A infestação por carrapatos em bovinos gera grandes problemas, sendo esse parasito responsável por elevados prejuízos econômicos em diversos rebanhos de produção de corte e também de leite na maioria das regiões brasileiras, inclusive em Santa Catarina. A resistência dos carrapatos frente aos diversos princípios ativos pode ser decorrente de fatores como o uso indiscriminado de carrapaticidas, bem como a inadequada forma de aplicação dos mesmos. Tendo em vista a grande importância em combater essas infestações, buscou-se avaliar a eficiência de oito diferentes princípios ativos de contato contra o carrapato Rhipicephalus microplus, bem como identificar os principais fatores que possam contribuir com a seleção de carrapatos resistentes nas propriedades avaliadas. Para isso, teleóginas de R. microplus foram coletadas em 39 propriedades de diferentes municípios do Oeste de Santa Catarina. No momento da coleta também foram obtidas informações referentes ao sistema de manejo dos animais, histórico dos carrapaticidas utilizados, número de aplicações anuais, frequência de rodízios de acaricidas, e frequência de acompanhamento técnico para controle estratégico, com a finalidade de evidenciar os diferentes fatores que possam estar contribuindo para o surgimento de eventual resistência das populações do R. microplus. A avaliação de suscetibilidade foi realizada através da técnica de biocarrapaticidograma, utilizando 10 teleóginas ingurgitadas para cada princípio e mais o controle. Dentre os principais resultados obtidos, a maioria das associações entre piretróides e organofosforados demonstraram ter eficiência entre 96,6% e 100%, com exceção da associação de Alfacipermetrina 15%, Ethion 16% e Clorpirifós 8,5% com 93,4% de eficiência. Dentre as bases químicas sem nenhum tipo de associação o Amitraz 12,5% demonstrou ser eficaz em 98,3%, em contrapartida a Cipermetrina 15% teve eficiência em apenas 25,8% das propriedades analisadas. A partir dos resultados obtidos, pode se afirmar que as formas de controle e manejo adotados pelos produtores podem estar relacionadas com os baixos índices de resistência aos carrapaticidas de contato, nas diferentes populações de carrapato na região do estudo. | Tick infestation causes major problems in cattle. The results of our study show that most associations between pyrethroids and organophosphates had an efficiency between 96.6% and 100% in the control of R. microplus except for the combination of Alfacipermetrine 15%, Ethion 16%, and Chlorpyrifos 8.5% that had 93.4% of efficiency. Among the chemicals not associated with any other type of compound, Amitraz 12.5% was effective in 98.3% of the cases. In contrast, Cypermethrin 15% had an efficiency in only 25.8% of the farms/herds analyzed. Based on the results of the present study, we may infer that the methods of control and management used by producers may be related to the low indexes of resistance to topical acaricides in the different populations of ticks in the study area.

Este estudo relata os fatores que levaram um Residencial localizado em Bragança Paulista (SP, Brasil), área não-endêmica para Febre Maculosa Brasileira (FMB), a ser classificado como Área de Risco para a doença, mostrando que uma crescente população residente de capivaras (Hydrochoerus hydrochaeris) na área era a provável responsável por uma proliferação de carrapatos Amblyomma sculptum e estava atuando como hospedeiro amplificador da bactéria Rickettsia rickettsii, principal agente etiológico da FMB. Relatamos as ações de manejo ambiental propostas para controlar a quantidade de carrapatos no local e reduzir o risco de transmissão da doença, incluindo medidas para o controle de populações de carrapatos parasíticas e no ambiente e a eliminação da população residente de capivaras. Análises de dados populacionais de carrapatos e testes serológicos para R. rickettsii indicaram que as medidas tomadas foram efetivas, causando grande redução da população de carrapatos no ambiente e reduzindo o risco de transmissão de FMB na área. | This study reports the factors which led a gated community located in Bragança Paulista (SP, Brazil), a non-endemic area for Brazilian Spotted Fever (BSF), to be classified as a Risk Area for transmission of this disease, showing that an increasing resident population of capybaras (Hydrochoerus hydrochaeris) in the area was likely responsible for a proliferation of Amblyomma sculptum ticks and acted as an amplifying host for Rickettsia rickettsii, the main etiologic agent of BSF. O presente trabalho analisou a fauna ectoparasitária de peixes ornamentais de água doce comercializados em um estabelecimento