Effects of sodium arsenate exposure on liver fatty acid profiles and oxidative stress in rats Texto completo

The present study aimed to evaluate the effect of arsenic on liver fatty acids (FA) composition, hepatotoxicity and oxidative status markers in rats. Male rats were randomly devised to six groups (n = 10 per group) and exposed to sodium arsenate at a dose of 1 and 10 mg/l for 45 and 90 days. Arsenate exposure is associated with significant changes in the FA composition in liver. A significant increase of saturated fatty acids (SFA) in all treated groups (p < 0.01) and trans unsaturated fatty acids (trans UFA) in rats exposed both for short term for 10 mg/l (p < 0.05) and long term for 1 and 10 mg/l (p < 0.001) was observed. However, the cis UFA were significantly decreased in these groups (p < 0.05). A markedly increase of indicator in cell membrane viscosity expressed as SFA/UFA was reported in the treated groups (p < 0.001). A significant increase in the level of malondialdehyde by 38.3 % after 90 days of exposure at 10 mg/l was observed. Compared to control rats, significant liver damage was observed at 10 mg/l of arsenate by increasing plasma marker enzymes after 90 days. It is through the histological investigations in hepatic tissues of exposed rats that these damage effects of arsenate were confirmed. The antioxidant perturbations were observed to be more important at groups treated by the high dose (p < 0.05). An increase in the level of protein carbonyls was observed in all treated groups (p < 0.05). The present study provides evidence for a direct effect of arsenite on FA composition disturbance causing an increase of SFA and TFAs isomers, liver dysfunction and oxidative stress. Therefore, arsenate can lead to hepatic damage and propensity towards liver cancer.

Pesticides are frequently detected in estuaries among the pollutants found in estuarine and coastal areas and may have major ecological consequences. They could endanger organism growth, reproduction, or survival. In the context of high-mortality outbreaks affecting Pacific oysters, Crassostrea gigas, in France since 2008, it appears of importance to determine the putative effects of pesticides on oyster susceptibility to infectious agents. Massive mortality outbreaks reported in this species, mainly in spring and summer, may suggest an important role played by the seasonal use of pesticides and freshwater input in estuarine areas where oyster farms are frequently located. To understand the impact of some pesticides detected in French waters, their effects on Pacific oyster hemocytes were studied through short-term in vitro experiments. Bivalve immunity is mainly supported by hemocytes eliminating pathogens by phagocytosis and producing compounds including lysosomal enzymes and antimicrobial molecules. In this study, oyster hemocytes were incubated with a mixture of 14 pesticides and metaldehyde alone, a molecule used to eliminate land mollusks. Hemocyte parameters including dead/alive cells, nonspecific esterase activities, intracytoplasmic calcium, lysosome number and activity, and phagocytosis were monitored by flow cytometry. No significant effect of pesticides tested at different concentrations was reported on oyster hemocytes maintained in vitro for short-term periods in the present study. It could be assumed that these oyster cells were resistant to pesticide exposure in tested conditions and developing in vivo assays appears as necessary to better understand the effects of pollutants on immune system in mollusks.

A field-based evaluation of the biological effects of potential nickel (Ni) exposures was conducted using monitoring data for benthic macroinvertebrates and water chemistry parameters for streams in England and Wales. Observed benthic community metrics were compared to expected community metrics under reference conditions using RIVPACS III+ software. In order to evaluate relationships between Ni concentrations and benthic community metrics, bioavailable Ni concentrations were also calculated for each site. A limiting effect from Ni on the 90th percentile of the maximum achievable ecological quality was derived at “bioavailable Ni” exposures of 10.3 μg l⁻¹. As snails have been identified as particularly sensitive to nickel exposure, snail abundance in the field in response to nickel exposure, relative to reference conditions, was also analysed. A “low effects” threshold for snail abundance based on an average of spring and autumn data was derived as 3.9 μg l⁻¹bioavailable Ni. There was no apparent effect of Ni exposure on the abundance of Ephemeroptera (mayflies), Plecoptera (stoneflies) or Tricoptera (caddisflies) when expressed relative to a reference condition within the range of “bioavailable Ni” exposures observed within the dataset. Nickel exposure concentrations co-vary with the concentrations of other stressors in the dataset, and high concentrations of Ni are also associated with elevated concentrations of other contaminants.

