Salinity adaptation of tobacco salt-tolerant callus variant

By increasing the salt concentration gradually, a cell line tolerant to 2% NaCl was established. The 2% NaCl-tolerant variant callus was subcultured through 52 passages in a modified MS medium containing 2% NaCl, then transferred to a medium containing 0.5%, 1.0%, 1.5% or 2.0% NaCl respectively with original type, its fresh wt., dry wt., relative growth rate and proline content were all superior to original type. The callus line possessed NaCl-tolerance thoroughly and the NaCl-tolerance was positively correlative with proline content. But when the 2% NaCl-tolerant variant callus was subcultured through 29 passages in a medium containing 2% NaCl, then transferred to NaCl-free for 11 and 20 passages, it did not maintain the tolerance. As table 2, 3 showed, they could only tolerate NaCl levels of 1.5% or 1.0%. Self-pollinated progenies of regenerated plant from the salt-tolerant variant were obtained. The germinating seeds, seedlings and adult plants of the progenies were all not salt tolerant. The results showed that the increased tolerance to salt of tobacco callus variant obtained as reported in this paper was only a physiological adaptation