atacadista de peixes ornamentais na grande São Paulo no período de janeiro a dezembro de 2016. Foram avaliados 268 peixes ornamentais de água doce de diversas espécies que apresentaram sinais de doença após a chegada ao estabelecimento. Nos indivíduos foi efetuada a pesquisa de ectoparasitas por meio da raspagem de pele e brânquias e visualização em microscópio óptico em aumento 40x, 100x, 200x e 400x. Foram encontrados parasitos em 220 animais (82%) incluindo 11 parasitos distintos: Ichthyophthirius multifiliis, Monogêneas, Metacercárias (Centrocestrus formousus), Epistylis spp., Chillodonella spp., Lernaea spp., Tetrahymena spp., Piscinoodinium spp., Trichodina spp., Ichthyobodo spp. e Argulus spp. A ocorrência dos parasitas no estabelecimento em estudo pode ser indicativa da falta de boas práticas sanitárias e de higiene dos fornecedores no manejo de suas instalações e, consequentemente, na distribuição de animais infestados, o que coloca em risco toda cadeia. | This study analyzed the ectoparasitic fauna of freshwater ornamental fish marketed by a wholesaler in the São Paulo city. O objetivo do presente estudo foi determinar a frequência de anticorpos anti-Toxoplasma gondii em soro de galinhas abatidas na região do Triângulo Mineiro, Minas Gerais, detectar molecularmente o parasito em tecidos (coração e cérebro) de algumas das aves sorologicamente positivas e averiguar variáveis de risco associadas à infecção. Foram testados soros de 417 galinhas, criadas nos sistemas extensivo, semi-intensivo e intensivo. Para a pesquisa de anticorpos anti-T. gondii foi utilizada a Reação de Imunofluorescência Indireta (RIFI) e Hemaglutinação Indireta (HAI). A Reação em Cadeia da Polimerase (PCR) foi utilizada para detectar o DNA de T. gondii em fragmentos de cérebro e coração. Anticorpos foram detectados no soro de 37,65% (157/417) das aves pela RIFI e em 75,06% (313/417) pela HAI. O índice Kappa mostrou uma fraca concordância entre as técnicas (0,087). Baseado na RIFI, foi verificada associação estatisticamente significativa (p < 0,0001) entre a soropositividade e as variáveis: idade, tipo de alimentação e criação em conjunto com outras espécies animais. Não foi observada associação estatística (p > 0,01) entre as variáveis: sexo, presença de gatos e presença de ratos. Pelo diagnóstico molecular DNA do parasito foi detectado em duas amostras de cérebro, de indivíduos diferentes criados em sistema intensivo e semi-intensivo. Na toxicologia da reprodução e do desenvolvimento, a atenção necessária foi dada aos efeitos da exposição do organismo às inúmeras drogas durante o período gestacional somente após a metade dos anos 50, quando a tragédia da talidomida atingiu a humanidade. Assim, esta área alcançou desenvolvimento científico com a contribuição da experimentação animal nas últimas décadas. O uso de animais de laboratórios para a pesquisa científica expôs a necessidade de melhorar a qualidade de vida destas espécies. Portanto, este trabalho tem como objetivo investigar como a ciência do bem-estar animal pode melhorar a pesquisa científica como um todo, incluindo na área de toxicologia da reprodução e de desenvolvimento, enfatizando o enriquecimento ambiental em biotérios. Uma revisão integrativa de literatura foi realizada, incluindo abordagens quantitativas e qualitativas, quais podem ser aplicadas para estudos de toxicologia. Aqui, são mostradas evidências de que o enriquecimento ambiental melhora o bem-estar animal e previne ou reduz os efeitos negativos do cativeiro, qual deve um princípio da pesquisa toxicológica por razões éticas, argumentos legais e garantias científicas. | Reproductive and developmental toxicology has focused on the need to approach the effects of organism exposure to various drugs during pregnancy after the mid-50’s, when the thalidomide tragedy stroke humanity. In recent decades, this area of study has developed a lot due to animal testing, raising awareness on the need to improve the quality of life of such animals. Therefore, this paper aims to investigate how the science of animal welfare can improve scientific research as a whole, including the reproductive and developmental toxicology fields, by emphasizing environmental enrichment in animal facilities. To do so, we conducted an integrative literature review on several quantitative and qualitative methodological approaches that are applicable to toxicology studies. Here, we present evidence that environmental enrichment improves animal welfare and prevents or reduces the negative effects of captive housing, which must be a principle of toxicological research for ethical, legal and scientific reasons.

