Determination of amino acid content in feedstuffs by the method of HPLC

Using Lichro CART Superspher CH-8, 4um, 250x4mm i.d.as stable phase and acetonitrile-Na-Acetat as mobile phase, the amino acids in the standard solution and in the hydrolyzed feedstuff solutions were derivatized with 9-fluorenylmethyl chloroformate (FMOC) at room temperature, separated with the reversed-phase system by adjusting the elution gradient and polarity of the mobile phase, and finally determined by fluoresence detector at emmission 310 mm and excitation 260 mm. The results showed that FMOC had high sensitivity and stability. A linearity was obtained between the concentration of amino acid and intensity of fluoresence within the range of concentrations in our experiment. The CA% of amino acid determined from the standard solutions was less than 4%, with the exception of lysine (CA=9.6%, n=12). The CA of the different hydrolyzed feedstuff solutions was less than 10%.
