potential degradation of toxins. Laboratory diagnosis is a complex and problematic process which should utilise different prescribed methods for specific types of sample.

Fasciola hepatica is a trematode infecting ruminants worldwide and occasionally affecting other animal species, including humans. It causes significant economic losses. Geographic distribution and patterns of infection must be considered before control and management measures are developed for this parasite. DNA molecular markers are useful for the identification of flukes and elucidation of their genetic evolution. Therefore, the population structure of F. hepatica was studied using this method in sheep in Xinjiang, China. The molecular characteristics, genetic relationships within the population and dispersal patterns of F. hepatica isolates were analysed based on the cox1 and nad1 genes. The population structure of F. hepatica from three regions of Xinjiang was explored and a neutrality test was conducted. The cox1 and nad1 genes have 21 and 42 variable sites, respectively, which can be classified into 34 and 33 haplotypes. Median-joining network and phylogenetic tree analyses showed that there was no significant variation in F. hepatica isolates between the three geographical regions. Analysis of variance revealed that the genetic variation of F. hepatica was mainly present within the populations. The neutrality test indicated that the populations were relatively stable but the Hami population may have undergone short-term expansion. This study revealed for the first time the molecular characteristics, genetic diversity and dispersal patterns of F. hepatica isolates from sheep in Xinjiang, thus providing new insights into the genetic variation and haplotype diversity of F. hepatica from indigenous sheep.

Objective: The study aimed to determine the effect of using a combination of cassava tuber (Manihot esculenta) and Indigofera zollingeriana leaf flour as an energy source supplemented with citric acid in the ration on performance, carcass quality, digesta pH, viscosity, and the number of villi in the intestines of broilers. Materials and Methods: The research design was completely randomized with five treatments and five replications, each containing four broilers. The treatment was a substitution of corn in the ration with a combination of cassava tuber and I. zollingeriana leaf (CIF): without CIF, 5% CIF substitute for corn + 0.2% citric acid, 10% CIF substitute for corn + 0.2% citric acid, 15% CIF corn substitute + 0.2% citric acid, and CIF corn substitute + 0.2% citric acid. Each treatment ration was supplemented with 0.2% citric acid. The variables that were looked at were ration consumption, body weight gain, feed conversion, live weight, percentage of the carcass, percentage of abdomi¬nal fat, pH, viscosity, and the number of broiler villi. Results: This study showed that the combination of CIF flour supplemented with 0.2% citric acid had a significant effect (p < 0.05) on body weight gain, ration conversion, live weight, carcass percentage, and the number of villi in broiler intestines. But it did not have a significant effect (p > 0.05) on the amount of feed eaten, the amount of fat in the abdomen, the pH of the digesta, or the thickness of the broilers' blood. Conclusions: The combination of CIF flour at a level of 10% supplemented with 0.2% citric acid can be used as an energy source to replace corn without having a bad effect on production performance, carcass quality, and small intestine characteristics of broilers. Objectives: Domestic pigeons (Columba livia domestica) have diverse plumage pigmentations. Melanocortin 1 receptor (MC1R) gene variation has been correlated with color traits. The associa¬tion between MC1R and plumage coloration in African domestic pigeons is yet to be investigated. Materials and Methods: Herein, we report the relationships between single nucleotide polymor¬phisms (SNPs) in MC1R and plumage of 35 domestic pigeons from Nigeria with 4 different plum¬age phenotypes plus 37 published MC1R sequences from France (n = 14) and Russia (n = 11). Results: We obtained 14 SNP sites among 72 individuals. Missense mutations C206T (Ser69Leu) and G253A (Val85Met) were observed in 16 and 8 Nigerian pigeons, respectively. The chi-squared test (p < 0.05) for C206T, G253A, and A520G has the advantage of homozygous genotypes CC, GG, and AA, respectively. The association of C206T loci showed the advantage of CC genotype in ash-red, spread, and white pigeons, and TT in blue-bar, spread, and white feather pigeons. For G253A and A520G loci, GG and AA were dominant in all plumages except for genotype AA in G253A, which was prominent in ash-red, spread, and white plumages. The three SNPs were assigned to seven haplotypes. The median-joining network revealed 20 haplotypes, including 5 in Nigeria and 2 shared. Conclusion: This study provides an insight into the association of MC1R