Gut microbiota of aquatic organisms: A key endpoint for ecotoxicological studies

Gut microbial communities constitute a compartment of crucial importance in regulation of homeostasis of multiple host physiological functions as well as in resistance towards environmental pollutants. Many chemical contaminants were shown to constitute a major threat for gut bacteria. Changes in gut microbiome could lead to alteration of host health. The access to high-throughput sequencing platforms permitted a great expansion of this discipline in human health while data from ecotoxicological studies are scarce and particularly those related to aquatic pollution. The main purpose of this review is to summarize recent body of literature providing data obtained from microbial community surveys using high-throughput 16S rRNA sequencing technology applied to aquatic ecotoxicity. Effects of pesticides, PCBs, PBDEs, heavy metals, nanoparticles, PPCPs, microplastics and endocrine disruptors on gut microbial communities are presented and discussed. We pointed out difficulties and limits provided by actual methodologies. We also proposed ways to improve understanding of links between changes in gut bacterial communities and host fitness loss, along with further applications for this emerging discipline.

Size-resolved samples were collected using a 14-stage impactor during four seasons in Zhengzhou and analyzed for 26 elements to calculate the health risks from atmospheric particle-bound metals. High concentrations of heavy metals were observed in ultrafine (10.2 (Ni)–66.9 (Cd) ng m⁻³) or submicrometer (11.4 (Ni)–134 (Pb) ng m⁻³) mode in winter. Two size-dependent models were used to estimate the deposition of inhaled toxic metals in various regions of the human respiratory system. Results show that heavy metals deposited in the alveolar region ranged from 7.6 (As)–375 (Al) ng m⁻³ and were almost concentrated in ultrafine and fine modes. Cd (2.2–8.6) may cause accumulative non-carcinogenic health effects on children, and Cr (1.0 × 10⁻⁴–2.2 × 10⁻⁴) may lead to carcinogenic health risks for nearby residents around the sampling site. The major sources by principal component analysis that contributed to Cr and Cd in ultrafine and fine particles were coal combustion, vehicular and industrial emissions. The atmospheric dry deposition fluxes of Cr and Cd were between 0.7 and 1.9 μg m⁻² day⁻¹ calculated by a multi-step method. From the environmental and public health perspective, environmental agencies must control the emission of heavy metals in the atmosphere.

In present study, the analyses of essential [copper (Cu), cobalt (Co), selenium (Se) and zinc (Zn)] and non-essential elements [mercury (Hg), lead (Pb), cadmium (Cd) and arsenic (As)] in 7 fish species consumed by the indigenous people of the European Russia Arctic were conducted. The Nenets Autonomous Region, which is located in the north-eastern part of European Russia, was chosen as a Region of interest. Within it, the Nenets indigenous group (n = 6000) constitutes approximately 10% of the total population. Nearly all of the Nenets live a traditional life with fish caught in the local waters as a subsistence resource.We found that northern pike contained twice the amount of Hg compared with roach, and 3–4 times more than other fish species commonly consumed in the Russian Arctic (namely, Arctic char, pink salmon, navaga, humpback whitefish and inconnu). Fish Hg concentrations were relatively low, but comparable to those reported in other investigations that illustrate a decreasing south-to-north trend in fish Hg concentrations. In the current study, northern pike is the only species for which Hg bioaccumulated significantly. In all fish species, both Cd and Pb were present in considerably lower concentrations than Hg. The total As concentrations observed are similar to those previously published, and it is assumed to be present primarily in non-toxic organic forms. All fish tissues were rich in the essential elements Se, Cu and Zn and, dependent on the amount fish consumed, may contribute significantly to the nutritional intake by indigenous Arctic peoples. We observed large significant differences in the molar Se/Hg ratios, which ranged from 2.3 for northern pike to 71.1 for pink salmon. Values of the latter <1 may increase the toxic potential of Hg, while those >1 appear to enhance the protection against Hg toxicity.

Ammonia oxidation is the rate-limiting step in nitrification process and dominated by ammonia-oxidizing bacteria (AOB) and ammonia-oxidizing archaea (AOA). In the present study, a highly enriched culture of AOA was obtained from urban polluted water in Shahe River, Shenzhen, China. The optimum growth conditions were identified by orthogonal analysis as 37 °C, with pH 7.0 and initial ammonia concentration of 1.0 mM. Under these conditions, the highest abundance of AOA was obtained as 4.6 × 10⁷ copies/ng DNA. Growth of AOA in polluted river water showed significant reduction in ammonia concentration in AOA-enriched cultures without antibiotics after 10 days of incubation, while synchronous increase in nitrate concentration was up to 12.7 mg/L. However, AOA-enriched by antibiotic showed insignificant changes in ammonia or nitrite concentration. This study showed that AOB play an important role in ammonia oxidation of polluted river water, and AOA alone showed insignificant changes in ammonia or nitrite concentrations. Therefore, the ammonia oxidation performance of natural water could not be improved by adding high concentration AOA bacterial liquid.