In recent decades, this area of study has developed a lot due to animal testing, raising awareness on the need to improve the quality of life of such animals. Therefore, this paper aims to investigate how the science of animal welfare can improve scientific research as a whole, including the reproductive and developmental toxicology fields, by emphasizing environmental enrichment in animal facilities. To do so, we conducted an integrative literature review on several quantitative and qualitative methodological approaches that are applicable to toxicology studies. Here, we present evidence that environmental enrichment improves animal welfare and prevents or reduces the negative effects of captive housing, which must be a principle of toxicological research for ethical, legal and scientific reasons.

As neuropatias do glossofaríngeo e vago são frequentemente associadas com enfermidade das bolsas guturais. A ocorrência desta lesão secundária a compressão pelo músculo estilofaríngeo não é reportada. Relata-se o caso de uma égua quarto de milha, 8 anos de idade, 450 kg, apresentando dispneia, ruído respiratório e perda de peso. No exame clínico observou-se dispnéia mista, taquicardia, disfagia, sialorréia, crepitação pulmonar e linfadenopatia submandibular e parotídea. Ao exame endoscópico identificou-se condrite aritenóide direita, colapso nasofaríngeo, edema generalizado da laringe e deslocamento dorsal do palato mole. Na avaliação da bolsa gutural direita identificou-se aumento de volume na origem do músculo estilofaríngeo com consequente compressão dos nervos cranianos XII, X e IX. Foi realizada traqueotomia, tratamento sistêmico com corticosteróides e antibióticos. Nenhuma resolução foi observada, após 16 dias o animal apresentou piora clínica, pleuropneumonia, uveíte, sepse grave, insuficiência renal aguda e foi submetido à eutanásia. A neuropatia mista resultou em rápida deterioração clínica do animal, devido à dificuldade de deglutição e processos respiratórios associados. Salienta-se a importância de se avaliar a origem do músculo estilofaríngeo em casos de colapso nasofaríngeo associado a disfagia em cavalos, dada a possibilidade de que alterações estruturais nesse músculo possam resultar em neuropatia laríngea. | Neuropathies of pharyngeal branches of glossopharyngeal and vagus are often associated with guttural pouches diseases; however, these branches of injury due to stylopharyngeus muscle compression are not reported. The mixed neuropathy resulted in rapid clinical deterioration of the animal, due to the difficulty in swallowing and consequent associated respiratory processes. This report emphasizes the importance of evaluating stylopharyngeus muscle origin in cases of nasopharyngeal collapse associated with dysphagia in horses, given the possibility that structural changes in this muscle can result in laryngeal neuropathy.

