Year-to-year Variations in Antioxidant Components of High-Lycopene Tomato (Solanum lycopersicum L.) Breeding Lines Texto completo

A two-year field study (2013−2014) was performed to evaluate phytochemical content and antioxidant activity of two high-lycopene tomato breeding lines (HLT-F71 and HLT-F72) grown under open field conditions as compared to the traditional cultivar (cv) Rio Grande. Two open-field trials were conducted in 2013 and 2014. Three tomato cvs were used: two high-lycopene tomato advanced breeding lines (HLT-F71 and ‘HLT-F72’) (F7 generation), and the open-pollinated cv Rio Grande commonly grown in Tunisia. Regardless the growing years and cvs, lycopene content ranged from 98.8 to 280.0 mg/kg fw, total phenolics from 176.1 to 831.8 mg GAE/kg fw flavonoids from 169.7 to 552.1 mg RE/kg fw and total vitamin C from 183.4 to 370.0 mg/kg fw. Significant year-to-year variability was only detected in total phenolics and flavonoid content. No significant year-to-year variability in antioxidant activity was detected in high-lycopene lines. Disregarding the cv, tomato berries harvested on July 2014 had 143% - 224% higher total phenolics and 8% to 47% higher flavonoid contents compared to tomato berries harvested on July 2013. However, disregarding the cv, tomato berries harvested on July 2013 had 6% -17% higher lycopene and 8% -21% higher LAA compared to berries harvested on 2014. During the second growing year, the higher temperature, particularly prior to harvest, determined a decrease in lycopene content and a significant increase in total phenolics regardless the cv. Although the huge variability in total phenolics and the decrease in lycopene content during the second harvest, the antioxidant activity was not affected by year-to-year variability. This reveals consistent functional quality of these cvs and minimal genotype*environmental conditions interaction.

In this study was carried out to determine the effects of humic acid in heavy metal stress of lettuce changes in some of the morphological and physiological characteristics. In studies conducted in controlled greenhouse conditions, lettuce plants subjected to four different 4 different humic acid levels humic acid doses (0, 2, 4, 8 L/da) and 4 different heavy metal irrigation levels (Control: 0 ppm; I. mixture: 0.2 ppm Cu + 0.01 ppm Cd + 5 ppm Pb + 2 ppm Zn; II. mixture: : 0.4 ppm Cu + 0.02 ppm Cd + 10 ppm Pb + 4 ppm Zn; III. mixture: 0.8 ppm Cu + 0.04 ppm Cd + 20 ppm Pb + 8 ppm Zn). Four weeks after at heavy metal and humic acid treatment young plants were harvested and the shoot fresh and dry weight, root fresh and dry weight, shoot and root length, and leaf areas, MDA, superoxyde dismutase (SOD) and glutathione reductase (GR) were determined. The most highly toxic effects were determinate III. mixture level (0.8 ppm Cu + 0.04 ppm Cd + 20 ppm Pb + 8 ppm Zn). MDA and antioxidative enzyme activities increased in plants irrigated with water containing a mixture of heavy metal. Humic acid applications had a positive effect on reducing of the limiting effect of heavy metal stress on growth and development.

In the present study effects of different levels of hydrolyzed oak tannin on Japanese quail, live body weight and live body weight gain, food intake, food conversion efficiency and mortality rate were investigated. Therefore, a total of 240 quails were weighed and divided into 4 treatment groups each with 3 replicates containing 20 birds and reared in flat for 42 days as experimental period. While one of the groups was fed control diet with no additives (0% tannin A), other groups were fed with the different percentages of tannin on their diets, which are 0.5% (B), 1.0% (C), 1.5% (D) respectively. It has been observed that Japanese quails that fed the oak tannin containing diet had slower growth rate and poorer feed conversion efficiency and high mortality rate than birds fed with a tannin-free diet.

Extraction of deoxyribose nucleic acid (DNA) from plants is preliminary step in molecular biology. Fast and cost effective genomic DNA isolation from Glycine species for downstream application is a major bottleneck. Here we report a high throughput and trouble free method for genomic DNA extraction from leaf and seeds of Glycine species with high quality and quantity. Protocol reports the optimization by employing different concentrations of CTAB and PVP in extraction buffer. Efficiency of optimized protocol was compared with frequently used DNA extraction methods. Wide adoptability and utility of this protocol was confirmed by DNA extraction from leaves as well as seeds of G. max, G. soja, G. tomentella and G. latifolia. Extracted DNA was successfully subjected to PCR amplification of five microsatellite markers and four putative glycosyltransferase genes. DNA extraction protocol is reproducible, trouble free, rapid and can be adopted for plant molecular biology applications.

This study analyses the consciousness levels, attitudes and behaviours of farmers against stubble burning and the damages of stubble burning which is a part of land misuse. 86 farmers from 9 villages in Zile county of Tokat province were surveyed for the study. These data was used to state farmers' socio-demographical characteristics and their behaviours against stubble burning was analysed. According to the study results, 99% of the farmers says that stubble burning is a wrong application. They states that stubble burning causes natural damages and the most importantly it is harmful by 76% to the living creatures in the nature. 57% of them prefers the method of mixing stubble to the soil.