Cadmium (II) can potentially alter the microbial community structure and molecular ecological network in activated sludge systems. In this study, we used Illumina sequencing combined with an RMT-based network approach to show the response of the microbial community and its network structure to Cd (II) in activated sludge systems. The results demonstrated that 1 mg/L Cd (II) did not have chronic negative effects on chemical oxygen demand (COD) reduction and denitrification processes, but negatively affected the nitrification process and phosphorus removal. In contrast, 10 mg/L Cd (II) adversely affected both COD and nutrient removal, and reduced the microbial diversity and changed the overall microbial community structure. The relative abundances of Nitrosomonadaceae, Nitrospira, Accumulibacter and Acinetobacter, which are involved in nitrogen removal, significantly decreased with increases in the Cd (II) concentration. In addition, molecular ecological network analysis showed that the networks sizes in the presence of higher levels of Cd (II) were smaller than in the control, but the nodes were more closely connected with neighbors. These shifts in bacterial abundance and the bacterial network structure may be responsible for the deterioration of COD and nutrient removal. Overall, this study provides new insights into the effects of Cd (II) on the bacterial community and its interactions in activated sludge systems.

In wheat production areas of China, soil lead (Pb) pollution is generally accompanied by cadmium (Cd) pollution and it is of considerable significance in repairing the Cd and Pb co-contaminated soils for safe agronomic production. Organosilicon fertilizer (OSiF) is a new type of silicon (Si) fertilizer that can effectively alleviate heavy metal toxicity in plants, but the mechanisms on its heavy metal detoxification are poorly understood. A soil pot experiment was conducted to evaluate and compare the effects of two OSiFs (OSiFA and OSiFB) and an inorganic silicon fertilizer (InOSiF) on wheat heavy metal uptake and biochemical parameters in a Cd and Pb co-contaminated soil. The results demonstrated that OSiFA, OSiFB and InOSiF could alleviate the Cd and Pb toxicity of wheat, as indicated by increasing wheat grain yield by 65%, 45% and 22%, respectively. The Si fertilizers enhanced leaf gas exchange attributes and chlorophyll content, whereas diminished the oxidative damage, as indicated by a lower level of hydrogen peroxide (H2O2) and malondialdehyde (MDA) content, and lower activity of superoxide dismutase (SOD) and catalase (CAT) activity, as compared with control. Adding OSiFA, OSiFB and InOSiF increased Si uptake in roots and shoots, thus reducing Cd and Pb accumulation in the wheat shoot, bran and flour, especially, flour Cd contents by 17%, 10% and 31% respectively, flour Pb contents by 74%, 53% and 48% respectively. Also, Si fertilizers application decreased the health risk index (HRI) of both Cd and Pb. The grey correlation degrees of OSiFA, OSiFB and InOSiF are 0.72, 0.77 and 0.61, respectively, indicating that the effects of OSiFs on detoxifying Cd and Pb could be better than that of InOSiF in wheat. Thus, the use of OSiFs might be a feasible approach to reduce Cd and Pb entry into the human body through crops.

Emissions from passenger cars are one of major sources that deteriorate urban air quality. This study presents characterization of real-drive emissions from three Euro 6 emission level passenger cars (two gasoline and one diesel) in terms of fresh particles and secondary aerosol formation. The gasoline vehicles were also characterized by chassis dynamometer studies. In the real-drive study, the particle number emissions during regular driving were 1.1–12.7 times greater than observed in the laboratory tests (4.8 times greater on average), which may be caused by more effective nucleation process when diluted by real polluted and humid ambient air. However, the emission factors measured in laboratory were still much higher than the regulatory value of 6 × 1011 particles km−1. The higher emission factors measured here result probably from the fact that the regulatory limit considers only non-volatile particles larger than 23 nm, whereas here, all particles (also volatile) larger than 3 nm were measured. Secondary aerosol formation potential was the highest after a vehicle cold start when most of the secondary mass was organics. After the cold start, the relative contributions of ammonium, sulfate and nitrate increased. Using a novel approach to study secondary aerosol formation under real-drive conditions with the chase method resulted mostly in emission factors below detection limit, which was not in disagreement with the laboratory findings.

