Polymorphism of prolactin, growth differentiation factor 9, and calpastatin genes and their effects on weight traits in Awassi lambs

Objectives: This study was conducted to determine the correlation among prolactin gene (PRG), growth differentiation factor 9 (GDF-9), and calpastatin (CAG) genes polymorphism with growth traits in Awassi lambs. Materials and Methods: Records of 779 Awassi lambsfrom 264 dams and 15 sires were used. The sex, type of birth (single versus twins), weight at birth, average daily gain (ADG), and the adjusted weight at 60 days of age were determined for each animal. Blood samples were collected from all lambs to determine PRG, GDF-9, and CAG polymorphism using polymerase chain reaction-Restriction fragment length polymorphism. Results: Three PRG genotypes (AA, AB, and BB) were determined with a frequency of 0.88, 0.05, and 0.07, respectively. The frequency of each allele was 0.84 and 0.16 for A and B alleles, respectively. For GDF-9, there were only two genotypes detected (AB and BB) with a frequency of 0.96 and 0.04, respectively. The frequency of each allele was 0.92 and 0.08 for A and B alleles, respectively. For CAG, two genotypes were also detected (AB and BB) with a frequency of 0.92 and 0.08, respectively. The frequency of each allele was 0.96 and 0.04 for A and B alleles, respectively. A significant (p ≤ 0.04) effect of PRG genotype on birth weight was detected but this effect was not significant on ADG and weight at weaning. There were no associations between any of the pre-weaning growth traits and GDF-9 and CAG variants. Conclusion: The results of this study show that PRG could be used to select dams with a high frequency of dystocia to reduce birth weight of newborn lambs and therefore conserve the dams reproductive functions and improve lamb survivability. Objective: The objective of this study was to assess the risk of rabies entry through the movement of hunting dog from Garut District to Sumatera Island with a semi-quantitative approach. Materials and Methods: Rabies entry assessment used the standard risk analysis according to the World Organization for Animal Health, with a semi-quantitative approach referring to Australian Biosecurity. Risk estimation calculation used Microsoft Excel and probabilities were estimated using Monte Carlo stochastic simulation modeling with @Risk (Palisade Corporation). Results: Risk estimation were considered as very low with a 0.02 (90%; 0.010.03) probability. The probability of undetected rabies-infected dog during Veterinary Certificate issuance [node probability (NP4)] was considered as the highest, with moderate likelihood and 0.63 (90%; 0.510.75) of probability value. The number of dog movement to Sumatera reached 27,000 heads per year which 5,050 heads of them come from Garut District. There were 2 of 100 dogs from Garut District entered to Sumatera possibly infected by rabies. The five highest parameters most determinant of the risk were dog vaccination before transported (0.66), dog obtained from other District (0.41), vaccination program (0.32), serologically test (0.27), and history of vaccination (0.23). Conclusion: Risk estimation from assessing on rabies entry to Sumatera through hunting dogs movement from Garut District was considered very low. Risk mitigation is focused on the highest parameters that contribute the most to risk based on the results of the sensitivity analysis. Semi-quantitative likelihood evaluations can consider the volume of dog traffic which is an important issue in risk analysis which is not easy to get with a simpler qualitative approach. Objective: Investigating the antibiotic and antioxidant benefits of medicinal herbs to enrich the serum immune responses of chicken meat.Materials and Methods: A total of 1,080 Ross 308 broilers were reared up to 42 days. The broilers were divided randomly into nine assemblies, with each sectioned into three replicates. The first and second were supplemented with 0.25% and 0.50% of coriander seeds, respectively, while the third and fourth with 0.25% and 0.50% of rosemary leaves, respectively. A mixture of herbs from the two plants were added to fifth, sixth, seventh, and eighth treatments [(0.50% coriander seeds + 0.50% rosemary leaves), (0.25% coriander seeds + 0.50% rosemary leaves), (0.50% coriander seeds + 0.25% rosemary leaves), and (0.25% coriander seeds + 0.25% rosemary leaves)], respectively, whereas chicks in the ninth as a control group.Results: The results showed the pH for the thigh and breast of the carcass were measured. Glycogen levels, serum immunity (H, L, Hlration, Albumin, Globulin, and A/G ratio at 28 days and alanine aminotransferase, aspartate aminotransferase, and cholesterol at 42 days). The mineral deposits in the chicken meat were measured for Mg, Fe, Ca, Na, J, and total N. The fifth treatment had a significantly higher glycogen ratio (p < 0.05). pH measurements for the thigh and breast were done immediately, 4, 12, and 24 h after slaughter. For the thigh, the seventh treatment was highest immediately and at 12 h. For the breast, significant differences were only noted at 12 h for chickens on a coriander diet.Conclusion: It is concluded that these additives have a positive effect on some of the blood pro-files, carcass characteristics, and mineral composition of chicken meat. OBJECTIVE To describe diffusion and perfusion characteristics of the prostate gland of healthy sexually intact adult dogs as determined by use of diffusion-weighted and perfusion-weighted MRI. ANIMALS 12 healthy sexually intact adult Beagles. PROCEDURES Ultrasonography of the prostate gland was performed. Subsequently, each dog was anesthetized, and morphological, diffusion-weighted, and perfusion-weighted MRI of the caudal aspect of the abdomen was performed. The apparent diffusion coefficient was calculated for the prostate gland parenchyma in diffusion-weighted MRI images in the central ventral and peripheral dorsal areas. Perfusion variables were examined in multiple regions of interest (ROIs) in the ventral and dorsal areas of the prostate gland and in the gluteal musculature. Signal intensity was determined, and a time-intensity curve was generated for each ROI. RESULTS Results of ultrasonographic examination of the prostate gland revealed no abnormalities for any dog. Median apparent diffusion coefficient of the prostate gland was 1.51 × 10-3 mm2/s (range, 1.04 × 10-3 mm2/s to 1.86 × 10-3 mm2/s). Perfusion-weighted MRI variables for the ROIs differed between the prostate gland parenchyma and gluteal musculature. CONCLUSIONS AND CLINICAL RELEVANCE Results provided baseline information about diffusion and perfusion characteristics of the prostate gland in healthy sexually intact adult dogs. Additional studies with dogs of various ages and breeds, with and without abnormalities of the prostate gland, will be necessary to validate these findings and investigate clinical applications.

