Contribution to the study of production and characterization of polygalacturonase from Kluyveromyces marxianus and its mutant KF28.

A hyperproducing mutant of Kluyveromyces marxianus (KF28) was selected, the optimal culture conditions of which were different from the wild-type (KF). The production of polygalacturonase (PG) was obtained under three types of oxygenation. Yield was significantly affected by pH under aerobic conditions. The patterns of isoenzymes were identical between KF and KF28. Both have four isoenzymes shown by electrophoresis and chromatography. Two of which with different molecular weights, neutral sugars contents and Michaelis-Menten constant (Km) were purified and characterized. The mechanism of hydrolysis was investigated by HPLC. The isoenzymes mixture and one purified isoenzyme have the same mode of action, including activity 'ndo' and 'multichain' attack. The most important differences relating to the degradation of oligogalacturonoids were observed between endo-PG of K. marxianus and Aspergillus niger. The properties and degradation mechanism of poly-and oligogalacturonoids were affected differently by immobilization on eupergit or chitin.