A crescente casuística de cães obesos na clínica de cães tem sido um alerta aos médicos veterinários sobre a necessidade do desenvolvimento de novas ferramentas para a abordagem do paciente obeso, com o intuito de alcançar resultados satisfatórios em programas de redução de peso. Nesse estudo, foram selecionados 18 cães obesos ou acima do peso ideal, sem indícios de distúrbios endócrinos. Esses animais foram divididos aleatoriamente em dois grupos de nove animais. Em um dos grupos foram utilizadas ferramentas estratégicas para modificação comportamental e ambiental para seus tutores e esse grupo foi assistido diariamente por comunicação em aplicativo digital de mensagens instantâneas (WhatsApp(R)). No outro grupo, essas ferramentas não foram utilizadas e os animais foram acompanhados somente na consulta mensal. A variável analisada foi o percentual de perda de peso de cada animal em um mês de aplicação dométodo. Os animais do grupo assistido diariamente apresentaram média de perda de peso de 5,17%, enquanto que os animais do grupo não assistido apresentaram média de 3,31%. Pelos resultados é possível afirmar que os animais do grupo assistido perderam aproximadamente 1,6 vezes a mais de peso que os animais não assistidos. Embora os resultados sejam motivadores, não houve diferença estatística com grau de significância de 0,05, isso provavelmente se deve ao número amostral de dezoito animais ser muito pequeno. | The increasing number of obese dogs in veterinary practice has been a warning to veterinarians about the need to develop new tools to approach the obese patient, aiming to attain satisfactory results in weight loss programs. A carne de aves é um alimento que frequentemente encontra-se envolvido como veículo de micro-organismos causadores de enfermidades, sendo Salmonella sp. um desses agentes comumente envolvidos nessas enfermidades em todo o mundo. Com isso, o objetivo deste estudo foi determinar a ocorrência de Salmonella sp. em carcaças de frango congeladas e refrigeradas, abatidas e comercializadas na região oeste do estado do Paraná. Um total de 340 amostras foram coletadas entre janeiro de 2007 e abril de 2013. Das amostras analisadas, 66 (19,41%) das carcaças foram positivas para Salmonella sp. De acordo com os resultados obtidos pode-se concluir que a ocorrência de Salmonella sp. em carcaças de frangos permanece alta, mesmo sendo produtos comercializados refrigerados ou congelados, métodos consideradosadequados para conservação de alimentos. Um resultado de menor ocorrência era esperado devido à implementação de 14 anos do Programa Nacional de Redução de Patógenos estabelecido pelo Ministério da Agricultura, Pecuária e Abastecimento visando uma redução gradual na ocorrência de Salmonella sp. nestes produtos através de monitoramento constante de carcaças imediatamente após o abate. | Poultry meat is often involved as a vehicle for microorganisms that cause food-borne diseases. Os organismos podem desenvolver diferentes tipos de assimetria quando ocorrem desvios da pefeita simetria esperada. Entre os diversos tipos de assimetria existentes duas merecem especial destaque: a flutuante (AF) e a directional (AD). A AF é representada por pequenas diferenças casuais entre as partes correspondents das laterais direita e esquerda de um indivíduo em estruturas pareadas bilateralmente; acredita-se que elas reflitam a habilidade de um organismo adaptar-se a fatores estressantes genéticos e ambientais observados durante o seu crescimentos. A AD ocorre quando um lado da assimetria plana desenvolve-se mais do que o outro e há um componente genético. No presente trabalho foi analisada a expresssão da simetria morfológica de 38 crânios de diferentes grupos etários de javalis (Sus scrofa), nos seus aspectos ventrais, com o emprego de duas coordenadas dimensionais de 27 pontos anatômicos homólogos. As análises efetuadas revelaram a existência de valores significantes de AF e AD em toda a amostra trabalhada, detectando inclusive diferenças entre os grupos etários. Herpesvirus bovino 5 é um alfaherpesvírus causador de meningoencefalite não supurativa em bovinos. Esta doença possui ocorrência natural em surtos ou casos isolados, associadas a baixa morbidade e alta letalidade. Embora estudos anteriores tenham elucidado aspectos relacionados a patogenia da doença, há uma lacuna de conhecimento relacionado aos eventos moleculares que contribuem para a infecção e replicação do BoHV-5. O objetivo do presente estudo foi determinar a expressão gênica in vitro de genes virais (i.e., alfa, beta e gama) e das células hospedeiras (GAPDH e 18S) durante a infecção considerando diferentes momentos de infecção e quantidade de vírus utilizado. Três genes do BoHV-5 (bICP0, UL9, US4), um gene estrutural (GAPDH) e um gene constitutivo (18S) da célula bovina tiveram suas expressões avaliadas por PCR quantitativa (qPCR). Enquanto os genes virais tiveram sua expressão aumentada ao longo do tempo de infecção, o gene hospedeiro teve sua expressão diminuída, demonstrando a ação do vírus na expressão gênica de células bovinas in vitro. O gene constitutivo 18S teve sua expressão mantida durante todos os momentos do experimento. Nossos resultados