Microscopic study on the spleen of five healthy water buffaloes revealed that the capsule was loosely covered by a serous membrane and was composed of two distinct layers viz. an outer collagen and elastic fibrous and inner smooth muscle layer. The trabeculae compositions were related to their place of origin. The trabeculae of capsular origin were composed of an outer layer of smooth muscle fibers which originated from the inner layer of capsule and collagen and elastic fibers at the core originated from outer layer of capsule. The trabeculae originated from the hilus were rich in collagen and elastic fibers with few smooth muscle. Two cells thick smooth muscle fibers were noticed in the fine trabeculae of red pulp of the spleen in the water buffalo.