The proposed advantages of intra-arterial chemotherapy (IAC) are based on the premises of local dose escalation to the tumor and reduced availability of systemic drugs. There is a lack of objective pharmacokinetic data to confirm the advantage of IAC in dogs with naturally occurring urogenital tumors. The objective of this study was to determine if IAC administration in urogenital tumors would result in decreased systemic drug exposure when compared to intravenous routes. Twenty-two dogs with naturally occurring urogenital tumors were enrolled in this prospective case-controlled study. Mitoxantrone, doxorubicin, or carboplatin were administered by IAC and intravenous routes [intravenous awake (intravenous chemotherapy - IVC) and under general anesthesia (IVGAC)] 3 weeks apart. Serum assays were used to determine the extent of systemic drug exposure. Dose-normalized peak systemic serum concentration (Cmax) and area under the serum drug concentration-time curve (AUC) were used to quantify systemic exposure. A total of 26 mitoxantrone treatments were administered to 10 dogs. While there was no significant difference in Cmax, the AUC was significantly lower after IAC compared with IVGAC. Ten doxorubicin treatments were administered to 5 dogs. There were no significant differences in Cmax or AUC. A total of 14 carboplatin treatments were administered to 7 dogs. The Cmax was significantly lower for IAC compared to IVC, while the AUC values were equivocal. This study demonstrates certain lower serum values may be achieved after IAC delivery of carboplatin and mitoxantrone. These chemotherapy agents may have a preferred pharmacological profile for regional chemotherapy delivery in dogs with urogenital tumors.