variation and plumage diversity in Nigerian domestic pigeons. However, due to the limitation of the current data, we could not make further conclusions; this necessitates the need for more genomics studies on Nigerian pigeons. Objective: This study has been designed to determine the effectiveness of heterologous plate¬let-rich plasma (hPRP) against infected wounds in rabbits. Materials and Methods: Staphylococcus aureus contamination was present in 24 surgical inci¬sions, each 8 mm in diameter. The animals were then divided into two groups as follows: group A, also known as the hPRP group, received topically applied, freshly manufactured hPRP twice weekly, and group B, also known as the control group, only received sterile saline. Evaluations of the histological architecture of wounds, posttherapeutic morphology, morphometry, and in-vitro and in-vivo antimicrobial potentials of hPRP were made. Results: Rabbits that were given hPRP exhibited quicker rates of wound contraction and shorter healing times. The samples from day 7 in the hPRP group showed less inflammation and more structured fibroblasts than those from the control wounds, according to histological analysis. On day 21 of the histological examination, the hPRP group's epidermis showed notable thickening. As demonstrated by in-vitro antibacterial activity, undiluted hPRP successfully suppressed S. aureus growth. A serum biochemical analysis showed that hPRP had no harmful effects on the liver or kidneys. Conclusions: Based on the findings of the histological features, antibacterial properties, and wound morphology, it can be said that hPRP gel holds promise as an alternative to antibiotics for the treatment of wound infections. Objective: The treatment of syncope depends largely on its possible etiology. Therefore, identifying the cause of syncope is very important in treatment planning. Herein, we report an etiology of syncope caused by pulmonary hypertension (PH) associated with canine filariasis, followed by syncope due to bradyarrhythmia 1 year later. Materials and Methods: An 8-year-old male English Cocker Spaniel was referred to our hospital for a second opinion regarding syncope that the dog had started experiencing approximately 2 months prior. Based on the examination findings, we diagnosed that the fainting was due to heartworm disease and associated PH. After increasing the dose of pimobendan (0.50 mg/kg, q12h), the syncope subsided. However, syncope recurred on the 215th day of the first episode. Results: The findings that differed from those during the initial examination were that car¬diac arrest was observed for approximately 5 sec during auscultation, along with sinus arrest. Therefore, to further investigate for syncope, a Holter electrocardiograph was obtained for 3 days. Consequently, sinus arrest was identified as the etiology of the recurrent syncope, and the patient was diagnosed with sick sinus syndrome, Rubenstein classification type II. Following cilostazol (10 mg/kg, q12h) administration, the syncope subsided. Conclusion: This case reports syncope in a dog, which typically occurs due to different etiologies. When a dog has PH, it may be important to think about the possibility of arrhythmias caused by a bigger right heart. [J Adv Vet Anim Res 2022; 9(3.000): 440-446]

Objective: This research aims to examine the effect of different fermentation times on the results of scanning electron microscope energy dispersive X-ray (SEM-EDX), chemical, organoleptic, and microbiological quality of banana peel. Materials and Methods: The design in this study used a completely randomized design (four treatments and four replications). The treatments in this study were different durations of fer¬mentation of banana peels; T0 = no fermentation; T1 = 6 days; T2 = 12 days; and T3 = 18 days. The research parameters were chemical, organoleptic, microbiological quality, composition, and SEM-EDX images. Analysis of chemical, organoleptic, and microbiological quality data was done using analysis of variance, followed by Duncan's multiple range testing at the 5% significance level, while the elemental composition and SEM-EDX images were analyzed descriptively. Results: The results of the study showed that the length of fermentation had a significant effect (p < 0.05) on moisture content, ash, crude fiber, nitrogen-free extract, and all organoleptic quality variables of banana peels. Different fermentation durations had no effect (p > 0.05) on crude protein, ether extract, metabolic energy, total lactic acid bacteria, total bacteria, Gram positive or negative bacteria, and total fungi on banana peels. The analysis showed that the elemental composition of banana peels is C, Na2O, Cl, K2O, MgO, SO3, SiO2, and P2O5. Conclusion: Eighteen days of fermentation improved the chemical quality, organoleptic, microbi¬ological, elemental composition, and SEM-EDX image of fermented banana peel. Objective: The increase and