This study was conducted to determine the better hygienic behaviour determination method that is used in controlling against bee diseases and pests. Total forty honeybee colonies (Apis mellifera anatoliaca) were used and they were randomly divided into two groups (each group consists of twenty colonies) in first year. Liquid nitrogen method was used in the first group' colonies and pin-killing (needling process) method was used in the second group’ colonies to determine the effectiveness of methods for hygienic behaviour. Average clearance rate was found as 66.25% and 78.10% in the first (Liquid nitrogen application) and the second (pin-killing) groups respectively. In the second year, forty colonies were divided into five equal groups and each group consisted eight colonies. The first group consisted of 9 frames bees in standard langstroot hive (10 frame capacity), the second group consisted of 5 frames bees in standard langstroot hive (10 frame capacity), the third group consisted of 5 frames bees in ruşet hive (5 frame capacity), the forth group’s consisted of 3 frames bees in ruşet hive (5 frame capacity) and the fifty group consisted of queen mating hive. The pin-killing (needling process) method, tested in first year, was used for all groups to determine the effectiveness of colony population and the size of hive. Average clearance rates of the first, second, third, forth and fifth groups were 70.54%, 58.38%, 70.63%, 54.96% and 58.21% respectively. The colonies that belonged to the pin-killing (needling) group showed a higher cleaning behaviour rate than the other colonies. The density of bees in hive had an important effect on the clearance rate of colonies. The colonies of group 1 and group 3, which had the more density of bees in per unit area, had the higher clearance rate than the other groups.

Binary logit regression model of econometrics was used to identify the factors affecting adoption of recommended agricultural technology by the commercial farmers of Nepal. A survey was carried out in 2012 in 120 households from Kavre district, Nepal. The objective was to appraise factors affecting adoption of recommended technology of cauliflower, finding out the level of adoption, identifying the constraints of cauliflower cultivation, assessing the perceived level of satisfaction and studying the relationship of certain selected variables on the adoption. Education, Occupation, contact with Personal Localite sources of information, Group membership and Experience were the most influencing factors for adoption of recommended technology. Though, other factors were not strong enough to contribute significantly but indirectly influence the adoption decision of farmers as combined effects. Occupation was found as negatively associated. Landholding size was strongly and positively influencing among the non significant factors. The level of satisfaction and the constraints of cauliflower production were also identified. Transfer of technology will be effective if and only if the client adopts the technology. For an effective transfer of technology, it is better to go for activities for the welfare of the socioeconomic factors of the farmers.

Before serving the meals, the managements that operate in the production and distribution of table d’hôte meals, the data related to whether it's appropriate for the food hygiene regulation and the other legal requirements are so insufficient. So, an interview study was performed for the present conditions of the manufactures producing and serving meals and the good hygiene practices profile of their working personal in the plants with regard to Law No.5966 on Veterinary Services, Plant health, Food and Feed in the Samsun city. The survey was applied in catering enterprises and face to face interviews with a total of 258 people (employees and managers responsible for the kitchen area) and at the same time observing the operating conditions of 32 companies in 9 different counties. The questions of survey included gender, age, education level, professional experience of workers, real production capacities, the power of engine, number of employees in production, and findings related to good hygiene practice. It was observed that the design and layout of the production area in terms of good hygiene practices were not sufficient in some manufactures and only four manufactures had positive air pressure ventilation. It was stated that tools and equipment were required to take more precautions against the risk of corrosion. It was established that cleaning and disinfection process, personal hygiene rules and dissolving operations for frozen food were inadequate. It was determined that record forms were not sufficient to provide the traceability. Hazard and risk factors will be controlled by establishment of practical food safety management system in catering sector.

The study aimed to identify physiological response of potato to drought. For this aim, a drought experiment was carried out by using two different potato varieties, cv. Desiree and Russet Burbank, under environmentally controlled greenhouse conditions. Drought treatment was initiated at 45 days after emergence (early tuber bulking period) by withholding irrigation for 10 days. Physiological traits such as stomatal conductance, transpiration rate, photosynthetic rate, chlorophyll index, leaf temperature, proline content, malondialdehyde (MDA) accumulation and hydrogen peroxide (H2O2) accumulation, in addition, some yield components average tuber weight, number of tubers and plant tuber yield were evaluated in the study. While the first significant decline in stomatal conductance, transpiration rate, and photosynthetic rate of both varieties was occurred at the 5th day of withholding irrigation, the highest decline was observed at 9th and 10th days of withholding irrigation. Proline content in both varieties increased two times at 10th day of withholding irrigation, however, H2O2 accumulation was not changed significantly by drought treatment. Even though MDA accumulation was increased in both varieties under drought stress conditions, the increase was significant in Desiree whereas, it was not significant in Russet Burbank. In addition, while drought treatment did not change the plant tuber yield in both varieties, it caused to a significant decline in average tuber yield of Russet Burbank, being an important trait for marketable tuber yield.

The article focuses on the impact of different dairy management types on quality characteristics of organic and conventional milk. The study was conducted during 9 months (spring-autumn), raw milk samples were collected from organic and conventional dairy farms from two Ukrainian regions. The milk samples were analyzed for dry matter, total protein and non-protein nitrogen, ammonia content and values of stable isotopes 13С/12С were measured in fat and milk protein fractions. The values of dry matter, total protein, true protein and non-protein nitrogen content do not represent a statistically significant effect of farming type. The values of ammonia content, ammonia/non-protein nitrogen and ammonia/total protein ratios were statistically significant and reflected the impact of farming type which is, probably, linked with different protein content in cows’ diet. Conversion of some parameters on the dry matter allows more precise differentiation among types of milk, including statistically significant differences (e.g. total protein and ammonia content). The values of δ13C in fat fraction (-26.00‰) and protein fraction (-22.89‰) of organic milk were statistically significant differed in comparison with conventional milk (-23.14 ‰ and -19.15‰ respectively), due to the high amount of maize in the diet on conventional farm. The values of Δδ13C also were statistically significant: 3.99‰ in conventional milk and 3.11‰ in organic. It was established that conventional milk has a greater range of the different parameter seasonal variations than organic milk.
