Determination of multiple bisphenol analogues and their metabolites in human serum by liquid chromatography tandem mass spectrometry

To date, knowledge of internal human exposure to BPA and its analogues (particularly bisphenol S and bisphenol F, etc.) remains limited. In the present study, a method involving dispersive solid-phase extraction and LC/MS was proposed to investigate the contamination levels of 28 precursor bisphenols and 9 major metabolites in serum. The critical variables of preparation method were screened out by Plackett-Burman design and further optimized by central composite design. Left in optimal conditions, a total of 286 samples consisting of 153 males and 133 females were analyzed. The results showed that BPA dominated over all the cases with the highest positive rate (82.2% of all the surveyed people), and totally four metabolites (BPA β-D-glucuronide, BPA monosulfate, BPA bis-(β-D-glucuronide) and BPS monosulfate) were detectable. The occurrence of BPA bis-(β-D-glucuronide) in serum is reported for the first time and its higher positive rate and contamination concentrations suggested that it may be a more important metabolite of BPA than others. Negligible potential risk of health effects to blood donors was observed, since the estimated exposure levels (mean 32.1 ng/kg bw/day, 95th 123.2 ng/kg bw/day) were well below far less than the temporary tolerable reference dose of BPA that recommended by the European Food Safety Authority (4 μg/kg bw/day by). The reference level of BPA for healthy population was determined to be 4.09 μg/L via the percentile method.

Aquaculture has significant impacts on freshwater lakes, but plankton communities, as key components of the microbial food web, are rarely considered when assessing the impacts of aquaculture. Revealing the dynamics of plankton communities, including bacterioplankton, phytoplankton and zooplankton, under anthropological disturbances is critical for predicting the freshwater ecosystem functioning in response to future environmental changes. In the present study, we examined the impacts of aquaculture on water quality, plankton diversity and the co-occurrence patterns within plankton metacommunities in a shallow freshwater lake. The study zones are influenced by the 20-year historical intensive aquaculture, but now they are undergoing either ecological aquaculture or ecological restoration. Our results showed that ecological aquaculture was more efficient in nitrogen removal than ecological restoration. Moreover, lower bacterioplankton diversity but higher phytoplankton and zooplankton diversity were found in the ecological aquaculture and ecological restoration zones compared to the control zone. The lower network connectivity of the plankton metacommunities in the ecological aquaculture and ecological restoration zones indicated the decreasing complexity of potential microbial food web, suggesting a possible lower resistance of the plankton metacommunities to future disturbance. Furthermore, plankton communities of different trophic levels were driven under distinct mechanisms. The bacterioplankton community was primarily affected by abiotic factors, whereas the phytoplankton and zooplankton communities were explained more by trophic interactions. These results revealed the impacts of aquaculture on the plankton communities and their potential interactions, thereby providing fundamental information for better understanding the impacts of aquaculture on freshwater ecosystem functioning.

Steroid estrogens have been detected in oceans, rivers, lakes, groundwaters, soils, and even urban water supply systems, thereby inevitably imposing serious impacts on human health and ecological safety. Indeed, many estrogen-degrading bacterial strains and degradation pathways have been reported, with the 4,5-seco pathway being particularly important. However, few studies have evaluated the use of the 4,5-seco pathway by actinomycetes to degrade 17β-estradiol (E2). In this study, 5 genes involved in E2 degradation were identified in the Rhodococcus equi DSSKP-R-001 (R-001) genome and then heterologously expressed to confirm their functions. The transformation of E2 with hsd17b14 reached 63.7% within 30 h, resulting in transformation into estrone (E1). Furthermore, we found that At1g12200-encoded flavin-binding monooxygenase (FMOAₜ₁g₁₂₂₀₀) can transform E1 at a rate of 51.6% within 30 h and can transform E1 into 4-hydroxyestrone (4-OH E1). In addition, catA and hsaC genes were identified to further transform 4-OH E1 at a rate of 97–99%, and this reaction was accomplished by C–C cleavage at the C4 position of the A ring of 4-OH E1. This study represents the first report on the roles of these genes in estrogen degradation and provides new insights into the mechanisms of microbial estrogen metabolism and a better understanding of E2 degradation via the 4,5-seco pathway by actinomycetes.