claramente demonstraram que o GAPDH não deve ser usado como gene de referência em estudos com infecção por BoHV-5 pois é influenciado pela infecção viral. Entretanto, o 18S rRNA foi constitutivamente expresso e pode ser recomendado para normalização em células bovinas infectadas pelo vírus. A expressão de mRNA viral não foi alterada pela quantidade de vírus usada. Todos os genes virais demonstraram o mesmo padrão de expressão ao longo do tempo de infecção. Nossos resultados trazem importantes diferenças comparando aos estudos clássicos que avaliaram a expressão de genes alfa, beta e gama. Mais estudos são necessários para aumentar o conhecimento da biologia molecular do BoHV-5. Estudo utilizando sequenciamento de última geração (i.e., RNA-seq), usando modelos in vitro e in vivo, aparentam ser o próximo passo lógico para acessar as alterações do transcriptoma do hospedeiro e viral ao longo do curso da infecção. | Bovine herpesvirus 5 is an alphaherpesvirus that causes nonsuppurative meningoencephalitis in cattle. This disease occurs naturally in either outbreaks or isolated cases, and exhibits low morbidity and high lethality. Although previous studies elucidated crucial aspects involved in the pathogenesis of the disease, there is a paucity of information regarding the molecular events contributing to infection and replication of BoHV-5. The objective of the present study was to determine the in vitro gene expression pattern of BoHV-5 (e.g., alpha, beta, and gamma genes) and host cells genes (GAPDH and 18S) over time utilizing different quantities of inoculated virus. Three BoHV-5 genes (bICP0, UL9, US4) and one structural bovine cell gene had their expression accessed by real-time PCR. While the expression of BoHV-5 genes increased during the course of infection, GAPDH gene expression decreased in the host cells, evidencing the effect of viral infection on the expression of bovine cell genes. The 18S ribosomal RNA (rRNA) gene was constitutively expressed throughout BoHV-5 infection. Our data clearly demonstrates that GAPDH gene should not be used as a reference gene in studies of BoHV-5 infection because it was influenced by viral infection. However, 18S rRNA was constitutively expressed and, therefore, is recommended for normalization of BoHV-5 infection studies in bovine cells. The expression of viral genes transcripts was not altered by increasing number of viral particles added to the culture. All viral genes included here demonstrated the same expression pattern over time and there was no difference in the expression of viral genes among the various time points. Our data show important differences comparing to classical studies regarding herpesvirus alpha, beta, and gamma genes expression. More research is necessary to improve our understanding about the BoHV-5 biology during infection. Studies employing next-generation sequencing (i.e., RNA-seq), using both in vitro and in vivo models, would be the next logical step to grasp the virus and host cell’s transcriptome changes over the course of infection.

Bovine herpesvirus 5 is an alphaherpesvirus that causes nonsuppurative meningoencephalitis in cattle. This disease occurs naturally in either outbreaks or isolated cases, and exhibits low morbidity and high lethality. Although previous studies elucidated crucial aspects involved in the pathogenesis of the disease, there is a paucity of information regarding the molecular events contributing to infection and replication of BoHV-5. The objective of the present study was to determine the in vitro gene expression pattern of BoHV-5 (e.g., alpha, beta, and gamma genes) and host cells genes (GAPDH and 18S) over time utilizing different quantities of inoculated virus. Three BoHV-5 genes (bICP0, UL9, US4) and one structural bovine cell gene had their expression accessed by real-time PCR. While the expression of BoHV-5 genes increased during the course of infection, GAPDH gene expression decreased in the host cells, evidencing the effect of viral infection on the expression of bovine cell genes. The 18S ribosomal RNA (rRNA) gene was constitutively expressed throughout BoHV-5 infection. Our data clearly demonstrates that GAPDH gene should not be used as a reference gene in studies of BoHV-5 infection because it was influenced by viral infection. However, 18S rRNA was constitutively expressed and, therefore, is recommended for normalization of BoHV-5 infection studies in bovine cells. The expression of viral genes transcripts was not altered by increasing number of viral particles added to the culture. All viral genes included here demonstrated the same expression pattern over time and there was no difference in the expression of viral genes among the various time points. Our data show important differences comparing to classical studies regarding herpesvirus alpha, beta, and gamma genes expression. More research is necessary to improve our understanding about the BoHV-5 biology during infection. Studies employing next-generation sequencing (i.e., RNA-seq), using both in vitro and in vivo models, would be the next logical step to grasp the virus and host cell’s transcriptome changes over the course of infection.
