Antibacterial efficacy of ethanolic extract of Camellia sinensis and Azadirachta indica leaves on methicillin-resistant Staphylococcus aureus and shiga-toxigenic Escherichia coli

Objective: This study aims at investigating the antibacterial potential of ethanolic extract of Camellia sinensis (common name: Green tea) and Azadirachta indica (common name: Neem) leaves on methicillin-resistant Staphylococcus aureus (MRSA) and shiga-toxigenic Escherichia coli (STEC). Materials and Methods: Fresh leaves were processed and extracted by 99% ethanol and recon¬stituted with 50% ethanol before testing. Disk diffusion and broth microdilution methods were used to determine zone diameter of inhibition (ZDI) and minimum inhibitory concentration (MIC), respectively. Nutrient agar plate was used to estimate the minimum bactericidal concentration (MBC). Results: Maximum ZDI value was observed for green tea against MRSA (7.5 mm) and minimum for neem (4.9 mm). Moreover, the highest ZDI against STEC was also for green tea and the com¬bination of green tea and neem (4.5 mm). The MIC values of green tea extract were 15.625 and 31.25 mg/ml against MRSA and STEC, respectively, whereas the MIC of neem was 31.25 and 125 mg/ml, respectively. The combination had similar MIC (46.87 mg/ml) against both organ¬isms. Green tea showed the lowest MBC values, 31.25 and 62.5 mg/ml, against MRSA and STEC, respectively. However, MBC of neem and the combination against MRSA and STEC were found >250 mg/ml, >500 mg/ml and 93.75 mg/ml, >375 mg/ml, respectively. Conclusion: Green tea and neem leaves showed good antimicrobial effects and can be used to explore novel antimicrobial compounds against MRSA and STEC. [J Adv Vet Anim Res 2019; 6(2.000): 247-252]

Objective: The study was carried out to detect the carriage of methicillin-resistant Staphylococcus aureus (MRSA) and their antimicrobial susceptibilities in village chickens sold at Maiduguri poultry markets using phenotypic characterization. Materials and Methods: This was a cross-sectional study where 120 samples comprised 50% each of Nasal and cloacal swabs, were, respectively, collected from live village chickens sold at Maiduguri poultry markets and examined for the presence MRSA based on bacterial culture, biochemical characteristics, growth on oxacillin resistant screening agar base, and antimicrobial susceptibility tests. Results: The study revealed an overall occurrence of 38.33% S. aureus and 32.60% MRSA, respectively. Antimicrobial susceptibility test was carried out on MRSA positive isolates against seven antimicrobials. All the isolates (100%) exhibited resistance against cefoxitin, whereas the least antimicrobial resistance was recorded against erythromycin and streptomycin each with 26.6%, respectively. In the same way, the highest antibiotic susceptibility in this study was observed against erythromycin (60%) and least susceptibility was against vancomycin and streptomycin with 20% each. A varying intermediate antibiotic susceptibility ranging from 13.33% to 53.33% was observed. Multiple-drug resistance patterns were exhibited by MRSA isolates from this study with 73.3% of the isolates exhibiting resistance to two or more antibiotics. Conclusion: This study has shown the carriage of MRSA by village chickens which calls for serious public health concern and concludes that these birds might have acquired these pathogens from the environment or infected humans since they normally receive no less medical attention. Objective: Jicama (Pachyrhizus erosus) fiber has been documented to exert an immunomodu¬latory effect both in vitro and in vivo. However, its beneficial effect against metabolic syndrome remains unknown. This study aimed to reveal whether the jicama fiber (JF) could prevent the development of diabetes and obesity caused by a high-sugar diet (HSD). Materials and Methods: The JF was isolated from its tuberous part and subsequently used as a supplemental diet for adult male Bagg and Albino (BALB)/c mice fed with a HSD. Four different diet paradigms including normal diet, HSD (30% sucrose), and HSD in combination with 10% and 25% of JF, respectively, were deployed continuously for 8 weeks. Furthermore, the blood glucose level, glucose tolerance, body weight, food and water consumption as well as epididymal white adipose tissue (WAT) and interscapular brown adipose tissue (BAT) mass were determined. Results: Our results revealed that supplementation of 25% JF could significantly prevent the blood glucose increase, excessive body weight gain, and glucose intolerance in mice fed with HSD. Moreover, 10% and 25% JF blunted the HSD-induced WAT mass gain but failed to counteract the depletion of BAT mass. Furthermore, the fiber supplementation elicited a minimum effect on rhythm and total food and water intake. Conclusion: The JF could effectively sustain blood glucose homeostasis as well as improve body weight and WAT mass profile against the development of diabetes and obesity caused by HSD. Objective: The study was carried out to develop a wet litter model with magnesium chloride to assess the effectiveness of a polyherbal formulation (PHF) on growth performance, litter and cecal moisture (LCM) level, cecal consistency (CC) score, and footpad lesions (FPLs) score in Ross 308 broiler chickens. Materials and Methods: 1,200 one-day-old chicks were assigned into five groups: normal control, negative control [NTC; treated with 1.7% magnesium chloride hexahydrate (MgCl2.6H2O)], and three treatment groups, T1, T2, and T3, where 750, 1,000, and 2,000 gm/ton of PHF, respectively, were supplemented. All the groups were fed a basal diet until day 7. However, the NTC and treat¬ment groups were fed a diet with MgCl2 from days 8 to 42. Results: The addition of MgCl2 for 35 days worsened the growth performance traits in broilers and induced wet litter problems (FPL, high LCM, and poor CC) in the NTC group. However, PHF (750, 1,000, and 2,000 gm/ton) ameliorated the negative effect of a diet with MgCl2 on growth perfor¬mance and wet litter problems, but a better response with respect to LCM and CC was observed in 2,000 gm/ton of PHF group, followed by that in 1,000 gm/ton of PHF group and 750 gm/ton of PHF group on day 42. Conclusion: The wet litter broiler model was developed through excessive feeding of MgCl2, which caused the performance parameters to worsen and the emergence of problems associated with the wet litter. Objective: This study was conducted to investigate different respiratory diseases in broiler and sonali birds in some selected districts of Bangladesh. Materials and Methods: We were collected a total of 460 blood samples from 46 farms with 36 broiler farms and 10 sonali farms (cross-breed) from 2015 to 2017. All the collected serum sam¬ples were tested for determining specific antibodies of avian rhinotracheitis (ART) virus, infectious laryngotracheitis (ILT) virus, infectious bronchitis (IBV) virus, and Ornithobacterium rhinotracheale (ORT) infection using commercially available enzyme-linked immunosorbent assay kits. Results: The overall seropositivity was highest in ORT (45.9%), followed by IBV (37.6%), ART (2.6%), and ILT (0.4%). Out of 360 broiler samples, highest seropositivity was recorded in ORT (43.3%) and lowest in IBV (31.4%). Surprisingly, no broiler samples were found positive for ART and ILT. In case of sonali, the seropositivity was highest in IBV (60%) and lowest in ILT (2%). With respect to types of birds and age groups, the seropositive percentage of all four pathogens was found higher in sonali than broiler. Between two age groups of sonali, the seropositive percentage of ART (12%), ORT (55%), ILT (2%), and IBV (60%) was highest at 2160 weeks of age compared to 520 weeks of age. However, based on location, the seropositive of ORT and IBV was highest in Jamalpur (63.3%) and Fulbariya and Trishal (50%) and lowest in Sreepur (16.7%) and Jamalpur (3.3%). Conclusion: The four pathogens are ubiquitous in nature for the sonali chickens, and the prev¬alence of ORT and IBV was the most prevalent viruses in the study areas. This study indicates a need for improved surveillance and characterization of ORT and ART circulating in all types of poultry in Bangladesh. [J Adv Vet Anim Res 2019; 6(4.000): 561-566]

Objective: This study aimed to evaluate the respiratory methane emission and ultimate methane yield (B0) of goat feces that fed roughage consisted of Pennisetum purpureum and Gliricidia) and fed roughage and concentrate with different protein source in the ration (fish meal and soybean meal). Materials and Methods: Fifteen Kacang bucks were allocated to the control group (T0): goats were fed roughage only, T1: goats were fed roughage and concentrate with fish meal as protein sources, and T2: goats were fed roughage and concentrate and the protein source in the ration was soybean meal. Results: The protein content of feces from T0 was significantly lower (p < 0.05) than that from the other treatments. The same phenomenon was also found in the respiratory methane emission in terms of l/head/d, l/kg digestible dry matter, and l/kg body weight. However, there was no signif¬icant effect (p > 0.05) of different ration composition on the ultimate methane yield (B0) of goat feces. This study found that B0 of goat feces from treatment T0, T1, and T2 was 17.40%, 25.78%, and 61.29%, respectively, higher than that from the international default value for developing countries. Conclusion: Feeding grass and legume can reduce methane respiration emission in goat. B0 of feces in the present study was higher than that in the international default value; therefore, the potential emission of goat manure in tropical developing countries could be higher than that in the present estimation. Objective: The purpose of this study was to evaluate the clinical outcome of surgical decompression and rehabilitation therapy in dogs with thoracolumbar intervertebral disk herniation (IVDH). Materials and Methods: After surgery, physiotherapeutic rehabilitation was performed by a combination of electrotherapy, infrared therapy, training for standing, deep tendon reflex, and aquatic treadmill exercise. A total of 186 dogs were selected from the hospital records and included in two groups: the rehabilitated group (RG, n = 96) and non-rehabilitated group (NRG, n = 90). Dogs in each group were subdivided into three groups