Oxidative stress in duckweed (Lemna minor L.) induced by glyphosate: Is the mitochondrial electron transport chain a target of this herbicide?

We investigated the physiological responses of Lemna minor plants exposed to glyphosate. The deleterious effects of this herbicide on photosynthesis, respiration, and pigment concentrations were related to glyphosate-induced oxidative stress through hydrogen peroxide (H2O2) accumulation. By using photosynthetic and respiratory electron transport chain (ETC) inhibitors we located the primary site of reactive oxygen species (ROS) production in plants exposed to 500 mg glyphosate l−1. Inhibition of mitochondrial ETC Complex I by rotenone reduced H2O2 concentrations in glyphosate-treated plants. Complex III activity was very sensitive to glyphosate which appears to act much like antimycin A (an inhibitor of mitochondrial ETC Complex III) by shunting electrons from semiquinone to oxygen, with resulting ROS formation. Confocal evaluations for ROS localization showed that ROS are initially produced outside of the chloroplasts upon initial glyphosate exposure. Our results indicate that in addition to interfering with the shikimate pathway, glyphosate can induce oxidative stress in plants through H2O2 formation by targeting the mitochondrial ETC, which would explain its observed effects on non-target organisms.
Mostrar más [+] Menos [-]Evaluating the predictability of PM10 grades in Seoul, Korea using a neural network model based on synoptic patterns

As of November 2014, the Korean Ministry of Environment (KME) has been forecasting the concentration of particulate matter with diameters ≤ 10 μm (PM10) classified into four grades: low (PM10 ≤ 30 μg m−3), moderate (30 < PM10 ≤ 80 μg m−3), high (80 < PM10 ≤ 150 μg m−3), and very high (PM10 > 150 μg m−3). The KME operational center generates PM10 forecasts using statistical and chemistry-transport models, but the overall performance and the hit rate for the four PM10 grades has not previously been evaluated. To provide a statistical reference for the current air quality forecasting system, we have developed a neural network model based on the synoptic patterns of several meteorological fields such as geopotential height, air temperature, relative humidity, and wind. Hindcast of the four PM10 grades in Seoul, Korea was performed for the cold seasons (October–March) of 2001–2014 when the high and very high PM10 grades are frequently observed. Because synoptic patterns of the meteorological fields are distinctive for each PM10 grade, these fields were adopted and quantified as predictors in the form of cosine similarities to train the neural network model. Using these predictors in conjunction with the PM10 concentration in Seoul from the day before prediction as an additional predictor, an overall hit rate of 69% was achieved; the hit rates for the low, moderate, high, and very high PM10 grades were 33%, 83%, 45%, and 33%, respectively. Our findings also suggest that the synoptic patterns of meteorological variables are reliable predictors for the identification of the favorable conditions for each PM10 grade, as well as for the transboundary transport of PM10 from China. This evaluation of PM10 predictability can be reliably used as a statistical reference and further, complement to the current air quality forecasting system.

Spatial-temporal distributions, sources identification and risk assessment of polycyclic aromatic hydrocarbons (PAHs) in overlying water and surface sediments in urban river networks of Shanghai were studied. Analytical results showed that there was a significant seasonal variation in concentrations of ∑16PAHs in water, suspended particulate matter (SPM) and sediment phases in this study area. The PAHs pollution in these multi-phases were in the medium level compared with other areas around the world, and the levels of PAHs contamination in SPM and sediment phases in hierarchical rivers showed TS (the third-order stream) > FS (the first-order stream) > SS (the second-order stream). Two manners of isomer ratios and principal component analysis (PCA) were used to identify PAHs origins, and suggested that combustion processes are dominant for PAHs sources. The ratios of PAHs origins by fossil fuels combustion, coke burning and crude oil in hierarchical rivers were determined with FS > SS > TS in SPM and sediment phases, and the ratio of PAHs origins by traffic emissions was analyzed with TS > SS > FS. PAHs in water samples have a certain impact on aqueous ecological system especially due to the fact that the ∑ceq values of nine PAHs were calculated from 0.715 to 15.831 μg/L in winter, which inferred serious ecological risk to some special aquatic organisms. The calculations of MERMQ in sediment samples showed that the MERMQ values ranged from 0.021 to 1.209 in winter and 0.019 to 0.643 in summer, which suggested high toxicity at six sampling sites in winter and only one location in summer due to high levels of PAHs. Furthermore, the toxicity degree of sediments were demonstrated with TS > FS > SS.
Mostrar más [+] Menos [-]Fractionation of airborne particulate-bound elements in haze-fog episode and associated health risks in a megacity of southeast China

