Epigenetic control of heavy metal stress response in mycorrhizal versus non-mycorrhizal poplar plants

It was previously shown that arbuscular mycorrhizal fungi (AMF) exert a significant improvement of growth in a tolerant white poplar (Populus alba L.) clone (AL35) grown on Cu- and Zn-polluted soil via foliar alterations in the levels of defence/stress-related transcripts and molecules. However, nothing is known about the epigenetic changes which occur during tolerance acquisition in response to heavy metals (HMs) in the same mycorrhizal vs. non-mycorrhizal poplar plants. In order to analyse the epigenome in leaves of AL35 plants inoculated or not with AMF and grown in a greenhouse on multimetal polluted or unpolluted soil, the Methylation Sensitive Amplification Polymorphism (MSAP) approach was adopted to detect cytosine DNA methylation. Modest changes in cytosine methylation patterns were detected at first sampling (4 months from planting), whereas extensive alterations (hypomethylation) occurred at second sampling (after 6 months) in mycorrhizal plants grown in the presence of HMs. The sequencing of MSAP fragments led to the identification of genes belonging to several Gene Ontology categories. Seven MSAP fragments, selected on the basis of DNA methylation status in treated vs control AL35 leaves at the end of the experiment, were analysed for their transcript levels by means of qRT-PCR. Gene expression varied in treated samples relative to controls in response to HMs and/or AMF inoculation; in particular, transcripts of genes involved in RNA processing, cell wall and amino acid metabolism were upregulated in the presence of AMF with or without HMs.

In order to examine whether the leaves of the Colorado blue spruce (Picea pungens) are damaged or not by traffic pollution, the traits of the anatomy and physiology of its leaves are investigated by exposure to vehicle exhausts in a laboratory experiment lasting 30 days. The results show that both the anatomical structures and physiological traits of the leaves are significantly affected by vehicle exhausts. The anatomical structures, including epidermis, cuticle, palisade, and spongy parenchyma are modified when exposed to the high concentrations (≥0.4 mg/m³) of vehicle exhausts. However, physiological traits such as total chlorophyll content are not changed when exposed to different concentrations of vehicle exhaust. Unlike the total chlorophyll content, the electrical conductivities increased, whereas the POD activities decreased when presented in vehicle exhausts. The present study indicates that the Colorado blue spruce changes its anatomical structures and physiological traits to avoid possible damage by vehicle exhausts.

This study aims to investigate the influence of the coexistence of halogen ions (bromide/iodide) and biological source matters on the speciation and yield of trihalomethanes (THMs), haloacetic acids (HAAs), and N-nitrosodimethylamine (NDMA) during the ozonation and subsequent chlorination of water. The results show that the concentrations of brominated THMs and iodinated THMs increased with increasing bromide and iodide concentration. These results may be attributed to the higher reactivity of hypobromous acid and hypoiodous acid generated from the ozonation and subsequent chlorination in the presence of bromide or iodide ions. The presence of bromide increased the species of brominated HAAs. There was a shift from chlorinated HAAs to brominated HAAs after increasing the concentration of bromide. The effect of iodide on HAA formation was more complex than bromide. For most samples, the concentration of total HAAs (T-HAAs) increased to the maximum and then decreased with increasing iodide concentration. The components of the organic precursors also significantly influenced the formation of brominated and iodinated disinfection by-products (Br-DBPs and I-DBPs). Humic acids produced more CHBr₃(596.60 μg/L) than other organic materials. Microcystis aeruginosa cells produced the most tribromoacetic acid (TBAA, 84.16 μg/L). Furthermore, the yield of NDMA decreased with increasing bromide concentration, indicating that the formation of NDMA was inhibited by the high concentration of bromide.