The objective of this study was to assess veterinarians' understanding of nutraceutical use in humans and companion animals and their motivation and circumstances for recommending nutraceuticals to clients. We administered a cross-sectional survey to veterinarians attending continuing education sessions at the University of Georgia (USA) College of Veterinary Medicine from 2012 to 2015 (N = 126). Information regarding veterinarians' age, year of graduation, practice focus, and typical approaches to nutraceutical use was compiled from the returned surveys. The results indicated that veterinarians are more familiar with nutraceutical use in animals than in humans and primarily recommend nutraceuticals to their clients for preventative purposes and/or due to client interest. Veterinarians believed that nutraceuticals were most useful for osteoarthritis and therefore use omega-3 fatty acid and glucosamine/chondroitin products more often than other products for both their patients and their own pets. Safety and efficacy were the most important considerations when deciding which nutraceuticals to recommend to clients. The survey results show that veterinarians are familiar with nutraceuticals and open to their use in patients when they perceive these products to be safe and efficacious.

OBJECTIVE To determine the impact of mechanical ventilation (MV) and perfusion conditions on the efficacy of pulse-delivered inhaled nitric oxide (PiNO) in anesthetized horses. ANIMALS 27 healthy adult horses. PROCEDURES Anesthetized horses were allocated into 4 groups: spontaneous breathing (SB) with low (< 70 mm Hg) mean arterial blood pressure (MAP; group SB-L; n = 7), SB with physiologically normal (≥ 70 mm Hg) MAP (group SB-N; 8), MV with low MAP (group MV-L; 6), and MV with physiologically normal MAP (group MV-N; 6). Dobutamine was used to maintain MAP > 70 mm Hg. Data were collected after a 60-minute equilibration period and at 15 and 30 minutes during PiNO administration. Variables included Pao2, arterial oxygen saturation and content, oxygen delivery, and physiologic dead space-to-tidal volume ratio. Data were analyzed with Shapiro-Wilk, Mann-Whitney U, and Friedman ANOVA tests. RESULTS Pao2, arterial oxygen saturation, arterial oxygen content, and oxygen delivery increased significantly with PiNO in the SB-L, SB-N, and MV-N groups; were significantly lower in group MV-L than in group MV-N; and were lower in MV-N than in both SB groups during PiNO. Physiologic dead space-to-tidal volume ratio was highest in the MV-L group. CONCLUSIONS AND CLINICAL RELEVANCE Pulmonary perfusion impacted PiNO efficacy during MV but not during SB. Use of PiNO failed to increase oxygenation in the MV-L group, likely because of profound ventilation-perfusion mismatching. During SB, PiNO improved oxygenation irrespective of the magnitude of blood flow, but hypoventilation and hypercarbia persisted. Use of PiNO was most effective in horses with adequate perfusion.

OBJECTIVE To assess recording accuracy of right atrial and ventricular depolarization during 12-lead ECG when precordial lead V1 was positioned at each of 5 locations on the thorax of dogs with various thoracic conformations. ANIMALS 60 healthy client-owned dogs. PROCEDURES 20 dogs were allocated to each of 3 groups (brachymorphic, mesomorphic, or dolichomorphic) on the basis of thoracic conformation. Each dog remained unsedated and was positioned in right lateral recumbency for a series of five 12-lead surface ECGs, with V1 located adjacent to the sternum in the fifth intercostal space (ICS; control), at the costochondral junction (CCJ) of the right first ICS (1st-R), at the CCJ of the right third ICS, at the right third ICS where the thorax was the widest, and at the CCJ of the left first ICS. Electrocardiographic variables were compared among the 5 ECG tracings. RESULTS When V1 was at the control location, the P wave was positive for all dogs; however, consistent recording of right atrial and ventricular depolarization (ie, R wave-to-S wave ratio [R/S] < 1) occurred more frequently for brachymorphic dogs (16/20) than for dolichomorphic (7/20) and mesomorphic (6/20) dogs. When V1 was at the 1st-R location, the P wave was negative for most dogs, and R/S was < 1 for the majority of dogs in the brachymorphic (19/20), mesomorphic (17/20), and dolichomorphic (16/20) groups. The median R/S for V1 at the 1st-R location was significantly lower than that for the other 4 V1 locations. CONCLUSIONS AND CLINICAL RELEVANCE Results indicated that placement of V1 at the 1st-R location provided correct evaluation of right atrial and ventricular depolarization in most dogs regardless of thoracic conformation.