During winter operations at airports, large amounts of organic deicing chemicals (DIC) accumulate beside the runways and infiltrate into the soil during spring. To study the transport and degradation of DIC in the unsaturated zone, eight undisturbed soil cores were retrieved at Oslo airport, Norway, and installed as lysimeters at a nearby field site. Before snowmelt in 2010 and 2011, snow amended with a mix of the DICs propylene glycol (PG) and formate as well as bromide as conservative tracer was applied. Water samples were collected and analyzed until summer 2012. Water flow and solute transport varied considerably among the lysimeters but also temporally between 2010 and 2011. High infiltration rates during snowmelt resulted in the discharge of up to 51 and 82 % PG in 2010 and 2011, respectively. The discharge of formate remained comparatively low, indicating its favored degradation even at freezing temperatures compared with PG. Manganese (Mn) and iron (Fe) were observed in the drainage in autumn owing to the anaerobic degradation of residual PG during summer. Our findings suggest that upper boundary conditions, i.e., snow cover and infiltration rate, and the extent of preferential flowpaths, control water flow and solute transport of bromide and PG during snowmelt. PG may therefore locally reach deeper soil regions where it may pose a risk for groundwater. In the long term, the use of DIC furthermore causes the depletion of potential electron acceptors and the transport of considerable amounts of Fe and Mn. To avoid an overload of the unsaturated zone with DIC and to maintain the natural redox system, the development of suitable remediation techniques is required.

Spectral confocal microscope visualizations of microsphere movement in unsaturated porous media showed that attachment at the Air Water Solid (AWS) interface was an important retention mechanism. These visualizations can aid in resolving the functional form of retention rates of colloids at the AWS interface. In this study, soil adsorption isotherm equations were adapted by replacing the chemical concentration in the water as independent variable by the cumulative colloids passing by. In order of increasing number of fitted parameters, the functions tested were the Langmuir adsorption isotherm, the Logistic distribution, and the Weibull distribution. The functions were fitted against colloid concentrations obtained from time series of images acquired with a spectral confocal microscope for three experiments performed where either plain or carboxylated polystyrene latex microspheres were pulsed in a small flow chamber filled with cleaned quartz sand. Both moving and retained colloids were quantified over time. In fitting the models to the data, the agreement improved with increasing number of model parameters. The Weibull distribution gave overall the best fit. The logistic distribution did not fit the initial retention of microspheres well but otherwise the fit was good. The Langmuir isotherm only fitted the longest time series well. The results can be explained that initially when colloids are first introduced the rate of retention is low. Once colloids are at the AWS interface they act as anchor point for other colloids to attach and thereby increasing the retention rate as clusters form. Once the available attachment sites diminish, the retention rate decreases.

Fifteen plant species—Alternanthera bettzickiana, Drimiopsis botryoides, Aloe vera, Chlorophytum comosum, Aglaonema commutatum, Cordyline fruticosa, Philodendron martianum, Sansevieria hyacinthoides, Aglaonema rotundum, Fittonia albivenis, Muehlenbeckia platyclada, Tradescantia spathacea, Guzmania lingulata, Zamioculcas zamiifolia, and Cyperus alternifolius—were evaluated for the removal efficiency of xylene from contaminated air. Among the test plants, Z. zamiifolia showed the highest xylene removal efficiency. Xylene was toxic to Z. zamiifolia with an LC₅₀of 3,464 ppm. Higher concentrations of xylene exhibited damage symptoms, including leaf tips turning yellow, holonecrosis, and hydrosis. TEM images showed that a low concentration of xylene vapors caused minor changes in the chloroplast, while a high concentration caused swollen chloroplasts and damage. The effect of photosynthetic types on xylene removal efficiency suggests that a mixture of Z. zamiifolia, S. hyacinthoides, and A. commutatum which represent facultative CAM, CAM, and C₃plants, is the most suitable system for xylene removal. Therefore, for maximum improvement in removing xylene volatile compounds under various conditions, multiple species are needed. The effect of a plant’s total leaf area on xylene removal indicates that at lower concentrations of xylene, a small leaf area might be as efficient as a large leaf area.