prevalence of multidrug-resistant bacteria in livestock animals are serious public health concerns. This study aimed to identify the presence of the blaTEM gene in extended-spectrum beta-lactamase (ESBL)-producing Escherichia coli isolated from rectal swabs of apparently healthy pigs in Malang District, East Java, Indonesia. Materials and Methods: A total of 120 rectal swab samples were collected from the pigs. The rectal swabs were screened for the presence of E. coli using standard microbiological identifi¬cation procedures. The Kirby–Bauer disk diffusion method identified multidrug-resistant E. coli. Five different classes of antibiotics were used to identify multidrug-resistant isolates, including Ciprofloxacin, Trimethoprim, Tetracycline, Streptomycin, and Aztreonam. Multidrug-resistant E. coli isolates were characterized for the presence of ESBL using double-disk synergy test methods. The presence of blaTEM genes was determined using polymerase chain reaction methods. Results: The results of this study indicated that 107 (89.2%) out of 120 samples analyzed were positive for E. coli isolates. A total of 32 (29.9%) E. coli isolates were identified to be multidrug-re¬sistant and further subjected to molecular testing. The molecular analysis revealed (5; 15.6%) E. coli isolates to harbor the blaTEM gene. Conclusion: The results of this study revealed that pigs and products of pork origin must be considered a source of transmission of ESBL-producing E. coli to public health important under the food chain. Objective: The study was conducted to determine the marbling score, fat and meat color, choles¬terol, high-density lipoprotein (HDL), low-density lipoprotein (LDL), and physical–chemical con¬tent of male Bali beef fed fermented pineapple peel. Materials and Methods: Twelve heads of male Bali cattle with an initial weight of 168.46 ± 11.95 kg were put into individual cages at random based on a completely randomized design with three treatments and four heads of Bali cattle as replicates, namely T0 = NG + (39% MC + 61% RB + 0% fermented pineapple peel); T1 = NG + (10% MC + 70% RB + 20% fermented pineapple peel with yeast culture); and T2 = NG + (15% MG + 65% RB + 20% pineapple peel fermented by lactic acid bacteria). The sample of Bali cattle meat used in testing the research variables was the LD muscle, with as many as 24 samples for each treatment. The data were analyzed based on the analysis of variance using the Statistical Product and Service Solutions software program, following Duncan's test with 5% confidence. Results: The results showed that the treatment T2 could increase the marbling quality of the meat from 2.58% to 4.00%. The cholesterol content (80 mg/100 gm), HDL (60 mg/100 gm), LDL (10 mg/100 gm), water-holding capacity (36.10%), cooking loss (29.16%), tenderness/shear force (4.08 kg/cm2), crude protein (22.99%), crude fat (4.23%), and meat collagen (1.65%) were determined. Conclusions: Adding 20% of fermented pineapple peel by lactic acid bacteria to the ration can improve the quality of marbling, produce cholesterol, and the physical–chemical value of meat that meets the Indonesian National Standard. Objectives: The purpose of this research was to look into the impacts after the implication of feeding broiler chickens with spirulina in arsenic-incited toxicities. Materials and Methods: Birds (n = 125) were distributed equally (n = 25) into four groups treated (T1, T2, T3, T4) and a group controlled, T0 (normal feed and water without supplement), the group taking in arsenic trioxide (100 mg/l)-induced diet (T1), and the groups T2, T3, and T4 (feed supplemented with 50, 100, and 200 mg/l of spirulina along with Arsenic Trioxide, respectively). The body weight and hematobiochemical parameters were recorded every 7 days. Results: Different growth development indicators, e.g., body weight, feed intake ratio, feed conversion ratio, depression, and skin lesions, were weak in arsenic trioxide groups and upstanding in the arsenic plus spirulina group. Over and above, the lack of body weight gain in chicken (2.7%–13.00%) in the arsenic-introduced groups given spirulina (T2, T3, and T4) overtook the mere groups exposed to arsenic, where the lack of weight gain was optimum (54.90%). Thereafter, in arsenic-instituted groups given spirulina (T2, T3, and T4), the drop in total erythrocyte count, total leukocyte count, hemoglobin, and packed cell volume values became less notable than in arsenic pollutant groups (T1, p < 0.01). Two measurable factors (serum glutamate pyruvate transaminase and serum glutamic oxaloacetic transaminase) were substantially (p < 0.01) raised in the group (T1) treated with arsenic, but in the arsenic-induced groups (T2, T3, and T4) treated with spirulina, they were elevated less. Conclusion: This study demonstrates that arsenic is a threat to poultry. However, spirulina may be advantageous for alleviating the effects of arsenic in poultry.