Haze caused by high particulate matter loadings is an important environmental issue. PM2.5 was collected in Nanjing, China, during a severe haze–fog event and clear periods. The particulate-bound elements were chemically fractionated using sequential extractions. The average PM2.5 concentration was 3.4 times higher during haze–fog (96–518 μg/m³) than non-haze fog periods (49–142 μg/m³). Nearly all elements showed significantly higher concentrations during haze–fog than non-haze fog periods. Zn, As, Pb, Cd, Mo and Cu were considered to have higher bioavailability and enrichment degree in the atmosphere. Highly bioavailable fractions of elements were associated with high temperatures. The integrated carcinogenic risk for two possible scenarios to individuals exposed to metals was higher than the accepted criterion of 10⁻⁶, whereas noncarcinogenic risk was lower than the safe level of 1. Residents of a city burdened with haze will incur health risks caused by exposure to airborne metals.

The rising application of silver nanoparticles (AgNPs) and subsequent release into aquatic systems have generated public concerns over their potential risk and harm to aquatic organisms and human health. Effective and practical analytical methods for AgNPs are urgently needed for their risk assessment. In this study we established an innovative approach to detect trace levels of AgNPs in environmental water through integrating a filtration technique into surface-enhanced Raman spectroscopy (SERS) and compared it with previously established centrifuge-based method. The purpose of filtration was to trap and enrich salt-aggregated AgNPs from water samples onto the filter membrane, through which indicator was then passed and complexed with AgNPs. The enhanced SERS signals of indicator could reflect the presence and quantity of AgNPs in the samples. The most favorable benefit of filtration is being able to process large volume samples, which is more practical for water samples, and greatly improves the sensitivity of AgNP detection. In this study, we tested 20 mL AgNPs-containing samples and the filter-based method is able to detect AgNPs as low as 5 μg/L, which is 20 folds lower than the centrifuge-based method. In addition, the speed and precision of the detection were greatly improved. This approach was used to detect trace levels of AgNPs in real environmental water successfully. Meanwhile, the heteroaggregation of AgNPs with minerals in water was reliably monitored by the new method. Overall, a combination of the filtration-SERS approach provides a rapid, simple, and sensitive way to detect AgNPs and analyze their environmental behavior.

Waterborne metal uptake has been extensively studied and dietary metal assimilation is increasingly recognized in fish, whilst the interaction between the two uptake routes is largely overlooked. This study compared the waterborne Cu bioaccumulation (⁶⁵Cu as tracer) in a juvenile rabbitfish at different feeding regimes (starvation (SG), feeding normal diet (NDG) or diet supplemented with extra Cu (DCG)) to test the hypothesis that feeding can influence waterborne metal uptake in marine fish. NDG and DCG diet was fed as a single meal and then all fish were exposed to waterborne ⁶⁵Cu for 48 h, during which the time course sampling was conducted to determine ⁶⁵Cu bioaccumulation, chyme flow and dietary Cu assimilation. The results revealed that SG fish accumulated the highest ⁶⁵Cu, followed by NDG (61% of SG), whilst DCG fish accumulated the lowest ⁶⁵Cu (34% of SG). These results suggested a protective effect of feeding against waterborne Cu bioaccumulation. This effect was most notable between 10 min and 16 h when there was chyme in gastrointestinal tract (GT). Dietary Cu assimilation mainly occurred before 16 h after feeding. Waterborne ⁶⁵Cu influx rate in the GT was positively correlated with ⁶⁵Cu contents of chyme in NDG, whereas it was largely negatively correlated with ⁶⁵Cu contents of chyme in DCG. The waterborne Cu uptake in the GT was mainly influenced by the chyme flow and dietary Cu assimilation. Overall, our findings suggested that feeding has an important effect on waterborne metal uptake and that both the feeding status of the fish and the relative metal exposure through water and food should be considered in prediction of the metal bioaccumulation and biomonitoring programs.

This study presents a detailed kinetic investigation on the uptake, acropetal translocation and transformation of BDE-47, 6-OH-BDE-47 and 6-MeO-BDE-47 in maize (Zea mays L.) by hydroponic exposure. Root uptake followed the order: BDE-47 > 6-MeO-BDE-47 > 6-OH-BDE-47, while 6-OH-BDE-47 was the most prone to acropetal translocation. Debromination rates of BDE-47 were 1.31 and 1.46 times greater than the hydroxylation and methoxylation rates, respectively. Transformation from BDE-47 to lower brominated OH/MeO-PBDEs occurred mainly through debromination first followed by hydroxylation or methoxylation. There was no transformation from 6-OH-BDE-47 or 6-MeO-BDE-47 to PBDEs. Methylation rate of 6-OH-BDE-47 was twice as high as that of 6-MeO-BDE-47 hydroxylation, indicating methylation of 6-OH-BDE-47 was easier and more rapid than hydroxylation of 6-MeO-BDE-47. Debromination and isomerization were potential metabolic pathways for 6-OH-BDE-47 and 6-MeO-BDE-47 in maize. This study provides important information for better understanding the mechanism on plant uptake and transformation of PBDEs.