Metal-resistant bacteria can survive exposure to high metal concentrations without any negative impact on their growth. Biosorption is considered to be one of the more effective detoxification mechanisms acting in most bacteria. However, molecular-scale characterization of metal biosorption by wild metal-resistant bacteria has been limited. In this study, the Pb(II) biosorption behavior of Serratia Se1998 isolated from Pb-contaminated soil was investigated through macroscopic and microscopic techniques. A four discrete site non-electrostatic model fit the potentiometric titration data best, suggesting a distribution of phosphodiester, carboxyl, phosphoryl, and amino or hydroxyl groups on the cell surface. The presence of these functional groups was verified by the attenuated total reflection Fourier transform infrared (ATR-FTIR) spectroscopy, which also indicated that carboxyl and phosphoryl sites participated in Pb(II) binding simultaneously. The negative enthalpy (−9.11 kJ mol−1) and large positive entropy (81.52 J mol−1 K−1) of Pb(II) binding with the bacteria suggested the formation of inner-sphere complexes by an exothermic process. X-ray absorption fine structure (XAFS) analysis further indicated monodentate inner-sphere binding of Pb(II) through formation of C−O−Pb and P−O−Pb bonds. We inferred that C−O−Pb bonds formed on the flagellar surfaces, establishing a self-protective barrier against exterior metal stressors. This study has important implications for an improved understanding of metal-resistance mechanisms in wild bacteria and provides guidance for the construction of genetically engineered bacteria for remediation purposes.

The comprehensive sources of particulate matter (PM) require air purification materials to possess both high filtration efficiencies and low air resistances in an effort to provide healthcare. However, the assembly of multiple-layered filters with different functions leads to high pressure drop and high operating cost. Therefore, a multifunctional air filter that can provide excellent air filtration capacity and healthcare is highly desired. Here, a novel bifunctional polyacrylonitrile/attapulgite hierarchical-structured filter with low air resistance and high adsorption capacity was designed and fabricated by embedding attapulgite nanorods during a facile electrospinning process. The hierarchical polyacrylonitrile/attapulgite membranes showed only a ∼64 Pa resistance for 0.1 μm PM. Another benefit of using the attapulgite nanorods is an adsorption effect for hazardous heavy metal ions that accompany airborne ultrafine PMs. Thereby this hierarchical membrane simultaneously exhibits an enhanced filtration performance and hazardous protection ability. Furthermore, due to the electret effect of the attapulgite nanorods, the surface potential of the membrane remains at above 2.2 kV after 600 min of continuous use, which could improve the air filtration efficiency and ensure the long-term service life of the filters. This work may provide a new approach for the design and development of multifunctional air filters for simultaneously capturing ultrafine PMs and any other accompanying hazardous chemicals.

The degradation of trichloroethylene (TCE) and tetrachloroethylene (PCE), in incubations where ammonium was oxidized while iron was being reduced indicates that these compounds can be degraded during the Feammox process by Acidimicrobiaceae sp. A6 (ATCC, PTA-122488). None of these compounds were degraded in incubations to which no ammonium was added, indicating that they were degraded during the oxidation of ammonium. Degradation of TCE and PCE (ranging between 32% and 55%) was observed in incubations with a pure Acidimicrobiaceae sp. A6 culture as well as an Acidimicrobiaceae sp. A6 enrichment culture over a 2-week period. In addition to these batch studies, a column study, with a 5-h hydraulic residence time, was conducted contrasting the degradation of TCE in iron-rich soil columns that were either seeded with a pure or an enrichment culture of Acidimicrobiaceae sp. A6 to achieve ammonium oxidation under iron reduction, and a control column that was initially not seeded and later seeded with Geobacter metallireducens. While there was ∼22% TCE removal in the columns seeded with Acidimicrobiaceae sp. A6, there was no removal in the unseeded column or the column seeded with G. metallireducens which was being operated under iron reducing conditions. Feammox is an anoxic process that requires acidic conditions. Hence, these results indicate that this process might be harnessed where other bioremediation strategies are difficult, since many require neutral or alkaline conditions, and supplying ammonium to an anoxic aquifer is relatively easy, since there are not many processes that will oxidize ammonium in the absence of dissolved oxygen.