Severe perfluoroalkyl acid (PFAA) contamination resulting from the fast-growing semiconductor, electrochemical, and optoelectronic industries has been determined in the river water in the vicinity of the Taipei area, Taiwan, during recent years. However, little is known about body burdens of the PFAA contaminations in local residents, especially children living in the Taipei area recently. In this study, ten target PFAA analytes consisted of three perfluorosulfonates (PFSAs) and seven perfluorocarboxylates (PFCAs) in the blood serum samples, collected from 225 healthy children with an average age of 13.6 years in the Taipei area from 2009 to 2010, were analyzed via high-performance liquid chromatography–tandem mass spectrometry (HPLC–MS/MS). As the dominant PFAA contaminant in the blood serum samples from Taiwanese children, perfluorooctane sulfonate (PFOS) contributed 86 % of all the target PFAA analytes, while the other nine analytes contributed less than 5 % individually. PFOS showed the highest median up to 29 ng/mL, ranging from 0.03 to 148 ng/mL, which was higher than that observed in the serum samples collected from Taiwanese children between 2006 and 2008. Statistically, serum concentrations of perfluorobutane sulfonate (PFBS), perfluorohexane sulfonate (PFHxS), and perfluorooctanoic acid (PFOA) had significantly positive correlations with ages of children (p < 0.05). Furthermore, serum PFBS, PFHxS, and PFOA concentrations in the male children were considerably higher than those in the female children (p = 0.049, p = 0.000, p = 0.000).

To collect a complete dataset regarding the occurrence of organic substances in groundwater, this study presents the examination of 66 organic contaminants in the groundwater of overseas departments, including pesticides, pharmaceutical compounds, hormones and some industrial substances. The selective and sensitive analytical methods are described. These techniques begin with solid-phase extraction (SPE) followed by analysis using liquid chromatography–tandem mass spectrometry (LC-MS/MS) and gas chromatography–mass spectrometry (GC-ToF-MS). The paper summarises the analytical results from 40 sampling points collected during two campaigns in Guadeloupe, Martinique, Reunion, Mayotte and Guiana, representing 80 samples. Of the 66 target substances, 36 were determined at least once. Among the most frequently detected are bisphenol A (frequency, 96 %; max., 7,400 ng/L), caffeine (frequency, 91 %; max., 1,240 ng/L), pentachlorophenol (frequency, 55 %; max., 418 ng/L), and carbamazepine (frequency, 56 %; max., 22 ng/L). The results do not put in evidence that the origin of the sample or climatic characteristics of these regions influence the dilution and release of micropollutants.

Struvite precipitate obtained from yeast industry anaerobic effluent with high ammonium nitrogen (NH₄–N) was investigated for fertilizer effect on plant growth and nutrition according to applications of N, nitrogen/phosphorus/potassium (NPK), and control. Optimum struvite formation conditions were determined via Box–Behnken design. Optimum condition was obtained at pH 9.0 and Mg/N/P molar ratio of 1.5:1:1. Under these conditions, heavy metal concentrations in the obtained struvite precipitate (except Cu) were below the detection limits. In addition to high N, P, and Mg content, energy-dispersive X-ray (EDX) analysis showed that the struvite also included the nutritional elements Ca, K, Na, and Fe. X-ray diffraction (XRD) analysis revealed the complex structures of NaAl(SO₄)₂(H₂O)₁₂, NaMn²⁺Fe₂(PO₄)₃, and (Na₂,Ca)O₂(Fe,Mn)O.P₂O₅in the precipitate. High Na⁺and Ca²⁺concentrations in the anaerobic effluent reacted with phosphate during struvite precipitation. Different applications and struvite dosages significantly affected fresh and dry weights and nutrient element uptakes by plants (P < 0.05). N, P, and Mg uptakes of plants were significantly higher at struvite ×2, ×3, and ×4 dosages compared with NPK application. For adequate nutrition and supply of optimum dry weight, struvite ×2 dosage (5.71 g struvite/kg soil) was found appropriate for both maize and tomato plants.