The removal of 12 pharmaceuticals and personal care products (PPCPs) in two full-scale wastewater treatment plants (WWTPs) and a tertiary treatment system was studied. The ecological risks of effluents from both secondary and tertiary treatment systems as well as excess sludge were evaluated. Primary treatment and ultraviolet light disinfection showed limited ability to remove most selected PPCPs. The combination of an anaerobic process and triple-oxidation ditches can eliminate DEET better than the anaerobic/anoxic/oxic process. Adsorption to sludge played a key role in the removal of triclocarban. Multistage constructed wetlands as a tertiary treatment efficiently removed caffeine and ibuprofen from wastewater and could decrease the risk of partial selected PPCPs. Selected PPCPs residues in excess sludge generally produced higher risks to the ecological environment than effluents from WWTPs.

The occurrence of harmful algal blooms has been increasing significantly around the world. In order to ensure the safety of drinking water, procedures to screen potential materials as effective algicides are needed, and predictive methods which save both the labor and time compared with traditional experimental approaches, are particularly desirable. In this study, data from previous studies on the algal-growth inhibitory action of two kinds of compounds, namely, the action of fatty acids and thiazolidinediones on the harmful algae Heterosigma akashiwo and Chattonella marina, were modeled using multiple linear regression (MLR) based on quantitative structure–activity relationships (QSAR). The models were shown to have highly predictive ability and stability, and provided insight into the inhibitory mechanisms of congeneric compounds. The main descriptors in the fatty-acid models were the Connolly accessible area and the number of rotatable bonds, illustrating that molecular surface area and shape are important in their algicidal actions. In the thiazolidinedione models, the critical volume, octanol–water partition coefficient (LogP), and Connolly solvent-excluded volume were found to be significant, indicating that hydrophobicity, substituent group size, and mode of action are mechanistically important. Our results showed the algicidal activity of a series of compounds on different algae could be modeled, and each model is efficacious for compounds that fall into the application domain of the QSAR model. This work demonstrates how reliable predictions of the algicidal activity of novel compounds and explanations of their inhibitory mechanisms can be obtained.

An amorphous Zn biomineralization (“white mud”), occurring at Naracauli stream, Sardinia, in association with cyanobacteria Leptolyngbya frigida and diatoms, was investigated by electron microscopy and X-ray absorption spectroscopy. Preliminary diffraction analysis shows that the precipitate sampled on Naracauli stream bed is mainly amorphous, with some peaks ascribable to quartz and phyllosilicates, plus few minor unattributed peaks. Scanning electron microscopy analysis shows that the white mud, precipitated in association with a seasonal biofilm, is made of sheaths rich in Zn, Si, and O, plus filaments likely made of organic matter. Transmission electron microscopy analysis shows that the sheaths are made of smaller units having a size in the range between 100 and 200 nm. X-ray absorption near-edge structure and extended X-ray absorption fine structure data collected at the Zn K-edge indicate that the biomineral has a local structure similar to hemimorphite, a zinc sorosilicate. The differences of this biomineral with respect to the hydrozincite biomineralization documented about 3 km upstream in the same Naracauli stream may be related to either variations in the physicochemical parameters and/or different metabolic behavior of the involved biota.

In recent years, occupational and environmental exposure to toxic pollutants has increasingly contributed to declining sperm quality and increasing morbidity of human male genital diseases. This study explored the effects of electronic waste (e-waste) environmental pollutions on male genital health in Guiyu, one of the largest e-waste recycling centers in the world. We collected outpatient case information from 2001 to 2009 in Guiyu and a control hospital and performed statistical analysis on male genital diseases morbidity (MGDM). The MGDM in Guiyu and the control hospital per thousand from 2004 to 2009 were 1.410/0.403 (2004), 0.539/0.385 (2005), 0.248/0.284 (2006), 0.485/0.195 (2007), 1.107/0.272 (2008), and 0.741/0.586 (2009) while the average total MGDM from 2004 to 2009 were 0.753 and 0.355 per thousand, respectively. Percentage of occurrence of epididymitis, impotence and prospermia, redundant prepuce, gonorrhea, urethritis, sexual function dysfunction, azoospermia, asthenospermia, and unknown etiology male sterility were higher in Guiyu (P < 0.05), whereas the frequency of prostatitis, condyloma accuminatum, and genital herpes were higher in the control (P < 0.05). Morbidity of male genital diseases was higher in Guiyu than in the control area. Male reproductive health may be threatened by e-waste environmental pollution in Guiyu, especially for diseases that could be influenced by environmental factors, and it may influence local population diathesis.
