Kinetic parameters of nucleases in conditions of water deficiency

The aim of this investigation was to study the effect of water deficiency on kinetic parameters of the nucleases in the roots of young maize plants. Water deficiency was induced by water solutions of mannitol (55 g/l) and polyethylene glycol (200 g/l). The enzyme preparations from check seedlings and those grown in conditions of water deficiency showed the same substrate affinity, i.e., their Km values were very close. For ribonuclease 1, the Km values ranged from 1.67 to 1.82 g/l, for nuclease 1 from 1.45 to 1.52 g/l. The values obtained for V max were very close too. They were from 0.176 to 0.220 micromol/min for ribonuclease and from 0.071 to 0.147 micromol/min for nuclease 1. The results obtained suggest that no structural change occurred in the enzymes exposed to water deficiency.
