Genotypic differences of multiple shoot-forming capacity in soybean (Glycine max (L.) merr.) [in India]

Cultured cotyledonary node and shoot tip explants of 20 genotypes of soybean (Glycine max (L.) Merr.) grown in India were tested for their ability to form multiple shoots. Explants were cultured on three shoot induction media prepared with the B5 basal medium containing 3% sucrose and supplemented with three combinations of IBA (0.005 mg/l) and BAP (1.0, 5.0 and 10.0 mg/l) as growth regulators. The intensity of shoot multiplication increased during subculturing on B5 medium supplemented with IBA (0.005 mg/l) + BAP (1.0 mg/l). The average number of shoot buds/shoots ranged from 2 to 30 or more in different genotypes in the primary cultures which increased up to 100 or more from one explant of some genotypes after the 2nd passage. The unexpanded shoot buds were proliferated and elongated on B5 medium with lower levels of BAP (0.2 mg/l). The elongated shoots were rooted on half-strength hormone-free MS medium containing 6% sucrose. The degree of shoot bud formation was affected by genotype and explant types and the exogenous hormonal regime in the medium.