OBJECTIVE To evaluate the effects of injectable dexmedetomidine-ketamine-midazolam (DKM) and isoflurane inhalation (ISO) anesthetic protocols on selected ocular variables in captive black-tailed prairie dogs (Cynomys ludovicianus; BTPDs). ANIMALS 9 zoo-kept BTPDs. PROCEDURES The BTPDs received dexmedetomidine hydrochloride (0.25 mg/kg, IM), ketamine hydrochloride (40 mg/kg, IM), and midazolam hydrochloride (1.5 mg/kg, IM) or inhalation of isoflurane and oxygen in a randomized complete crossover design (2-day interval between anesthetic episodes). Pupil size, globe position, tear production, and intraocular pressure measurements were recorded at 5, 30, and 45 minutes after induction of anesthesia. For each BTPD, a phenol red thread test was performed in one randomly selected eye and a modified Schirmer tear test I was performed in the other eye. Intraocular pressure was measured by rebound tonometry. RESULTS Compared with findings for the DKM protocol, pupil size was smaller at all time points when the BTPDs underwent the ISO protocol. Globe position remained central during anesthesia with the DKM protocol, whereas it varied among central, ventromedial, and ventrolateral positions during anesthesia with the ISO protocol. Tear production and intraocular pressure decreased significantly over time when the BTPDs underwent either protocol. CONCLUSIONS AND CLINICAL RELEVANCE Results indicated that ophthalmic examination findings for anesthetized BTPDs can be influenced by the anesthetic protocol used. The DKM protocol may result in more consistent pupil size and globe position, compared with that achieved by use of the ISO protocol. Tear production and intraocular pressure measurements should be conducted promptly after induction of anesthesia to avoid the effect of anesthetic episode duration on these variables.

OBJECTIVE To evaluate erythema and number of CFUs on the skin of dogs with hair clipped by use of 2 sizes of clipper blades. ANIMALS 67 client-owned dogs receiving an epidural. PROCEDURES Hair was clipped with a No. 10 blade (approx hair length, 1.5 mm) on one half and a No. 40 blade (approx hair length, 0.25 mm) on the other half of each epidural site. Skin was surgically scrubbed with 2% chlorhexidine gluconate and 70% isopropyl alcohol. Samples were obtained immediately after clipping, after skin was scrubbed, and again 24 hours after clipping. Number of CFUs for both sides of the clipped areas, types of microorganisms, and growth on MacConkey agar were evaluated every 24 hours for 72 hours. Colonies were evaluated for bacterial morphology and Gram stain characteristics. Sites were evaluated 24 hours after clipping for evidence of erythema. RESULTS 24 hours after hair was clipped, there was a significantly higher incidence of erythema and higher number of Micrococcaceae bacteria for the side clipped with the No. 40 blade than the side clipped with the No. 10 blade. Number of CFUs did not differ significantly between size of clipper blades. CONCLUSIONS AND CLINICAL RELEVANCE Clipping hair with a No. 40 blade resulted in a significant increase in the incidence of erythema and higher number of Micrococcaceae bacteria, compared with results for clipping with a No. 10 blade. These results supported use of a No. 10 clipper blade to prevent erythema and reduce variation in the skin microbiome.