Increasing rates of commercialization and industrialization have led to the comprehensive evaluation of toxic effects of microplastics on crop plants. However, research on the impact of functionalized polystyrene nanoplastics on the toxicity of heavy metals remains limited. This study investigated the effects of polystyrene, carboxy-modified polystyrene, and amino-modified polystyrene on lead (Pb) toxicity in dandelion seedlings. The results showed that carboxy -modified polystyrene with a negative charge absorbed more Pb²⁺ than polystyrene and amino-modified polystyrene, and their maximum adsorption amounts were 5.328, 0.247, and 0.153 μg g⁻¹, respectively. The hydroponic experiment demonstrated that single amino-modified polystyrene was more toxic to dandelion seedlings than polystyrene and carboxy-modified polystyrene. The presence of Pb²⁺ was found to increase antioxidant enzymes (superoxide dismutase and catalase) and non-antioxidant enzymes (glutathione and ascorbic acid) activities in response to excessive reactive oxygen species in dandelion leaves and roots treated with polystyrene and carboxy-modified polystyrene, while it did not change much when amino-modified polystyrene was added. Interestingly, compared with single Pb²⁺, the addition of amino-modified polystyrene with positive charges induced an obvious decrease in the above parameters; however, they declined slightly in the treatments with polystyrene and carboxy-modified polystyrene despite a stronger adsorption capacity for Pb²⁺. Similarly, the bioactive compounds, including flavonoids, polyphenols, and polysaccharides in dandelion, showed a scavenging effect on O₂⁻ and H₂O₂, thereby inhibiting the accumulation and reducing medicinal properties. This study found that the effects of microplastics on the uptake, distribution, and toxicity of heavy metals depended on the nanoparticle surface charge.
Mostrar más [+] Menos [-]Effect of landfill age on the physical and chemical characteristics of waste plastics/microplastics in a waste landfill sites

The landfills store a lot of waste plastics, thus it has been confirmed a main source for the occurrence of plastics/microplastic. Although there are some reports that microplastics (MPs) can generate in leachate and refuse samples from the landfill, it exist many blanks for the evolution of physical and chemical characteristics of waste plastics and microplastics with different landfill age. To explore the process that large pieces of plastic are fractured into microplastics, the waste plastics with landfill age from 7 to 30 years are surveyed from a typical landfill in Shanghai. The results show that PE and PP are the most common types of landfilling plastics, and their chemical composition also have changed due to the creation of CO and –OH. Moreover, the crystallinity is affected by plastic type and landfill age. The crystallinity of PP increased from 24.9% to 56.8%, but for PE, the crystallinity decreased from 55.6% to 20.8%. The mechanical properties of waste plastics were reduced significantly, which may be caused by changes in carbon-chain molecules. Al, Ti, Co, and other metal elements were detected on the plastic surface. The hydrophobic behavior of waste plastic is constantly decreasing (102.2°–80.1°) under long-term landfilling. By investigating the changes in the physical and chemical characteristics of waste plastics with different landfill age can shed light upon the process of environmental weathering of waste plastics. This provide theoretical guidance for reducing the transport of microplastics to the environment.

Cities constitute an important source of greenhouse gases, but few results originating from long-term, direct CO₂ emission monitoring efforts have been reported. In this study, CO₂ emissions were quasi-continuously measured in an urban center in Sakai, Osaka, Japan by the eddy covariance method from 2010 to 2021. Long-term CO₂ emissions reached 22.2 ± 2.0 kg CO₂ m⁻² yr⁻¹ from 2010 to 2019 (± denotes the standard deviation) in the western sector from the tower representing the densely built-up area. Throughout the decade, the annual CO₂ emissions remained stable. According to an emission inventory, traffic emissions represented the major source of CO₂ emissions within the flux footprint. The interannual variations in the annual CO₂ flux were positively correlated with the mean annual traffic counts at two highway entrances and exits. The CO₂ emissions decreased suddenly, by 32% ± 3.1%, in April and May 2020 during the period in which the first state of emergency associated with COVID-19 was declared. The annual CO₂ emissions also decreased by 25% ± 3.1% in 2020. Direct long-term observations of CO₂ emissions comprise a useful tool to monitor future emission reductions and sudden disruptions in emissions, such as those beginning in 2020 during the COVID-19 pandemic.

In recent years, pollution of antibiotics and heavy metal has often been reported in organic wastes. Saprophytic insects have been recorded as biological control agents in organic waste management. During organic waste conversion, the intestinal bacteria of the saprophytic insects play an important role in digestion, physiology, immunity and prevention of pathogen colonization. Black soldier fly (BSF) Hermetia illucens has been widely used as saprophytic insects and showed tolerance to sulfonamides (SAs) and cadmium (Cd). Diversity and changes in gut microbiota of black soldier fly larvae (BSFL) were evaluated through 16S rRNA high-throughput sequencing, and a decrease in diversity of gut microbiota along with an increase in SAs stress was recorded. Major members identified were Actinomycetaceae, Enterobacteriaceae, and Enterococcaceae. And fourteen multi-resistance Klebsiella pneumoniae strains were isolated. Two strains BSFL7-B-5 (from middle midgut of 7-day BSFL) and BSFL11-C-1 (from posterior midgut of 11-day BSFL) were found to be low-toxic and multi-resistance. The adsorption rate of SAs in 5 mg/kg solutions by these two strains reached 65.2% and 61.6%, respectively. Adsorption rate of Cd in 20 mg/L solutions was 77.2% for BSFL7-B-5. The strain BSFL11-C-1 showed higher than 70% adsorption rates of Cd in 20, 30 and 40 mg/L solutions. This study revealed that the presence of multi-resistance bacterial strains in the gut of BSFL helped the larvae against SAs or Cd stress. After determining how and where they are used, selected BSFL gut bacterial strains might be utilized in managing SAs or Cd contamination at suitable concentrations in the future.

