Acacia angustissima intoxication of Menz lambs requires two components
2005
Brown, D.L. | Yimegnuhal, A. | McCrabb, G.J.
This study sought to determine if sheep suffer neurological symptoms when fed Acacia angustissima leaves, and whether an equivalent amount of 70% acetone extract would have the same effect. In addition, the study tried to determine if treatment of leaves with 70% acetone would destroy the activity of A. angustissima toxins, and whether extraction with 70% aqueous acetone extract would separate two components of a toxic system. Twenty-five Menz lambs were randomly assigned to one of five treatments (1) A angustissima leaves as half the diet, 2) dried extract (70% aqueous acetone) of the same quantity of leaves, 3) a corresponding amount of residues, 4) a recombination of the dried extract and dried residue, or (5) a control diet containing no A angustissima leaves or extract fractions. All animals fed the leaves and the recombined fractions died or were euthanized when they were observed to be dying of severe neurological derangement. None of the other animals showed any neurological signs of impairement. The results of this study indicate that healthy, well-fed sheep can be poisoned by A angustissima, that the toxins are not destroyed by acetone or oven drying, and that severe neurological intoxication requires two components, which can be separated by acetone extraction.

2005
Detilleux, Johann | Serteyn, Didier
peer reviewed | Techniques of cost-effectiveness analyses were applied to determine whether or not it is economically efficient to measure the packed cell volume (PCV) on a colic horse before deciding on abdominal surgery. The effects of this decision of uncertainty on the estimated values of the parameters (probability of survival after surgery, surgery costs, PCV positive predictive value, and length of survival after surgery) were considered along with the monetary values of collecting additional information on those parameters. The effects of uncertainty on the incremental net benefits of each alternative were depicted by tornado diagrams, cost-effectiveness acceptability curves, and posterior probability distributions. The worth of additional information was computed as the expected values of perfect and sampling information. Given previously published results, the best PCV cut-off point to distinguish between survivors and nonsurvivors was at 44%. At this threshold, the most economically effective alternative is to measure PCV before surgery providing the owner is willing to pay less than €672 for each year the horse survives. Uncertainty on probability of survival after surgery largely influenced the decision whether or not to measure the PCV, but one should spend at most €381 in research to reduce this uncertainty. A study of postoperative survival of 500 colic horses would ensure an expected gain of €370 associated with a reduction in uncertainty.

The present study was carried out to investigate the skin irritation potential of oregano oil in rabbits. A volume of 0.5 ml of test article was applied to intact and abraded skins, respectively, for 24 h in 6 healthy male New Zealand White rabbits. Parameters measured during 72 h observation period were mortality, clinical signs, body weight changes, and local irritation. Treatment-related toxic symptoms, as evidenced by anorexia and decreased locomotor activity, were observed in all rabbits tested. Two rabbits out of the 6 total died on day 2 after the application of test article due to treatment-related toxicity.

Resistance to antibiotics is a problem all over the world, and this problem also is so extended in veterinary fields. Therefore, this study aimed to find out the antibacterial substances from natural medicinal herbs against bovine mastitis pathogens, especially methicillin-resistant Staphylococcus aureus (MRSA). Among seventy two medicinal herbs, Terminaliae chebula extract showed antimicrobial effect against MRSA isolated from mastitis milk in cow and patient in human. However, Terminaliae chebula extract didn't show antibacterial effects on various strains including other contagious and environmental pathogens related with mastitis of dairy cows.

The present study was carried out to produce the hematological and blood chemical findings after hypophysectomy in rats. Hypophysectomy was performed by the parapharyngeal method and the sham surgery was performed for the control group. Two weeks after the operation, the body weight of the hypophysectomized and control rats was measured daily for 5 days. We deleted the rats the weight gain of which is less than 5 g during 5 days from the hypophysectomy group. The successful operation rate was approximately 40%. In the hypophysectomized and control rats, their blood samples were collected from posterior vena cava after celiotomy under generally anesthesia with ether.

Proteomics is a useful approach to know protein expression, post-translational modification and protein function. We investigated the protein expression pattern and identity in chickens fed with the tissue culture medium waste after harvest of Korean wild ginseng (TCM-KWG) (Panax ginseng). Two groups (n=60/group) of day old broiler chickens were administered with 0 (control) and 0.8% (treatment) TCM-KWG through drinking water. After 5 weeks, we examined the protein expression pattern of fibularis longus and superficial pectoral muscle by Two-dimensional electrophoresis analysis. Interestingly, TCM-KWG treatment significantly increased five spot's density, and markedly reduced five spot's density in the muscles.

This study was carried out to investigate the antibiotic resistance pattern of Enterococcus spp. and Staphylococcus aureus (S. aureus) isolated from chicken feces. All isolates showed high resistance to erythromycin (E) and tetracycline (TE). Of the 63 Enterococcus faecalis (E. faecalis) isolates, 73.0% were resistant to E and 98.4% to TE. Of the 44 Enterococcus faecium (E. faecium) isolates, 50.0% were resistant to E and 95.5% to TE. Of the 52 S. aureus isolates, 57.6% were resistant to E and 96.2% to TE. The prevalence of two and three drugs resistance pattern were 28.6% and 17.5% of E. faecalis, 40.9% and 25.0% of E. faecium and 38.5% and 23.1% of S. aureus, respectively.

We developed the one-step strip based on an immunochromatographic (IC) assay for the rapid detection of Listeria spp. Genus-specific monoclonal antibody to flagella of L. monocytogenes was conjugated with 40 nm colloidal gold particles which were prepared in our laboratory. The specificity of the IC strip was tested with pure cultured bacteria. All strains of the genus of Listeria spp. yielded positive reactions and 12 strains of non-Listeria were negative, resulting in a specificity of 100%. L. monocytogenes was artificially inoculated in raw pork macerated with listeria enrichment broth.

Epidemiological characteristics of a total of 48 swine herd with diarrhea or a history of diarrhea in Gyeongsang-do between 1999 and 2000 were performed to evaluate the prevalence of Salmonella spp., their serotypes and antibiotic resistance patterns with respect to the different stages of swine production system. A total of 139 Salmonella spp. (21%) were isolated from 662 fecal samples and the overall herd prevalence of Salmonella spp. ranged from 12.5% to 88%. The average prevalence of Salmonella spp. from swine stages of suckling/nursery, grow/finisher and sow stage were 25.7%, 19.2% and 18.4%, respectively.

Kim, T.J. (Chonnam National University, Gwangju, Republic of Korea) | Lee, B.J. (Chonnam National University, Gwangju, Republic of Korea) | Lee, J.I. (Chonnam National University, Gwangju, Republic of Korea), E-mail: jaeil@chonnam.ac.kr
Actinobacillus pleuropneumoniae is the causative agent of a porcine contagious pleuropneumonia. Among several virulence factors including exotoxin (Apx toxins), LPS, transferrin-binding proteins, OMPs, and some proteases, Apx toxins have been major targets for the protection study. In this study, cloning and expression of A. pleuropneumoniae Apx I and Apx Ⅱ toxin, which are produced by all highly virulent strains, were performed by Escherichia coli expression system. Genes coding Apx I and Ⅱ toxin were amplified from the A. pleuropneumoniae serotype 5 genomic DNA using polymerase chain reaction and cloned to a prokaryotic expression vector, pRSET.