Many polluted sites are typically characterized by contamination with multiple heavy metals, drought, salinity, and nutrient deficiencies. Here, an Australian native succulent halophytic plant species, Carpobrotus rossii (Haw.) Schwantes (Aizoaceae) was investigated to assess its tolerance and phytoextraction potential of Cd, Zn, and the combination of Cd and Zn, when plants were grown in soils spiked with various concentrations of Cd (20–320 mg kg⁻¹Cd), Zn (150–2,400 mg kg⁻¹Zn) or Cd + Zn (20 + 150, 40 + 300, 80 + 600 mg kg⁻¹). The concentration of Cd in plant parts followed the order of roots > stems > leaves, resulting in Cd translocation factor (TF, concentration ratio of shoots to roots) less than one. In contrast, the concentration of Zn was in order of leaves > stems > roots, with a Zn TF greater than one. However, the amount of Cd and Zn were distributed more in leaves than in stems or roots, which was attributed to higher biomass of leaves than stems or roots. The critical value that causes 10 % shoot biomass reduction was 115 μg g⁻¹for Cd and 1,300 μg g⁻¹for Zn. The shoot Cd uptake per plant increased with increasing Cd addition while shoot Zn uptake peaked at 600 mg kg⁻¹Zn addition. The combined addition of Cd and Zn reduced biomass production more than Cd or Zn alone and significantly increased Cd concentration, but did not affect Zn concentration in plant parts. The results suggest that C. rossii is able to hyperaccumulate Cd and can be a promising candidate for phytoextraction of Cd from polluted soils.

Materials based on polystyrene and starch copolymers are used in food packaging, water pollution treatment, and textile industry, and their biodegradability is a desired characteristic. In order to examine the degradation patterns of modified, biodegradable derivates of polystyrene, which may keep its excellent technical features but be more environmentally friendly at the same time, polystyrene-graft-starch biomaterials obtained by emulsion polymerization in the presence of new type of initiator/activator pair (potassium persulfate/different amines) were subjected to 6-month biodegradation by burial method in three different types of commercially available soils: soil rich in humus and soil for cactus and orchid growing. Biodegradation was monitored by mass decrease, and the highest degradation rate was achieved in soil for cactus growing (81.30 %). Statistical analysis proved that microorganisms in different soil samples have different ability of biodegradation, and there is a significant negative correlation between the share of polystyrene in copolymer and degree of biodegradation. Grafting of polystyrene on starch on one hand prevents complete degradation of starch that is present (with maximal percentage of degraded starch ranging from 55 to 93 %), while on the other hand there is an upper limit of share of polystyrene in the copolymer (ranging from 37 to 77 %) that is preventing biodegradation of degradable part of copolymers.

The degradation of aqueous Rhodamine B (RhB) was examined using a dual-channel spark switch module designed to regulate the steepness of pulsed high voltage with microsecond rise time. Depending on the energy per pulse, a spark along the water surface (SPWS) or streamer along the water surface (STWS) was formed. STWS was found to have a better degradation effect and energy efficiency toward RhB than SPWS at the same power; however, addition of H₂O₂amounts resulted in increased degradation, the effect being more pronounced using SPWS. The initial concentration of RhB also appeared to influence the rate constant of the degradation reaction. Furthermore, TiO₂films doped with Fe, Mn, and Ce were found to enhance the degradation performance of plasma. A possible reaction mechanism of plasma formation along the water surface was concluded by determination of the main inorganic products in the liquid and gas phases.

Severe eutrophication of surface water has been a major problem of increasing environmental concern worldwide. In the present study, economic plant annual ryegrass (Lolium multiflorum) was grown in floating mats as an economic plant-based treatment system to evaluate its potential after ion implantation for removing nutrients in simulated eutrophic water. The specific weight growth rate of L. multiflorum with ion implantation was significantly greater than that of the control, and the peroxidase, nitrate reductase, and acid phosphatase activities of the irradiated L. multiflorum were found to be greater than those plants without ion implantation. Higher total nitrogen (TN) and total phosphorus (TP) removal efficiencies were obtained for the L. multiflorum irradiated with 25 keV 5.2 × 10¹⁶ N⁺ions/cm²and 30 keV 4.16 × 10¹⁶ N⁺ions/cm², respectively (p < 0.05). Furthermore, the nitrogen and phosphorus contents in the plant biomass with ion implantation were also greater than those in the control and were positively correlated with TN and TP supplied. L. multiflorum itself was directly responsible for 39–49 and 47–58 % of the overall N and P removal in the experiment, respectively. The research results suggested that ion implantation could become a promising approach for increasing phytoremediation efficiency of nutrients from eutrophic water by L. multiflorum.
