RP-HPLC of storage proteins of developing triticale grain

Ethanol-soluble proteins from whole meal of 20-, 40- and 60-day old grain of two triticale varieties grown in 1988/89 in lysimeters under controlled optimal soil and humidity conditions and at two levels of N application were characterized and compared. Resulting fractions were analyzed by reversed-phase high-performance liquid chromatography (RP-HPLC). There were no qualitative differences between fractions of each variety grown at different N application levels, but minor quantitative differences did exist. Ethanol-soluble triticale proteins 20 days after anthesis exhibited a different HPLC pattern from 40- or 60-day old kernels. However, at the wax maturity stage (40 days after anthesis), alcohol-soluble protein patterns of fractions were the same as in fully ripened 60-day old kernels. HPLC results are also compared with amino acid compositions of triticale grains at different maturity stages; protein accumulation at different levels of N application is also discussed
