Induction of barley haploid by chromosome elimination

A cultivar and two F2 intraspecific hybrids of Hordeum vulgare L., as maternal parents, were crossed with H. bulbosum to induce haploids in their progeny. On average, 42.5% of the pollinated florets formed embryos. The immature embryos were cultured in vitro and 59.24% of them grew into seedlings. The root-tip cells of 30 of those seedlings were observed and their chromosome numbers were all found to be 7, indicating that they were all haploids. The N-banding pattern of their chromosomes was similar to that of their maternal parents. Knop solution added with 0.04% colchicine was used to double the chromosome number of the haploids, and a doubling rate of 73.14% was obtained. The technical key points, advantages and limitations of applying the chromosome elimination technique to barley breeding are discussed.