based on a pre-operative clinical severity grading system and those in grades 24 were included in this study. Post-operative neurologic functions, unassisted standing, walking, and the success rate of both groups were evaluated and compared Results: Overall, 86.46% (83/96) of dogs had a successful neurologic outcome in the RG group, which was significantly (p < 0.01) higher than the NRG group 52.22% (47/90). Interestingly, the success rate differed when the preoperative grading system was considered. The success rates of grades 2, 3, and 4 were 97.14% (34/35), 97.33% (42/45), and 43.75% (7/16), respectively, in the rehabilitated groups, whereas in the non-rehabilitated groups, success rates were 82.35% (28/34), 51.85% (14/27), and 17.24% (5/29), respectively. The differences in success rates among the groups according to grading were 14.79%, 41.48%, and 26.51%, respectively, indicating that the proposed rehabilitation therapy is remarkably advantageous for increasing the success rate. Conclusion: Rehabilitation therapy after surgical decompression of thoracolumbar IVDH improves neurologic functions and increases the success rate, especially when the preoperative pathological condition is severe. Zika virus, a member of Flaviviridae is the etiology of Zika or Zika fever or Zika virus (ZIKV) disease characterized by mild symptoms similar to very mild form of Dengue or Chikungunya. The virus transmits through Aedes mosquitoes, particularly by Aedes aegypti. The most dangerous effect of ZIKV infection is the ability of the virus to cause microcephaly and congenital malformation to the newborn baby if the mother is infected. The neurological disorders including Guillain-Barre syndrome might be associated with adults and children due to ZIKV infections. Zika has emerged as a serious global public health problem as it has been found in 87 countries, particularly in Africa, America, and Asia and has no vaccine and treatment so far. Bangladesh is at a high risk of ZIKV infection and we consider ZIKV as a possible emerging threat for Bangladesh. This short review summarizes the insights of ZIKV infection, present status of the disease in Bangladesh and its neighboring countries, and recommendations for necessary preparations and strategies to be taken for effective controlling of the ZIKV infection in Bangladesh before getting any havoc. Objective: The current research was carried out to evaluate the use of magnesium hydroxide as buffer to control acidosis in rumen culture fermenting carbohydrates in vitro. Materials and Methods: The experiments were carried out in the chemostat system in which the reactor used was a 200 ml of working volume. A series of fed-batch trials were carried out in fed-batch system with hydraulic retention time of 4 days. All digesters were completely mixed with the rotation of 55 rpm, and the temperature was controlled at 39°C ± 0.5°C. Results: Results showed that the supplementation of magnesium hydroxide (50 mM/day) to the corn starch feed (12.5 gm/l per day) for the rumen culture could prevent acidosis while at the same concentration of sodium bicarbonate addition to rumen culture, acidosis cannot be prevented in which lactic acid accumulated up to 200 mM. Supplementing magnesium hydroxide to the mixture of starch and sugar feeds prevented acidosis in which the major fermentation end product formed was acetate. A daily feeding with the ratio of 4.5:1 [starch: Mg(OH)2 ] was feasible to prevent rumen acidosis. Conclusion: Magnesium hydroxide added to the rumen culture could prevent lactic acid accumulation while sodium bicarbonate supplementation did not prevent acidosis and had lactic acid accumulation. Objective: The purpose of this study was to characterize the mitochondrial COX-1 gene of Sarcoptes scabiei in rabbits from three districts of Malang, Nganjuk, and Kediri, East Java, Indonesia. The gene was aligned with a DNA isolated from S. scabiei of Chongqing rabbit (accession number: EU256388.1) to construct a molecular analysis of phylogenetic in S. scabiei COX-1 gene. Materials and Methods: This study has been verified by the Committee Ethics (Faculty of Veterinary Medicine, Universitas Airlangga). The mites were collected and identified from rabbits that have an indication of scabies infection. DNA was extracted with QIAamp DNA mini kit and polymerase chain reaction (PCR) analysis was done. The PCR products were purified with the pro¬tocol of the BigDye XTerminator Purification Kit (Thermo Scientific) and were double-sequenced with the forward and reverse PCR primers of ABI PRISM 310 Genetic Analyzer. The sequence prod¬uct was confirmed with Clone Manager Professional 9 (Sci-Ed Software) and the Neighbor-Joining method was done with MEGA6 to build a phylogenetic tree. Results: The target product of DNA amplification in this PCR was around 290-bp. The amplicon was visualized in 2% of agarose gel electrophoresis. The homology analysis of these sequences showed that it had more than 99% similarity. Conclusion: COX-1 gene sequences of S. scabiei from rabbits in Malang, Nganjuk, and Kediri were very similar to COX-1 gene sequences in S. scabiei acquired from several hosts according to NCBI data.