Airborne fine particles can affect climate change and human health; moreover, they can be transported over significant distances. However, studies on characteristics of individual particles and their morphology, elemental composition, aging processes, and spatial distribution after long-range transport over the Yellow Sea are limited. Therefore, in this study, we conducted shipborne measurements of fine particulate matter of less than 2.5 μm in diameter (PM₂.₅) over the Yellow Sea and classified the individual particles into seven types based on their morphology and composition. Overall, the percentage of organic-rich particles was the highest, followed by that of sea spray, sulfur-rich, dust, metals, fly ash, soot, and other particles. Near Shandong, China, the percentage of fly ash and sulfur-rich particles increased, while an increased percentage of only sulfur-rich particles was observed near the Korean Peninsula. In the open sea, the PM₂.₅ concentrations were the lowest, and sea spray particles predominated. During the cruises, three types (Types 1, 2, and 3) of events with substantially increased PM₂.₅ concentrations occurred, each with different dominant particles. Type 1 events frequently featured air masses from northern China and Mongolia with high wind speeds and increased dust particles. Type 2 events involved air masses from China with high wind speeds; fly ash, soot, organic-rich particles, and the sulfate percentage in PM₂.₅ increased. Type 3 events displayed stagnant conditions and local transport (from Korea); soot, dust particles, and the secondary sulfate and nitrate percentages in PM₂.₅ increased. Thus, different types of transport affected concentrations and dominant types of fine particles over the Yellow Sea during spring.

The bioaccumulation and biomagnification of perfluoroalkyl acids (PFAAs) in temperate urban lacustrine ecosystems is poorly understood. We investigated the occurrence and trophic transfer of and probabilistic health risk from 15 PFAAs in the food web of Luoma Lake, a temperate urban lake in East China. The target PFAAs were widely distributed in the water (∑PFAA: 77.09 ± 9.07 ng/L), suspended particulate matter (SPM) (∑PFAA: 284.07 ± 118.05 ng/g dw), and sediment samples (∑PFAA: 67.77 ± 17.96 ng/g dw) and occurred in all biotic samples (∑PFAA: 443.27 ± 124.89 ng/g dw for aquatic plants; 294.99 ± 90.82 for aquatic animals). PFBA was predominant in water and SPM, with 40.11% and 21.35% of the total PFAAs, respectively, while PFOS was the most abundant in sediments (14.11% of the total PFAAs) and organisms (14.33% of the total PFAAs). Sediment exposure may be the major route of biological uptake of PFAAs. The PFAA accumulation capacity was the highest in submerged plants, followed by emergent plants > bivalves > crustaceans > fish > floating plants. Long-chain PFAAs were biomagnified, and short-chain PFAAs were biodiluted across the entire lacustrine food web. PFOS exhibited the greatest bioaccumulation and biomagnification potential among the target PFAAs. However, biomagnification of short-chain PFAAs was also observed within the low trophic-level part of the food web. Human health risk assessment indicated that perfluorooctanesulfonate (PFOS) and perfluorooctanoic acid (PFOA) posed health risks to all age groups, while the other PFAAs were unlikely to cause immediate harm to consumers in the region. This study fills a gap in the knowledge of the transfer of PFAAs in the food webs of temperate urban lakes.

The presence of contaminants of emerging concern in the aquatic environment directly impacts water-living organisms and can alter their living functions. These compounds are often metabolized and excreted, but they can also be accumulated and spread through the food chain. The metabolized contaminants can also lead to the formation of new compounds with unknown toxicity and bioaccumulation potential. In this work, we have studied the occurrence, bioconcentration, and biotransformation of CECs in glass eels (Anguilla anguilla) using UHPLC-HRMS. To select the target CECs, we first carried out an environmental risk assessment of the WWTP effluent that releases directly into the Adour estuary (Bayonne, Basque Country, France). The risk quotients of every detected contaminant were calculated and three ecotoxicologically relevant contaminants were chosen to perform the exposure experiment: propranolol, diazepam, and irbesartan. An experiment of 14 days consisting of 7 days of exposure and 7 days of depuration was carried out to measure the bioconcentration of the chosen compounds. The quantitative results of the concentrations in glass eel showed that diazepam and irbesartan reached BCF ≈10 on day 7, but both compounds were eliminated after 7 days of depuration. On the other hand, propranolol's concentration remains constant all along with the experiment, and its presence can be detected even in the non-exposed control group, which might suggest environmental contamination. Two additional suspect screening strategies were used to identify metabolization products of the target compounds and other xenobiotics already present in wild glass eels. Only one metabolite was identified, nordiazepam, a well-known diazepam metabolite, probably due to the low metabolic rate of glass eels at this stage. The xenobiotic screening confirmed the presence of more xenobiotics in wild glass eels, prominent among them, the pharmaceuticals exemestane, primidone, iloprost, and norethandrolone.