Urbanizations have increased the loadings of reactive nitrogen in urban riverine environments. However, limited information about dissimilatory nitrate reduction processes and associated contributions to nitrogen removal is available for urban riverine environments. In this study, sediment slurry experiments were conducted with nitrogen isotope-tracing technique to investigate the potential rates of denitrification, anaerobic ammonium oxidation (anammox) and dissimilatory nitrate reduction to ammonium (DNRA) and their contributions to nitrate reduction in sediments of urban river networks, Shanghai. The potential rates of denitrification, anammox and DNRA measured in the study area ranged from 0.193 to 98.7 nmol N g−1 h−1 dry weight (dw), 0.0387–23.7 nmol N g−1 h−1 dw and 0–10.3 nmol N g−1 h−1 dw, respectively. Denitrification and DNRA rates were higher in summer than in winter, while anammox rates were greater in winter than in summer for most sites. Dissolved oxygen, total organic carbon, nitrate, ammonium, sulfide, Fe(II) and Fe(III) were found to have significant influence on these nitrate reduction processes. Denitrification contributed 11.5–99.5%% to total nitrate reduction, as compared to 0.343–81.6% for anammox and 0–52.3% for DNRA. It is estimated that nitrogen loss of approximately 1.33 × 105 t N year−1 was linked to both denitrification and anammox processes, which accounted for about 20.1% of total inorganic nitrogen transported annually into the urban river networks of Shanghai. Overall, these results show the potential importance of denitrification and anammox in nitrogen removal and provide new insight into the mechanisms of nitrogen cycles in urban riverine environments.

The cycling of sulfur (S), iron (Fe) and phosphorus (P) in sediments and pore water can impact the water quality of overlying water. In a heavily polluted river estuary (Yantai, China), vertical profiles of fluxes of dissolved sulfide, Fe2+ and dissolved reactive phosphorus (DRP) in sediment pore water were investigated by the Diffusive Gradients in Thin films technique (DGT). Vertical fluxes of S, Fe, P in intertidal sediment showed the availability of DRP increased while the sulfide decreased with depth in surface sediment, indicating that sulfide accumulation could enhance P release in anoxic sediment. In sites with contrasting salinity, the relative dominance of iron and sulfate reduction was different, with iron reduction dominant over sulfate reduction in the upper sediment at an intertidal site but the reverse true in a freshwater site, with the other process dominating at depth in each case. Phosphate release was largely controlled by iron reduction.

The aim of this study was to assess the impact of treated wastewaters on native wild Prussian carp inhabiting effluent-receiving waters (ERC) receiving municipal and sugar plant treated wastewaters, further downstream waters (DW), and a detached canal unaffected by the WWTP activities. To that end, general fish health status was determined, including plasma biochemical, haematological, oxidative stress and tissue histopathological indices, over three seasons. The greatest tissue alterations were in fall in ERC during sugar beet processing, as hypertrophy of gill epithelial and interlamellar cells, necrosis and lymphocytic infiltration, hyperplasia and hypertrophy of renal tubules, distention of hepatic sinusoids. In fall the lowest leukocytes, lymphocytes and granulocytes (2467 ± 565, 1333 ± 264, 1133 ± 488 cells/μL respectively), as well as highest plasma ALP (52.7 ± 19.39 U/L) were measured. ERC in fall had the highest ammonium (20 mg/L), nitrite (1.48 mg/L), nitrate (13.4 mg/L), and lowest dissolved O2 (1.23 mg/L). Gill, kidney and liver alterations, and the highest plasma cholesterol (9.1 ± 1.98 mmol/L) were noted in DW fish in fall. Tissue morphology during sugar cane processing seems a consequence of cellular and structural tissue integrity loss. Structural heterogeneity of gills and spleen was enhanced with increasing concentrations of heavy metals and correlated with oxidative stress (SOD 392.5 ± 77.28 U/L). Monogenean infestation was moderate in ERC fish in all seasons compared with DW fish. Prussian carp biological responses to multiple stressors, measured by the effects of WWTP on blood and tissue parameters, reached far downstream and were not of localized nature. This study demonstrated that in aquatic environments impacted with complex contaminants acting synergistically, causal relationships between biological responses and environmental stressors should be interpreted. Integrated histopathological, haematological and biochemical findings are valuable biomarkers for native fish adaptive patterns and monitoring of water quality/pollution of freshwater ecosystems.
